Expression Patterns of Irx Genes in the Developing Chick Inner Ear

The molecular patterning of the developing otic epithelium creates various positional identities, consequently leading to the stereotyped specification of each neurosensory and non-sensory element of the membranous labyrinth. The Iroquois (Iro/Irx) genes, clustered in two groups (A: Irx1, Irx2, and Irx4; and B: Irx3, Irx5, and Irx6), encode for transcriptional factors involved directly in numerous patterning processes of embryonic tissues in many phyla. This work presents a detailed study of the expression patterns of these six Irx genes during chick inner ear development, paying particular attention to the axial specification of the otic anlagen. The Irx genes seem to play different roles at different embryonic periods. At the otic vesicle stage (HH18), all the genes of each cluster are expressed identically. Both clusters A and B seem involved in the specification of the lateral and posterior portions of the otic anlagen. Cluster B seems to regulate a larger area than cluster A, including the presumptive territory of the endolymphatic apparatus. Both clusters seem also to be involved in neurogenic events. At stages HH24/25-HH27, combinations of IrxA and IrxB genes participate in the specification of most sensory patches and some non-sensory components of the otic epithelium. At stage HH34, the six Irx genes show divergent patterns of expression, leading to the final specification of the membranous labyrinth, as well as to cell differentiation
Analysis of the Promoters Involved in Enterocin AS-48 Expression
The enterocin AS-48 is the best characterized antibacterial circular protein in prokaryotes. It is a hydrophobic and cationic bacteriocin, which is ribosomally synthesized by enterococcal cells and post-translationally cyclized by a head-to-tail peptide bond. The production of and immunity towards AS-48 depend upon the coordinated expression of ten genes organized in two operons, as-48ABC (where genes encoding enzymes with processing, secretion, and immunity functions are adjacent to the structural as-48A gene) and as-48C1DD1EFGH. The current study describes the identification of the promoters involved in AS-48 expression. Seven putative promoters have been here amplified, and separately inserted into the promoter-probe vector pTLR1, to create transcriptional fusions with the mCherry gene used as a reporter. The activity of these promoter regions was assessed measuring the expression of the fluorescent mCherry protein using the constitutive pneumococcal promoter PX as a reference. Our results revealed that only three promoters PA, P2(2) and PD1 were recognized in Enterococcus faecalis, Lactococcus lactis and Escherichia coli, in the conditions tested. The maximal fluorescence was obtained with PX in all the strains, followed by the P2(2) promoter, which level of fluorescence was 2-fold compared to PA and 4-fold compared to PD1. Analysis of putative factors influencing the promoter activity in single and double transformants in E. faecalis JH2-2 demonstrated that, in general, a better expression was achieved in presence of pAM401-81. In addition, the P2(2) promoter could be regulated in a negative fashion by genes existing in the native pMB-2 plasmid other than those of the as-48 cluster, while the pH seems to affect differently the as-48 promoter expression.This work was supported in part by the Ministerio de Ciencia e Innovación project BIO2008-01708, the Plan Propio from the University of Granada (Spain) and by the Research Plan Group (BIO 160)
Ceniza y lava: revelaciones científicas junto al volcán
Para la exposición se ha contado con el apoyo del Museo de la Ciencia y el Cosmos (La Laguna, Tenerife) del Cabildo de Tenerife, uno de los centros más visitados en la comunidad canaria por su enfoque interactivo de la divulgación científica. Ceniza y lava acerca el fenómeno geológico al público general para que experimente y comprenda la naturaleza, promoviendo la percepción de lo plausible en nuestro entorno y que adquiera conocimientos básicos para actuar. A diferencia de otros eventos divulgativos similares, esta exposición busca acercar a la población la experiencia investigadora implementada por tierra, mar y aire con sus hallazgos: materiales recopilados, metodologías utilizadas y conclusiones adquiridas, y lo hace de una manera inmersiva, interactiva y didáctica. La exhibición, programada del 29 de junio de 2023 al 30 de junio de 2024, permite ver, y en algunos casos manipular, materiales e instrumentos utilizados en las investigaciones y que por primera vez se han recopilado y museizado para ser exhibidos. Se compone de dos partes:
1) Ceniza y lava. En esta primera sala se recrea la erupción a través de diversos elementos: proyecciones impactantes, datos clave en una pared, línea del tiempo geológica de La Palma, visión nocturna de la erupción a gran escala, evolución diaria de las coladas sobre una reproducción 3D de la isla de 2 m de longitud y caleidoscopio inmersivo de un tubo lávico.
2) Revelaciones científicas. La transición de una sala a otra se realiza a través de una carpa, réplica de un puesto de mando avanzado que sirvió de zona de trabajo, reunión, atención a medios e incluso de descanso del personal investigador. En la segunda sala se accede a materiales, herramientas y descubrimientos realizados en la erupción desde las perspectivas de tierra, mar y aire.
