    Laboratory And Astronomical Detection Of The Negative Molecular Ion C3N-

    The negative molecular ion C3N- has been detected at millimeter wavelengths in a low-pressure laboratory discharge, and then with frequencies derived from the laboratory data in the molecular envelope of IRC+10216. Spectroscopic constants derived from laboratory measurements of 12 transitions between 97 and 378 GHz allow the rotational spectrum to be calculated well into the submillimeter-wave band to 0.03 km s(-1) or better in equivalent radial velocity. Four transitions of C3N- were detected in IRC+10216 with the IRAM 30 m telescope at precisely the frequencies calculated from the laboratory measurements. The column density of C3N- is 0.5% that of C3N, or approximately 20 times greater than that of C4H- relative to C4H. The C3N- abundance in IRC+10216 is compared with a chemical model calculation by Petrie & Herbst. An upper limit in TMC-1 for C3N- relative to C3N (< 0.8%) and a limit for C4H- relative to C4H (< 0.004%) that is 5 times lower than that found in IRC+10216, were obtained from observations with the NRAO 100 m Green Bank Telescope (GBT).     Dust-forming molecules in VY Canis Majoris (and Betelgeuse)

    The formation of inorganic dust in circumstellar environments of evolved stars is poorly understood. Spectra of molecules thought to be most important for the nucleation, i.e. AlO, TiO, and TiO2, have been recently detected in the red supergiant VY CMa. These molecules are effectively formed in VY CMa and the observations suggest that non-equilibrium chemistry must be involved in their formation and nucleation into dust.     Pure rotational spectra of TiO and TiO_2 in VY Canis Majoris

    We report the first detection of pure rotational transitions of TiO and TiO_2 at (sub-)millimeter wavelengths towards the red supergiant VY CMa. A rotational temperature, T_rot, of about 250 K was derived for TiO_2. Although T_rot was not well constrained for TiO, it is likely somewhat higher than that of TiO_2. The detection of the Ti oxides confirms that they are formed in the circumstellar envelopes of cool oxygen-rich stars and may be the "seeds" of inorganic-dust formation, but alternative explanations for our observation of TiO and TiO_2 in the cooler regions of the envelope cannot be ruled out at this time. The observations suggest that a significant fraction of the oxides is not converted to dust, but instead remains in the gas phase throughout the outflow.Comment: to appear in Astronomy and Astrophysic

    Identification of two novel activities of the Wnt signaling regulator Dickkopf 3 and characterization of its expression in the mouse retina

    <p>Abstract</p> <p>Background</p> <p>The Wnt signaling pathway is a cellular communication pathway that plays critical roles in development and disease. A major class of Wnt signaling regulators is the Dickkopf (Dkk) family of secreted glycoproteins. Although the biological properties of Dickkopf 1 (Dkk1) and Dickkopf 2 (Dkk2) are well characterized, little is known about the function of the related Dickkopf 3 (Dkk3) protein in vivo or in cell lines. We recently demonstrated that Dkk3 transcripts are upregulated during photoreceptor death in a mouse model of retinal degeneration. In this study, we characterized the activity of Dkk3 in Wnt signaling and cell death.</p> <p>Results</p> <p>Dkk3 was localized to Müller glia and retinal ganglion cells in developing and adult mouse retina. Western blotting confirmed that Dkk3 is secreted from Müller glia cells in culture. We demonstrated that Dkk3 potentiated Wnt signaling in Müller glia and HEK293 cells but not in COS7 cells, indicating that it is a cell-type specific regulator of Wnt signaling. This unique Dkk3 activity was blocked by co-expression of Dkk1. Additionally, Dkk3 displayed pro-survival properties by decreasing caspase activation and increasing viability in HEK293 cells exposed to staurosporine and H<sub>2</sub>O<sub>2</sub>. In contrast, Dkk3 did not protect COS7 cells from apoptosis.</p> <p>Conclusion</p> <p>These data demonstrate that Dkk3 is a positive regulator of Wnt signaling, in contrast to its family member Dkk1. Furthermore, Dkk3 protects against apoptosis by reducing caspase activity, suggesting that Dkk3 may play a cytoprotective role in the retina.</p

    Alterations of epithelial stem cell marker patterns in human diabetic corneas and effects of c-met gene therapy

