    The Glycolytic Pathway is the Predominate Path for Glucose Utilization in Human Pancreatic Beta Cells (1.1B4)

    The oxidative metabolism of energy substrates has a paramount role in the stimulus secretion pathway of insulin. However, the role of glycolytic pathway in pancreatic beta cells is not very well understood. To address this, we have investigated and compared the functional effects of two mitochondrial substrates (glucose and α-ketoisocaproate) between the human (1.1B4) and murine (MIN6) pancreatic beta cell lines. MTS assay was conducted as an indicator of the metabolic activity of both cell lines. Polarographic detection of (ΔO2) and lactate were used to measure the oxygen consumption rate and anaerobic glycolysis respectively. The mitochondrial redox state was monitored via RH123 distribution and NAD(P)H autofluorescence. The metabolic assays showed glucose stimulated MTS reduction in MIN6 cells in a time and concentration dependent manner and nor in 1.1B4. Both sub strates failed to affect OCR, NADPH and increased lactate production in 1.1B4 cells. However, they stimulated OCR, increased NADPH, increased lactate output but was less extent and hyperpolarized the mitochondria in MIN6 cells. The above results showed that 1.1B4 cells are mainly depending on the glycolytic pathway different from MIN6 cells which rely on mitochondrial respiration. In conclusion, 1.1B4 cell line represents a new model to study the bioenergetics profile because it depends on the anaerobic glycolysis rather than aerobic respiration of the other models such as MIN6 and islets

    Influence of uniaxial tensile stress on the mechanical and piezoelectric properties of short-period ferroelectric superlattice

    Tetragonal ferroelectric/ferroelectric BaTiO3/PbTiO3 superlattice under uniaxial tensile stress along the c axis is investigated from first principles. We show that the calculated ideal tensile strength is 6.85 GPa and that the superlattice under the loading of uniaxial tensile stress becomes soft along the nonpolar axes. We also find that the appropriately applied uniaxial tensile stress can significantly enhance the piezoelectricity for the superlattice, with piezoelectric coefficient d33 increasing from the ground state value by a factor of about 8, reaching 678.42 pC/N. The underlying mechanism for the enhancement of piezoelectricity is discussed

    Subfemtosecond steering of hydrocarbon deprotonation through superposition of vibrational modes

    Subfemtosecond control of the breaking and making of chemical bonds in polyatomic molecules is poised to open new pathways for the laser-driven synthesis of chemical products. The break-up of the C-H bond in hydrocarbons is an ubiquitous process during laser-induced dissociation. While the yield of the deprotonation of hydrocarbons has been successfully manipulated in recent studies, full control of the reaction would also require a directional control (that is, which C-H bond is broken). Here, we demonstrate steering of deprotonation from symmetric acetylene molecules on subfemtosecond timescales before the break-up of the molecular dication. On the basis of quantum mechanical calculations, the experimental results are interpreted in terms of a novel subfemtosecond control mechanism involving non-resonant excitation and superposition of vibrational degrees of freedom. This mechanism permits control over the directionality of chemical reactions via vibrational excitation on timescales defined by the subcycle evolution of the laser waveform

    Healthcare providers as patients: COVID-19 experience

    There is compelling evidence for the psychological effects of the COVID-19 pandemic and earlier epidemics. However, fewer studies have examined the subjective meaning experience of healthcare providers who have survived COVID-19 as patients. This qualitative study aimed to understand further and describe the life experiences of healthcare providers who have survived COVID-19 as patients in Saudi Arabia. Data was collected using unstructured in-depth individual interviews among n = 10 healthcare providers from public hospitals in Saudi Arabia. Data were analyzed based on a phenomenological approach, which resulted in five themes: (i) physical and psychological signs and symptoms; (ii) self-healing, hiding pain, and family; (iii) fear of complications; (iv) disease stigma & long-term psychological outcomes; (v) emotional support, mental well-being & resignation. The overall synthesis showed that healthcare providers, as patients, experience the same difficulties and stressors as the general public. In some cases, these factors are even worse, as family members, colleagues, and employers develop a new type of stigma. Given the impact of social media and the flow of information of any type, more research is needed to examine the sources used to obtain information by the general public, whether these sources are reliable, and how the public can be taught to use only scientific data and not social data. Understanding the experience of healthcare providers as patients during the pandemic has allowed to look at the feelings and needs of people during illness from a new perspective. As expressed by participants, being a healthcare provider does not reduce the fear of the disease and does not mitigate its consequences in the form of stigmatization and isolation

