    Role of histone modifications in marking and activating genes through mitosis

    The global inhibition of transcription at the mitotic phase of the cell cycle occurs together with the general displacement of transcription factors from the mitotic chromatin. Nevertheless, the DNase- and potassium permanganate-hypersensitive sites are maintained on potentially active promoters during mitosis, helping to mark active genes at this stage of the cell cycle. Our study focuses on the role of histone acetylation and H3 (Lys-4) methylation in the maintenance of the competency of these active genes during mitosis. To this end we have analyzed histone modifications across the promoters and coding regions of constitutively active, inducible, and inactive genes in mitotic arrested cells. Our results show that basal histone modifications are maintained during mitosis at promoters and coding regions of the active and inducible RNA polymerase II-transcribed genes. In addition we have demonstrated that, together with H3 acetylation and H3 (Lys-4) methylation, H4 (Lys-12) acetylation at the coding regions contributes to the formation of a stable mark on active genes at this stage of the cell cycle. Finally, analysis of cyclin B1 gene activation during mitosis revealed that the former occurs with a strong increase of H3 (Lys-4) trimethylation but not H3 or H4 acetylation, suggesting that histone methyltransferases are active during this stage.     Friendly Residential Environments and Subjective Well-Being in Older People with and without Help Needs.

    Previous studies have shown that friendly environments are associated with well-being and higher quality of life in older people. This study aimed to investigate the relationship between friendly environments and subjective well-being by segmenting the population according to the need for help in performing activities of daily living (ADLs) in a representative sample of people over 55 years of age in the Basque Country (Spain) (n = 2760). To determine the predictive power of friendliness on subjective well-being, two separate linear regression models were obtained according to the need for help in ADLs. The results obtained show a greater explanatory power of the model in the case of people who required help. However, in the case of people who do not need help, subjective health had a greater weight in the predictions. This paper’s findings support the greater importance of the characteristics of the physical and social environment, as people’s functional status worsens, with friendliness being an explanatory factor for people’s well-being as they age and their dependency increases.post-print399 K

    Every day the practice of hospital emergency and urgent care, we can find extreme life-threatening situations, situations in which the training of health personnel and the skills and knowledge on use of rescue material, will determine the evolution of injuries. It is therefore our aim is to provide more information than is currently known about the technique of fixation or immobilization of fractures. The treatment of these, in most cases, will depend on the characteristics of the injury although facts that are common to all the circumstances is the initial care and management, namely, immobilization and stabilization of the lesions with the appropriate material and in order to reduce risk of alteration in other anatomical structures as a result of the fracture. This, we can achieve with the use of vacuum immobilizers or vacuum cast.En la práctica diaria de las emergencias extrahospitalarias y atención urgente, nos podemos encontrar con situaciones de riesgo vital extremo, situaciones en las cuales la formación del personal sanitario así como las destrezas y conocimientos sobre utilización del material de rescate, van a determinar la evolución de las lesiones. Es por tanto nuestro objetivo, aportar mayor información de la que se conoce actualmente, sobre la técnica de fijación o inmovilización de fracturas. El tratamiento de estas, en la mayoría de los casos, va a depender de las características de la lesión aunque un factor común a todas las circunstancias es la atención y manejo inicial, es decir, la inmovilización y estabilización de las lesiones con el material adecuado y con el objetivo de disminuir riesgos de alteración en otras estructuras anatómicas como consecuencia de la fractura. El Enfoque tendrá un carácter mixto (cualitativo - cuantitativo), con predominio cualitativo, por lo tanto, se aplicarán diferentes instrumentos para comprender cómo se generan las Disonancias Cognitivas desde los discursos deterministas de los medios masivos. Los resultados esperados, pretenden evidenciar cómo los medios masivos al establecer los conceptos para tratar un tema, exacerban las disonancias cognitivas, que los ciudadanos esgrimen sin una previa investigación. The expected data coming out of this investigation aim to find out how mass media, by establishing concepts to develop a subject, exacerbate the cognitive dissonances that the citizens follow with no previous investigation. For the conclusion, it is established that the cognitive dissonances in the speech of the mass media developed a socio cultural judgment that stigmatize as gender ideology the content of the free discrimination environment book

