Effects of experience and opponents on pacing behavior and 2-km cycling performance of novice youths
Purpose: To study the pacing behavior and performance of novice youth exercisers in a controlled laboratory setting. Method: Ten healthy participants (seven male, three female, 15.8 ± 1.0 years) completed four, 2-km trials on a Velotron cycling ergometer. Visit 1 was a familiarization trial. Visits 2 to 4 involved the following conditions, in randomized order: no opponent (NO), a virtual opponent (starting slow and finishing fast) (OP-SLOWFAST), and a virtual opponent (starting fast and finishing slow) (OP-FASTSLOW). Repeated measurement ANOVAs (p < .05) were used to examine differences in both pacing behavior and also performance related to power output, finishing- and split times, and RPE between the four successive visits and the three conditions. Expected performance outcome was measured using a questionnaire.
Results: Power output increased (F3,27 = 5.651, p = .004, η2p = .386) and finishing time decreased (F3,27 = 9.972, p .05).
Conclusion: Performance was improved by an increase in experience after one visit, parallel with the ability to anticipate future workload
The value of technical characteristics for future performance in youth tennis players:A prospective study
The aim of this study is to examine whether technical characteristics predict current and future tennis performance of youth tennis players. Twenty-nine male youth tennis players (age 13.40 ± .51) were assessed on anthropometrical characteristics (height, weight, maturity status) and technical characteristics (ball speed, accuracy and percentage errors) using an on-court tennis test when they were under-14 (U14). Game situations were simulated, which were either fixed or variable. The variable game situations required players to consider the direction of the ball, as opposed to the fixed game situations where players needed to play every ball to the same side. Players' tennis ratings were obtained U14 ('current performance') and under-18 (U18) ('future performance'). According to their rating U18 players were classified as future elite (n = 9) or future competitive (n = 20). A multiple linear regression analysis showed that ball speed and accuracy were significant predictors of current and future performance (p < .001), with R2 of .595 and .463, respectively. When controlling for age, a one-way MANCOVA revealed that future elite players were more accurate than future competitive players (p = .048, 95% CI [.000 to .489]), especially in variable compared to fixed game situations (p < .05). In conclusion, the current study is the first to show that technical characteristics are crucial for current as well as future performance in youth male tennis players. Findings of this prospective study provide essential information to coaches about characteristics that require most attention in performance development in youth players
Pacing Behavior and Tactical Positioning in 500- and 1000-m Short-Track Speed Skating
Purpose: To explore pacing behavior and tactical positioning during the shorter 500- and 1000-m short-track competitions.
Methods: Lap times and intermediate rankings of elite 500- and 1000-m short-track-skating competitors were collected over the 2012–13 season. First, lap times were analyzed using a MANOVA, and for each lap, differences between sex, race type, final ranking, and stage of competition were determined. Second, Kendall tau-b correlations were used to assess relationships between intermediate and final rankings. In addition, intermediate rankings of the winner of each race were examined.
Results: Top-placed athletes appeared faster than bottom-placed athletes in every lap in the 500-m, while in the 1000-m no differences were found until the final 4 laps (P < .05). Correlations between intermediate and final rankings were already high at the beginning stages of the 50-m (lap 1: r = .59) but not for the 1000-m (lap 1: r = .21).
Conclusions: Although 500- and 1000-m short-track races are both relatively short, fundamental differences in pacing behavior and tactical positioning were found. A fast-start strategy seems to be optimal for 500-m races, while the crucial segment in 1000-m races seems to be from the 6th lap to the finish line (ie, after ± 650 m). These findings provide evidence to suggest that athletes balance between choosing an energetically optimal profile and the tactical and positional benefits that play a role when riding against an opponent, as well as contributing to developing novel insights in exploring athletic behavior when racing against opponents
Self-assessed tactical skills in tennis players:Psychometric evaluation of the Tactical Skills Questionnaire in Tennis
To our knowledge, no feasible, valid and reliable instrument exists to examine tactical skills over the course of multiple training and game situations in tennis yet. Therefore, the aim of this study was to develop and evaluate the psychometric properties of the Tactical Skills Questionnaire in Tennis (TSQT). The TSQT is a new instrument with closed-ended questions designed to examine tactical skills in tennis players. Participants were 233 competitive tennis players (age: 17.06 ± 4.74 years) competing on national or regional levels. With a principal component analysis (PCA) we identified four theoretically meaningful subscales for the 31-item TSQT: “Anticipation and positioning,” “Game intelligence and adaptability,” “Decision-making,” and “Recognizing game situations” and confirmed them with a confirmatory factor analysis (CFA) (χ2 = 527.02, df = 426, p 0.05). Relative test-retest reliability was moderate with ICC values ranging from 0.65 to 0.71. National players outperformed regional players on the subscales “Game intelligence and adaptability,” “Decision-making,” and “Recognizing game situations” (p < 0.05), and there was a trend toward significance for “Anticipation and positioning” (p = 0.07). This study supported the psychometric properties of the TSQT. Evaluating tactical skills with the TSQT provides players, coaches and other professionals with insight in players' self-assessed tactical skills over the course of multiple training and game situations. It creates the opportunity for players to reflect on their skills and detect personal development areas with their coach. We advise to use this information as input for tailor-made training programs