    PURPOSE:We have previously identified specific epithelial proteins with altered expression in human diabetic central corneas. Decreased hepatocyte growth factor receptor (c-met) and increased proteinases were functionally implicated in the changes of these proteins in diabetes. The present study examined whether limbal stem cell marker patterns were altered in diabetic corneas and whether c-met gene overexpression could normalize these patterns.METHODS:Cryostat sections of 28 ex vivo and 26 organ-cultured autopsy human normal and diabetic corneas were examined by immunohistochemistry using antibodies to putative limbal stem cell markers including ATP-binding cassette sub-family G member 2 (ABCG2), N-cadherin, \u394Np63\u3b1, tenascin-C, laminin \u3b33 chain, keratins (K) K15, K17, K19, \u3b2(1) integrin, vimentin, frizzled 7, and fibronectin. Organ-cultured diabetic corneas were studied upon transduction with adenovirus harboring c-met gene.RESULTS:Immunostaining for ABCG2, N-cadherin, \u394Np63\u3b1, K15, K17, K19, and \u3b2(1) integrin, was significantly decreased in the stem cell-harboring diabetic limbal basal epithelium either by intensity or the number of positive cells. Basement membrane components, laminin \u3b33 chain, and fibronectin (but not tenascin-C) also showed a significant reduction in the ex vivo diabetic limbus. c-Met gene transduction, which normalizes diabetic marker expression and epithelial wound healing, was accompanied by increased limbal epithelial staining for K17, K19, \u394Np63\u3b1, and a diabetic marker \u3b1(3)\u3b2(1) integrin, compared to vector-transduced corneas.CONCLUSIONS:The data suggest that limbal stem cell compartment is altered in long-term diabetes. Gene therapy, such as with c-met overexpression, could be able to restore normal function to diabetic corneal epithelial stem cells

    The S-DALINAC is a 130 MeV superconducting recirculating electron accelerator serving several nuclear and radiation physics experiments as well as driving an infrared free-electron laser. A system of normal conducting rf resonators for noninvasive beam position and current measurement was established. For the measurement of gamma-radiation inside the accelerator cave a system of Compton diodes has been developed and tested. Detailed investigations of the transverse phasespace were carried out with a tomographical reconstruction method of optical transition radiation spots. The method can be applied also to non-Gaussian phasespace distributions. The results are in good accordance with simulations. To improve the quality factor of the superconducting 3 GHz cavities, an external 2K testcryostat was commissioned. The influence of electro-chemical polishing and magnetic shielding is currently under investigation.     A major asymmetric ice trap in a planet-forming disk IV. Nitric oxide gas and a lack of CN tracing sublimating ices and a C/O ratio <1< 1

    [Abridged] Most well-resolved disks observed with ALMA show signs of dust traps. These dust traps set the chemical composition of the planet forming material in these disks, as the dust grains with their icy mantles are trapped at specific radii and could deplete the gas and dust of volatiles at smaller radii. In this work we analyse the first detection of nitric oxide (NO) in a protoplanetary disk. We aim to constrain the nitrogen chemistry and the gas-phase C/O ratio in the highly asymmetric dust trap in the Oph-IRS 48 disk. We use ALMA observations of NO, CN, C2_2H, and related molecules and model the effect of the dust trap on the physical and chemical structure using the thermochemical code DALI. Furthermore, we explore how ice sublimation contributes to the observed emission lines. NO is only observed at the location of the dust trap but CN and C2_2H are not detected in the Oph-IRS 48 disk. This results in an CN/NO column density ratio of <0.05< 0.05 and thus a low C/O ratio at the location of the dust trap. The main gas-phase formation pathways to NO through OH and NH in the fiducial model predict NO emission that is an order of magnitude lower than is observed. The gaseous NO column density can be increased by factors ranging from 2.8 to 10 when the H2_2O and NH3_3 gas abundances are significantly boosted by ice sublimation. However, these models are inconsistent with the upper limits on the H2_2O and OH column densities derived from observations. We propose that the NO emission in the Oph-IRS 48 disk is closely related to the nitrogen containing ices sublimating in the dust trap. The non-detection of CN constrains the C/O ratio both inside and outside the dust trap to be <1< 1 if all nitrogen initially starts as N2_2 and 0.6\leq 0.6, consistent with the Solar value, if (part of) the nitrogen initially starts as N or NH3_3.Comment: Accepted for publication in Astronomy and Astrophysic

    Grazing incidence x ray fluorescence analysis for non destructive determination of In and Ga depth profiles in Cu In,Ga Se2 absorber films

    Development of highly efficient thin film solar cells involves band gap engineering by tuning their elemental composition with depth. Here we show that grazing incidence X ray fluorescence GIXRF analysis using monochromatic synchrotron radiation and well characterized instrumentation is suitable for a non destructive and reference free analysis of compositional depth profiles in thin films. Variation of the incidence angle provides quantitative access to the in depth distribution of the elements, which are retrieved from measured fluorescence intensities by modeling parameterized gradients and fitting calculated to measured fluorescence intensities. Our results show that double Ga gradients in Cu In1 x,Gax Se2 can be resolved by GIXR
