    A Linear Epitope in the N-Terminal Domain of CCR5 and Its Interaction with Antibody.

    The CCR5 receptor plays a role in several key physiological and pathological processes and is an important therapeutic target. Inhibition of the CCR5 axis by passive or active immunisation offers one very selective strategy for intervention. In this study we define a new linear epitope within the extracellular domain of CCR5 recognised by two independently produced monoclonal antibodies. A short peptide encoding the linear epitope can induce antibodies which recognise the intact receptor when administered colinear with a tetanus toxoid helper T cell epitope. The monoclonal antibody RoAb 13 is shown to bind to both cells and peptide with moderate to high affinity (6x10^8 and 1.2x107 M-1 respectively), and binding to the peptide is enhanced by sulfation of tyrosines at positions 10 and 14. RoAb13, which has previously been shown to block HIV infection, also blocks migration of monocytes in response to CCR5 binding chemokines and to inflammatory macrophage conditioned medium. A Fab fragment of RoAb13 has been crystallised and a structure of the antibody is reported to 2.1 angstrom resolution

    Conventional dendritic cells (cDCs) are versatile activators of immune responses that develop as part of the myeloid lineage downstream of hematopoietic stem cells. We have recently shown that in mice precursors of cDCs, but not of other leukocytes, are marked by expression of DNGR-1/CLEC9A. To genetically deplete DNGR-1-expressing cDC precursors and their progeny, we crossed Clec9a-Cre mice to Rosa-lox-STOP-lox-diphtheria toxin (DTA) mice. These mice develop signs of age-dependent myeloproliferative disease, as has been observed in other DC-deficient mouse models. However, despite efficient depletion of cDC progenitors in these mice, cells with phenotypic characteristics of cDCs populate the spleen. These cells are functionally and transcriptionally similar to cDCs in wild type control mice but show somatic rearrangements of Ig-heavy chain genes, characteristic of lymphoid origin cells. Our studies reveal a previously unappreciated developmental heterogeneity of cDCs and suggest that the lymphoid lineage can generate cells with features of cDCs when myeloid cDC progenitors are impaired

    Off-Diagonal Deformations of Kerr Metrics and Black Ellipsoids in Heterotic Supergravity

    Geometric methods for constructing exact solutions of motion equations with first order α′\alpha ^{\prime} corrections to the heterotic supergravity action implying a non-trivial Yang-Mills sector and six dimensional, 6-d, almost-K\"ahler internal spaces are studied. In 10-d spacetimes, general parametrizations for generic off-diagonal metrics, nonlinear and linear connections and matter sources, when the equations of motion decouple in very general forms are considered. This allows us to construct a variety of exact solutions when the coefficients of fundamental geometric/physical objects depend on all higher dimensional spacetime coordinates via corresponding classes of generating and integration functions, generalized effective sources and integration constants. Such generalized solutions are determined by generic off-diagonal metrics and nonlinear and/or linear connections. In particular, as configurations which are warped/compactified to lower dimensions and for Levi-Civita connections. The corresponding metrics can have (non) Killing and/or Lie algebra symmetries and/or describe (1+2)-d and/or (1+3)-d domain wall configurations, with possible warping nearly almost-K\"ahler manifolds, with gravitational and gauge instantons for nonlinear vacuum configurations and effective polarizations of cosmological and interaction constants encoding string gravity effects. A series of examples of exact solutions describing generic off-diagonal supergravity modifications to black hole/ ellipsoid and solitonic configurations are provided and analyzed.     Closing the sea surface mixed layer temperature budget from in situ observations alone: Operation Advection during BoBBLE

    Sea surface temperature (SST) is a fundamental driver of tropical weather systems such as monsoon rainfall and tropical cyclones. However, understanding of the factors that control SST variability is lacking, especially during the monsoons when in situ observations are sparse. Here we use a ground-breaking observational approach to determine the controls on the SST variability in the southern Bay of Bengal. We achieve this through the first full closure of the ocean mixed layer energy budget derived entirely from in situ observations during the Bay of Bengal Boundary Layer Experiment (BoBBLE). Locally measured horizontal advection and entrainment contribute more significantly than expected to SST evolution and thus oceanic variability during the observation period. These processes are poorly resolved by state-of-the-art climate models, which may contribute to poor representation of monsoon rainfall variability. The novel techniques presented here provide a blueprint for future observational experiments to quantify the mixed layer heat budget on longer time scales and to evaluate these processes in models

    Truncated and Helix-Constrained Peptides with High Affinity and Specificity for the cFos Coiled-Coil of AP-1

    Protein-based therapeutics feature large interacting surfaces. Protein folding endows structural stability to localised surface epitopes, imparting high affinity and target specificity upon interactions with binding partners. However, short synthetic peptides with sequences corresponding to such protein epitopes are unstructured in water and promiscuously bind to proteins with low affinity and specificity. Here we combine structural stability and target specificity of proteins, with low cost and rapid synthesis of small molecules, towards meeting the significant challenge of binding coiled coil proteins in transcriptional regulation. By iteratively truncating a Jun-based peptide from 37 to 22 residues, strategically incorporating i-->i+4 helix-inducing constraints, and positioning unnatural amino acids, we have produced short, water-stable, alpha-helical peptides that bind cFos. A three-dimensional NMR-derived structure for one peptide (24) confirmed a highly stable alpha-helix which was resistant to proteolytic degradation in serum. These short structured peptides are entropically pre-organized for binding with high affinity and specificity to cFos, a key component of the oncogenic transcriptional regulator Activator Protein-1 (AP-1). They competitively antagonized the cJun–cFos coiled-coil interaction. Truncating a Jun-based peptide from 37 to 22 residues decreased the binding enthalpy for cJun by ~9 kcal/mol, but this was compensated by increased conformational entropy (TDS ≤ 7.5 kcal/mol). This study demonstrates that rational design of short peptides constrained by alpha-helical cyclic pentapeptide modules is able to retain parental high helicity, as well as high affinity and specificity for cFos. These are important steps towards small antagonists of the cJun-cFos interaction that mediates gene transcription in cancer and inflammatory diseases

    Activation of Al2O3 surface passivation of silicon: separating bulk and surface effects

    Understanding surface passivation arising from aluminium oxide (Al2O3) films is of significant relevance for silicon-based solar cells and devices that require negligible surface recombination. This study aims to understand the competing bulk and surface lifetime effects which occur during the activation of atomic layer deposited Al2O3. We demonstrate that maximum passivation is achieved on n- and p-type silicon with activation at ∼ 450 °C, irrespective of annealing ambient. Upon stripping the Al2O3 films and re-passivating the surface using a superacid-based technique, we find the bulk lifetime of float-zone and Czochralski silicon wafers degrade at annealing temperatures > 450 °C. By accounting for this bulk lifetime degradation, we demonstrate that the chemical passivation component associated with Al2O3 remains stable at activation temperatures of 450─500 °C, achieving an SRV of  300 °C, the interface becomes Si/SixAlyO2/Al2O3 due to diffusion of aluminium into the thin silicon oxide layer