Biliary Bicarbonate Secretion Constitutes a Protective Mechanism against Bile Acid-Induced Injury in Man
Background: Cholangiocytes expose a striking resistance against bile acids: while other cell types, such as hepatocytes, are susceptible to bile acid-induced toxicity and apoptosis already at micromolar concentrations, cholangiocytes are continuously exposed to millimolar concentrations as present in bile. We present a hypothesis suggesting that biliary secretion of HCO(3)(-) in man serves to protect cholangiocytes against bile acid-induced damage by fostering the deprotonation of apolar bile acids to more polar bile salts. Here, we tested if bile acid-induced toxicity is pH-dependent and if anion exchanger 2 (AE2) protects against bile acid-induced damage. Methods: A human cholangiocyte cell line was exposed to chenodeoxycholate (CDC), or its glycine conjugate, from 0.5 mM to 2.0 mM at pH 7.4, 7.1, 6.7 or 6.4, or after knockdown of AE2. Cell viability and apoptosis were determined by WST and caspase-3/-7 assays, respectively. Results: Glycochenodeoxycholate (GCDC) uptake in cholangiocytes is pH-dependent. Furthermore, CDC and GCDC (pK(a) 4-5) induce cholangiocyte toxicity in a pH-dependent manner: 0.5 mM CDC and 1 mM GCDC at pH 7.4 had no effect on cell viability, but at pH 6.4 decreased viability by >80% and increased caspase activity almost 10- and 30-fold, respectively. Acidification alone had no effect. AE2 knockdown led to 3- and 2-fold enhanced apoptosis induced by 0.75 mM CDC or 2 mM GCDC at pH 7.4. Discussion: These data support our hypothesis of a biliary HCO(3)(-) umbrella serving to protect human cholangiocytes against bile acid-induced injury. AE2 is a key contributor to this protective mechanism. The development and progression of cholangiopathies, such as primary biliary cirrhosis, may be a consequence of genetic and acquired functional defects of genes involved in maintaining the biliary HCO(3)(-) umbrella. The tactics of successful attacks in professional association football:large-scale spatiotemporal analysis of dynamic subgroups using position tracking data
Association football teams can be considered complex dynamical systems of individuals grouped in subgroups (defenders, midfielders and attackers), coordinating their behaviour to achieve a shared goal. As research often focusses on collective behaviour, or on static subgroups, the current study aims to analyse spatiotemporal behaviour of dynamic subgroups in relation to successful attacks. We collected position tracking data of 118 Dutch Eredivisie matches, containing 12424 attacks. Attacks were classified as successful (N = 1237) or non-successful (N = 11187) based on the potential of creating a scoring opportunity. Using unsupervised machine learning, we automatically identified dynamic formations based on position tracking data, and identified dynamic subgroups for every timeframe in a match. We then compared the subgroup centroids to assess the intra- and inter-team spatiotemporal synchronisation during successful and non-successful attacks, using circular statistics. Our results indicated subgroup-level variables provided more information, and were more sensitive to disruption, in comparison to team-level variables. When comparing successful and non-successful attacks, we found decreases (p < .01) in longitudinal inter- and intra-team synchrony of interactions involving the defenders of the attacking team during successful attacks. This study provides the first large-scale dynamic subgroup analysis and reveals additional insights to team-level analyses
Requirement of biphasic calcium release from the endoplasmic reticulum for Fas-mediated apoptosis
Fas receptor is a member of the tumor necrosis factor-α family of death receptors that mediate physiologic apoptotic signaling. To investigate the molecular mechanisms regulating calcium mobilization during Fas-mediated apoptosis, we have analyzed the sequential steps leading to altered calcium homeostasis and cell death in response to activation of the Fas receptor. We show that Fas-mediated apoptosis requires endoplasmic reticulum–mediated calcium release in a mechanism dependent on phospholipase C-γ1 (PLC-γ1) activation and Ca2+ release from inositol 1,4,5-trisphosphate receptor (IP3R) channels. The kinetics of Ca2+ release were biphasic, demonstrating a rapid elevation caused by PLC-γ1 activation and a delayed and sustained increase caused by cytochrome c binding to IP3R. Blocking either phase of Ca2+ mobilization was cytoprotective, highlighting PLC-γ1 and IP3R as possible therapeutic targets for disorders associated with Fas signaling
Fibrates for the treatment of cholestatic itch (FITCH): study protocol for a randomized controlled trial
BACKGROUND: Pruritus (itch) is a frequent, burdensome and difficult-to-treat symptom in patients with cholestasis. Fibrates are currently under investigation for the treatment of primary biliary cholangitis in patients with a suboptimal response to ursodeoxycholic acid. Moreover, there is empirical evidence for a possible antipruritic effect. We aim to prove this in a randomized controlled trial, including patients with cholestatic liver diseases other than primary biliary cholangitis that are accompanied by pruritus. METHODS: A multicenter investigator-initiated, double-blind, randomized placebo-controlled trial to evaluate the effect of bezafibrate on cholestatic pruritus in 84 adult patients with primary biliary cholangitis or primary/secondary sclerosing cholangitis. Primary outcome is the proportion of patients with a reduction of itch intensity of 50% or more (measured on a Visual Analog Scale) after 21 days of treatment with bezafibrate 400 mg qid or placebo. Secondary outcomes include the effect of bezafibrate on a five-dimensional itch score, liver disease-specific quality of life, serum liver tests and autotaxin activity. Safety will be evaluated through serum parameters for kidney function and rhabdomyolysis as well as precise recording of (serious) adverse events. We provide a schematic overview of the study protocol and describe the methods used to recruit and randomize patients, collect and handle data and perform statistical analyses. DISCUSSION: Given its favorable safety profile and anticholestatic properties, bezafibrate may become the new first-line treatment option for treating cholestatic pruritus