Esta exposición es fruto de la sinergia entre el personal científico de los centros CSIC en Canarias que se desplazó a la erupción y el de otras entidades como GE3BCN, el servicio de PRL del CSIC, el MUNA o el GES del Gobierno de Canarias, entre otros.Los equipos de investigación de los centros de la Delegación del CSIC en Canarias (IPNA, IEO e IGME) desempeñaron un papel crucial en la erupción de La Palma abordándola desde diferentes perspectivas: tierra, mar y aire. Su misión: entender el comportamiento del volcán y proporcionar información valiosa a las autoridades locales y equipos de emergencias para la toma de decisiones en gestión de desastres y protección de la población. Role of age and comorbidities in mortality of patients with infective endocarditis
[Purpose]: The aim of this study was to analyse the characteristics of patients with IE in three groups of age and to assess the ability of age and the Charlson Comorbidity Index (CCI) to predict mortality.
[Methods]: Prospective cohort study of all patients with IE included in the GAMES Spanish database between 2008 and 2015.Patients were stratified into three age groups:<65 years,65 to 80 years,and ≥ 80 years.The area under the receiver-operating characteristic (AUROC) curve was calculated to quantify the diagnostic accuracy of the CCI to predict mortality risk.
[Results]: A total of 3120 patients with IE (1327 < 65 years;1291 65-80 years;502 ≥ 80 years) were enrolled.Fever and heart failure were the most common presentations of IE, with no differences among age groups.Patients ≥80 years who underwent surgery were significantly lower compared with other age groups (14.3%,65 years; 20.5%,65-79 years; 31.3%,≥80 years). In-hospital mortality was lower in the <65-year group (20.3%,<65 years;30.1%,65-79 years;34.7%,≥80 years;p < 0.001) as well as 1-year mortality (3.2%, <65 years; 5.5%, 65-80 years;7.6%,≥80 years; p = 0.003).Independent predictors of mortality were age ≥ 80 years (hazard ratio [HR]:2.78;95% confidence interval [CI]:2.32–3.34), CCI ≥ 3 (HR:1.62; 95% CI:1.39–1.88),and non-performed surgery (HR:1.64;95% CI:11.16–1.58).When the three age groups were compared,the AUROC curve for CCI was significantly larger for patients aged <65 years(p < 0.001) for both in-hospital and 1-year mortality.
[Conclusion]: There were no differences in the clinical presentation of IE between the groups. Age ≥ 80 years, high comorbidity (measured by CCI),and non-performance of surgery were independent predictors of mortality in patients with IE.CCI could help to identify those patients with IE and surgical indication who present a lower risk of in-hospital and 1-year mortality after surgery, especially in the <65-year group

Late hepatic artery pseudoaneurysm: A rare complication after resection of hilar cholangiocarcinoma
We report an unusual pathological entity of a pseudoaneurysm of the right hepatic artery, which developed two years after the resection of a type II hilar cholangiocarcinoma and secondary to an excessive skeletonization for regional lymphadenectomy and neoadjuvant external-beam radiotherapy. After a sudden and massive hematemesis, a multidetector computed tomographic angiography (MDCTA) showed a hepatic artery pseudoaneurysm. Angiography with embolization of the pseudoaneurysm was attempted using microcoils with adequate patency of the hepatic artery and the occlusion of the pseudoaneurysm. A new episode of hematemesis 3 wk later revealed a partial revascularization of the pseudoaneurysm. A definitive interventional radiological treatment consisting of transarterial embolization (TAE) of the right hepatic artery with stainless steel coils and polyvinyl alcohol particles was effective and well-tolerated with normal liver function tests and without signs of liver infarction
Emergency adrenalectomy due to acute heart failure secondary to complicated pheochromocytoma: a case report
Abstract Pheochromocytomas are catecholamine producing tumors arising mostly from chromaffin cells of the adrenal medulla. The most common clinical presentation is hypertension, mainly in the form of paroxymal episodes. Cardiovascular manifestations include malignant arrhythmia and catecholamine cardiomyopathy, mimicking acute coronary syndromes and acute heart failure. There are reports of pheochromocytomas presenting as acute coronary syndrome and rapidly leading to cardiogenic shock; the failure of intensive medical treatment in these cases has prompted the need for emergency adrenalectomy as the only remaining option. We report on a case of complicated pheochromocytoma presenting as cardiogenic shock, in which emergency adrenalectomy was performed following a total lack of response to intensive medical treatment.</p
Monitoring and Interpreting External Load in Basketball: a Narrative Review
External load monitoring brings valuable information to practitioners and coaches about the physical requirements of the activity developed by their players, according to their level of competition, age and gender. This information can be used to individualize training programs and to optimize the time invested in them, making coaches and players more efficient in the process. This review asses the results reflected in the existing bibliography about the measurements of external load in the sport of basketball, synthesizing them in one article for the scientific reader.
