    Dome-type carcinoma of the colon; a rare variant of adenocarcinoma resembling a submucosal tumor: a case report

    <p>Abstract</p> <p>Background</p> <p>Dome-type carcinoma (DC) is a distinct variant of colorectal adenocarcinoma and less than 10 cases have been described in the literature. Most of the previously reported cases were early lesions and no endoscopic observations have been described so far. We herein report a case of a DC invading the subserosal layer, including endoscopic findings.</p> <p>Case presentation</p> <p>A highly elevated lesion in the transverse colon was diagnosed by colonoscopy in a 77-year-old man. The tumor appeared to be similar to a submucosal tumor (SMT), however, a demarcated area of reddish and irregular mucosa was observed at the top of the tumor. There were no erosions or ulcers. Laparoscopic-assisted right hemicolectomy was performed and pathological examination revealed a well-circumscribed tumor invading the subserosal layer. The tumor was a well-differentiated adenocarcinoma associated with a dense lymphocytic infiltration and showed expansive growth. The overlying mucosal layer showed high-grade dysplasia.</p> <p>Conclusion</p> <p>The present lesion was diagnosed as a DC of the colon invading the subserosal layer. Because the association of mucosal dysplasia is common in DCs, the detection of dysplastic epithelium would be important to discriminate DCs from SMTs.</p

    Application of precise neutron focusing mirrors for neutron reflectometry: latest results and future prospects

    超精密中性子集束ミラーによる電極界面のナノ構造解析技術の実用化 --測定精度の劇的な向上に向けた大きなマイルストーン--. 京都大学プレスリリース. 2020-10-28.Neutron reflectometry (NR) is a powerful tool for providing insight into the evolution of interfacial structures, for example via operando measurements for electrode–electrolyte interfaces, with a spatial resolution of nanometres. The time resolution of NR, which ranges from seconds to minutes depending on the reflection intensity, unfortunately remains low, particularly for small samples made of state-of-the-art materials even with the latest neutron reflectometers. To overcome this problem, a large-area focusing supermirror manufactured with ultra-precision machining has been employed to enhance the neutron flux at the sample, and a gain of approximately 100% in the neutron flux was achieved. Using this mirror, a reflectivity measurement was performed on a thin cathode film on an SrTiO3 substrate in contact with an electrolyte with a small area of 15 × 15 mm. The reflectivity data obtained with the focusing mirror were consistent with those without the mirror, but the acquisition time was shortened to half that of the original, which is an important milestone for rapid measurements with a limited reciprocal space. Furthermore, a method for further upgrades that will reveal the structural evolution with a wide reciprocal space is proposed, by applying this mirror for multi-incident-angle neutron reflectometry

    PRRX1 promotes malignant properties in human osteosarcoma

    Paired related homeobox 1 (PRRX1) is a marker of limb bud mesenchymal cells, and deficiency of p53 or Rb in Prrx1-positive cells induces osteosarcoma in several mouse models. However, the regulatory roles of PRRX1 in human osteosarcoma have not been defined. In this study, we performed PRRX1 immunostaining on 35 human osteosarcoma specimens to assess the correlation between PRRX1 level and overall survival. In patients with osteosarcoma, the expression level of PRRX1 positively correlated with poor prognosis or the ratio of lung metastasis. Additionally, we found PRRX1 expression on in 143B cells, a human osteosarcoma line with a high metastatic capacity. Downregulation of PRRX1 not only suppressed proliferation and invasion but also increased the sensitivity to cisplatin and doxorubicin. When 143B cells were subcutaneously transplanted into nude mice, PRRX1 knockdown decreased tumor sizes and rates of lung metastasis. Interestingly, forskolin, a chemical compound identified by Connectivity Map analysis using RNA expression signatures during PRRX1 knockdown, decreased tumor proliferation and cell migration to the same degree as PRRX1 knockdown. These results demonstrate that PRRX1 promotes tumor malignancy in human osteosarcoma

    Unitary-model-operator approach to Λ\Lambda hypernuclei

    A method is formulated for the description of lambda hypernuclei in the framework of the unitary-model-operator approach (UMOA). The method is applied to Λ17_{\Lambda}^{17}O. A lambda-nucleon effective interaction is derived, taking the coupling of the sigma-nucleon channel into account.     Clinical outcome of patients with recurrent or refractory localized Ewing's sarcoma family of tumors: A retrospective report from the Japan Ewing Sarcoma Study Group

    [Background] Patients with Ewing's sarcoma family of tumors (ESFT) who experience relapse or progression have a poor prognosis. [Aim] This study aimed to identify the prognostic and therapeutic factors affecting overall survival (OS) of patients with recurrent or refractory localized ESFT. [Methods and results] Thirty-eight patients with localized ESFT who experienced first relapse or progression between 2000 and 2018 were retrospectively reviewed. The 5-year OS rate of the entire cohort was 48.3% (95% confidence interval, 29.9%-64.5%). Multivariate analysis of OS identified time to relapse or progression, but not stem cell transplantation (SCT), as the sole independent risk factor (hazard ratio, 35.8; P = .002). Among 31 patients who received salvage chemotherapy before local treatment, 21 received chemotherapy regimens that are not conventionally used for newly diagnosed ESFT. The objective response rate to first-line salvage chemotherapy was 55.2% in the 29 evaluable patients. Time to relapse or progression was significantly associated with response to first-line salvage chemotherapy (P = .006). [Conclusions] The present study fails to demonstrate significant clinical benefit of SCT for recurrent or refractory localized ESFT. Recently established chemotherapy regimens may increase the survival rate of patients with recurrent or refractory localized ESFT while attenuating the beneficial effect of SCT