    First Report of 13 Species of Culicoides (Diptera: Ceratopogonidae) in Mainland Portugal and Azores by Morphological and Molecular Characterization

    The genus Culicoides (Diptera: Ceratopogonidae) contains important vectors of animal and human diseases, including bluetongue, African horse sickness and filariosis. A major outbreak of bluetongue occurred in mainland Portugal in 2004, forty eight years after the last recorded case. A national Entomological Surveillance Plan was initiated in mainland Portugal, Azores and the Madeira archipelagos in 2005 in order to better understand the disease and facilitate policy decisions. During the survey, the most prevalent Culicoides species in mainland Portugal was C. imicola (75.3%) and species belonging to the Obsoletus group (6.5%). The latter were the most prevalent in Azores archipelago, accounting for 96.7% of the total species identified. The Obsoletus group was further characterized by multiplex Polymerase Chain Reaction to species level showing that only two species of this group were present: C. obsoletus sensu strictu (69.6%) and C. scoticus (30.4%). Nine species of Culicoides were detected for the first time in mainland Portugal: C. alazanicus, C. bahrainensis, C. deltus, C. lupicaris, C. picturatus, C. santonicus, C. semimaculatus, C. simulator and C. subfagineus. In the Azores, C. newsteadi and C. circumscriptus were identified for the first time from some islands, and bluetongue vectors belonging to the Obsoletus group (C. obsoletus and C. scoticus) were found to be widespread

    Fluoxetine during Development Reverses the Effects of Prenatal Stress on Depressive-Like Behavior and Hippocampal Neurogenesis in Adolescence

    Depression during pregnancy and the postpartum period is a growing health problem, which affects up to 20% of women. Currently, selective serotonin reuptake inhibitor (SSRIs) medications are commonly used for treatment of maternal depression. Unfortunately, there is very little research on the long-term effect of maternal depression and perinatal SSRI exposure on offspring development. Therefore, the aim of this study was to determine the role of exposure to fluoxetine during development on affective-like behaviors and hippocampal neurogenesis in adolescent offspring in a rodent model of maternal depression. To do this, gestationally stressed and non-stressed Sprague-Dawley rat dams were treated with either fluoxetine (5 mg/kg/day) or vehicle beginning on postnatal day 1 (P1). Adolescent male and female offspring were divided into 4 groups: 1) prenatal stress+fluoxetine exposure, 2) prenatal stress+vehicle, 3) fluoxetine exposure alone, and 4) vehicle alone. Adolescent offspring were assessed for anxiety-like behavior using the Open Field Test and depressive-like behavior using the Forced Swim Test. Brains were analyzed for endogenous markers of hippocampal neurogenesis via immunohistochemistry. Results demonstrate that maternal fluoxetine exposure reverses the reduction in immobility evident in prenatally stressed adolescent offspring. In addition, maternal fluoxetine exposure reverses the decrease in hippocampal cell proliferation and neurogenesis in maternally stressed adolescent offspring. This research provides important evidence on the long-term effect of fluoxetine exposure during development in a model of maternal adversity

    Resource Sharing in Energy Communities: A Cooperative Game Approach

    We analyze the overall benefits of an energy community cooperative game under which distributed energy resources (DER) are shared behind a regulated distribution utility meter under a general net energy metering (NEM) tariff. Two community DER scheduling algorithms are examined. The first is a community with centrally controlled DER, whereas the second is decentralized letting its members schedule their own DER locally. For both communities, we prove that the cooperative game's value function is superadditive, hence the grand coalition achieves the highest welfare. We also prove the balancedness of the cooperative game under the two DER scheduling algorithms, which means that there is a welfare re-distribution scheme that de-incentivizes players from leaving the grand coalition to form smaller ones. Lastly, we present five ex-post and an ex-ante welfare re-distribution mechanisms and evaluate them in simulation, in addition to investigating the performance of various community sizes under the two DER scheduling algorithms.Comment: 8 page