    Differential Estrogenic Effects of the Persistent Organochlorine Pesticides Dieldrin, Endosulfan, and Lindane in Primary Neuronal Cultures

    The organochlorine chemicals endosulfan, dieldrin, and ghexachlorocyclohexane (lindane) are persistent pesticides to which people are exposed mainly via diet. Their antagonism of the g-aminobutyric acid-A (GABAA) receptor makes them convulsants. They are also endocrine disruptors because of their interaction with the estrogen receptor (ER). Here, we study the effects of dieldrin, endosulfan, and lindane on ERs in primary cultures of cortical neurons (CN) and cerebellar granule cells (CGC). All the compounds tested inhibited the binding of [3H]-estradiol to the ER in both CN and CGC, with dieldrin in CGC showing the highest affinity.We also determined the effects of the pesticides on protein kinase B (Akt) and extracellular-regulated kinase 1 and 2 (ERK1/2) phosphorylation. Dieldrin and endosulfan increased Akt phosphorylation in CN, which was inhibited by the ERb antagonist 4-[2-phenyl-5,7- bis(trifluoromethyl)pyrazolo[1,5-a]pyrimidin-3-yl]phenol. Instead, Akt and ERK1/2 phosphorylation induced by dieldrin in CGC was mediated by multiple activation of ERa, ERb, and G protein– coupled receptor 30. Lindane did not activate these pathways, but it inhibited estradiol-mediated Akt and ERK1/2 activation. In CN, all the chemicals activated ERK1/2 through a mechanism involving GABAA and glutamate receptors. Long-term exposure to these pesticides reduced the levels of ERa, but not of ERb. Moreover, extracts of CN treated with endosulfan, dieldrin, or lindane induced cell proliferation in MCF-7 human breast cancer–derived cells, whereas only extracts of CGC treated with dieldrin induced MCF-7 cell proliferation.     En el presente trabajo se estudia el funcionamiento y desarrollo histórico del Hospital de San Cosme y San Damián, la primera y más antigua de las instituciones existentes en Sevilla cuya función principal era la de «curar a los pobres enfermos faltos de juicio». Concretamente se estudia el origen, la evolución y significación del hospital a lo largo de los siglos, así como la información disponible acerca de los ingresos, patologías y tratamientos que constan en la documentación. También se exploran las competencias y funciones de las diferentes figuras que formaban parte del servicio a los enfermos.This article studies the functioning and the historical development of the Hospital de San Cosme y San Damián, the first and oldest institution of Seville which main function was «healing the insane without sense». Specifically, we study the origin, evolution and significance of the hospital through the centuries, as well as the available information regarding admissions, pathologies and treatment contained in the documentation.     RING1B contributes to Ewing sarcoma development by repressing the Na(V)1.6 sodium channel and the NF-kappa B pathway, independently of the fusion oncoprotein

    Ewing sarcoma (ES) is an aggressive tumor defined by EWSR1 gene fusions that behave as an oncogene. Here we demonstrate that RING1B is highly expressed in primary ES tumors, and its expression is independent of the fusion oncogene. RING1B-depleted ES cells display an expression profile enriched in genes functionally involved in hematological development but RING1B depletion does not induce cellular differentiation. In ES cells, RING1B directly binds the SCN8A sodium channel promoter and its depletion results in enhanced Nav1.6 expression and function. The signaling pathway most significantly modulated by RING1B is NF-κB. RING1B depletion results in enhanced p105/p50 expression, which sensitizes ES cells to apoptosis by FGFR/SHP2/STAT3 blockade. Reduced NaV1.6 function protects ES cells from apoptotic cell death by maintaining low NF-κB levels.
