    PinAPL-Py: A comprehensive web-application for the analysis of CRISPR/Cas9 screens.

    Large-scale genetic screens using CRISPR/Cas9 technology have emerged as a major tool for functional genomics. With its increased popularity, experimental biologists frequently acquire large sequencing datasets for which they often do not have an easy analysis option. While a few bioinformatic tools have been developed for this purpose, their utility is still hindered either due to limited functionality or the requirement of bioinformatic expertise. To make sequencing data analysis of CRISPR/Cas9 screens more accessible to a wide range of scientists, we developed a Platform-independent Analysis of Pooled Screens using Python (PinAPL-Py), which is operated as an intuitive web-service. PinAPL-Py implements state-of-the-art tools and statistical models, assembled in a comprehensive workflow covering sequence quality control, automated sgRNA sequence extraction, alignment, sgRNA enrichment/depletion analysis and gene ranking. The workflow is set up to use a variety of popular sgRNA libraries as well as custom libraries that can be easily uploaded. Various analysis options are offered, suitable to analyze a large variety of CRISPR/Cas9 screening experiments. Analysis output includes ranked lists of sgRNAs and genes, and publication-ready plots. PinAPL-Py helps to advance genome-wide screening efforts by combining comprehensive functionality with user-friendly implementation.     An Analysis of Federal Aviation Administration Enforcement Actions Against sUAS Operators

    The Federal Aviation Administration (FAA) has promulgated regulations to govern the commercial operation of small uncrewed aircraft systems (sUAS). Compliance with these regulations is essential for maintaining safety in the National Airspace System. And if sUAS operators fail to comply with applicable federal aviation regulations, the FAA has been granted the authority to enforce these regulations. This study explores how the FAA has been exercising its enforcement power in the context of sUAS operator regulatory noncompliance. Using data obtained from a Freedom of Information Act request, this study examines 62 FAA enforcement actions levied against sUAS operators from 2012 until 2020. Key findings include trends in enforcement activity following the promulgation of Part 107 regulations; timelines associated with civil penalty and certificate action cases; and the specific regulations most frequently prosecuted by the FAA in sUAS enforcement cases

    Prioritizing Tasks in Code Repair: A Psychological Exploration of Computer Code

    The current study explored the influence of task prioritization on how computer programmers reviewed and edited code. Forty-five programmers recruited from Amazon Mechanical Turk downloaded and edited a computer program in C#. Programmers were given instructions to review the code and told to prioritize either the reputation, transparency, or performance aspects of the code, or were given no prioritization instruction. Code changes and remarks about their changes to the code were analyzed with a between-within multivariate analysis of variance. Results indicate prioritizing an aspect of the code leads to increased performance on that aspect, but with deficits to other aspects of the code. Managers may want programmers to prioritize certain aspects of code depending on the stage of development of the software (i.e., testing, rollout, etc.). However, managers should also be cognizant of the effects task prioritization has on programmer perceptions of the code as a whole

    INVESTIGATING DRILL CONSTRAINT KINEMATICS IN MALE BASEBALL PITCHERS USING MARKERLESS MOTION CAPTURE

    This study investigated the kinematic differences that pitching constraint drills elicit compared to a baseball pitch. 18 male baseball pitchers with average height (183.7 ± 5.2cm), weight (87.4 ± 9.6kg), and skill level (Professional (4), Collegiate (5), High School (9)) were included. Video was recorded using a single camera from the open side. Each pitcher threw 3 maximum effort pitches from a mound. Next, 3 maximum effort throws were recorded for 8 different throwing drills: medicine ball hook’em drill, pivot pickoff drill, foot-up rocker drill, walk-in drill, towel drill, janitor drill, drop-step drill, and long toss. Videos were processed using pitchAITM, a markerless motion capture solution. The medicine ball hook’em drill was the most different to a pitch, and the towel drill was the most similar. This work demonstrates the first collective approach to studying the biomechanics of frequently used baseball pitching constraint drills

    Depression Is transiently increased in patients undergoing two-stage revision arthroplasty

    Background: The implications of two-stage revision on mental health are poorly understood. The purpose of this study is to determine (1) whether patients undergoing two-stage revision total hip and knee arthroplasty for prosthetic joint infection were more likely to get Patient-Reported Outcomes Measurement Information System (PROMIS) Depression scores consistent with major depressive disorder (MDD) than those undergoing aseptic revision and (2) whether these symptoms resolved after the procedure. Methods: Records of all 366 patients that underwent revision total hip or knee arthroplasty from January 1, 2015, - June 20, 2019, were reviewed. Forty-two patients were excluded for missing PROMIS Depression scores or incomplete treatment. Preoperative (\u3c3 months), early postoperative (2-8 weeks), and final postoperative (6-18 months) Depression scores were collected. Patients crossing the PROMIS Depression threshold equivalent to a Patient Health Questionnaire-9 score ≥10, indicative of MDD, were evaluated. Results: More two-stage revision patients developed Depression scores indicative of MDD perioperatively than the aseptic cohort (20.0% vs 6.5%, Conclusions: Twenty percent of two-stage revision arthroplasty patients, compared to \u3c7% of aseptic revision patients, developed PROMIS Depression scores consistent with MDD during treatment. At final follow-up, a clinically significant improvement in Depression scores from baseline was evident in both cohorts

    AbstractA 44-year-old female presented with a chief complaint of left knee pain and dysfunction. The patient had a complex surgical history including patellar fracture repair, subsequent patellar ligament repair, and ultimately allograft reconstruction which was complicated by septic arthritis requiring graft resection. On presentation to our clinic, she was noted to have significant degenerative disease in addition to chronic extensor mechanism deficiency. She underwent primary total knee arthroplasty with concomitant tibial tubercle osteotomy and advancement.
