    Meal Patterns and Food Choices of Female Rats Fed a Cafeteria-Style Diet Are Altered by Gastric Bypass Surgery

    After Roux-en-Y gastric bypass surgery (RYGB), rats tend to reduce consumption of high-sugar and/or high-fat foods over time. Here, we sought to investigate the behavioral mechanisms underlying these intake outcomes. Adult female rats were provided a cafeteria diet comprised of five palatable foodstuffs varying in sugar and fat content and intake was monitored continuously. Rats were then assigned to either RYGB, or one of two control (CTL) groups: sham surgery or a nonsurgical control group receiving the same prophylactic iron treatments as RYGB rats. Post-sur-gically, all rats consumed a large first meal of the cafeteria diet. After the first meal, RYGB rats reduced intake primarily by decreasing the meal sizes relative to CTL rats, ate meals more slowly, and displayed altered nycthemeral timing of intake yielding more daytime meals and fewer nighttime meals. Collectively, these meal patterns indicate that despite being motivated to consume a cafeteria diet after RYGB, rats rapidly learn to modify eating behaviors to consume foods more slowly across the entire day. RYGB rats also altered food preferences, but more slowly than the changes in meal patterns, and ate proportionally more energy from complex carbohydrates and protein and proportionally less fat. Overall, the pattern of results suggests that after RYGB rats quickly learn to adjust their size, eating rate, and distribution of meals without altering meal number and to shift their macronutrient intake away from fat; these changes appear to be more related to postingestive events than to a fundamental decline in the palatability of food choices

    Status of scientific knowledge, recovery progress, and future research directions for the Gulf Sturgeon, Acipenser oxyrinchus desotoi Vladykov, 1955

    The Gulf Sturgeon, Acipenser oxyrinchus desotoi, is an anadromous species of Acipenseridae and native to North America. It currently inhabits and spawns in the upper reaches of seven natal rivers along the northern coast of the Gulf of Mexico from the Suwannee River, Florida, to the Pearl River, Louisiana, during spring to autumn. Next to the Alligator Gar (Atractosteus spatula), the Gulf Sturgeon is currently the largest fish species occurring in U.S. Gulf Coast rivers, attaining a length of 2.35 m and weights exceeding 135 kg, but historically attained a substantially larger size. Historically, the spawning populations existed in additional rivers from which the species has been wholly or nearly extirpated, such as the Mobile and Ochlockonee rivers, and possibly the Rio Grande River. Most Gulf Sturgeon populations were decimated by unrestricted commercial fishing between 1895–1910. Subsequently most populations remained unrecovered or extirpated due to continued harvest until the 1970s–1980s, and the construction of dams blocking access to ancestral upriver spawning grounds. Late 20th Century harvest bans and net bans enacted by the several Gulf Coast states have stabilized several populations and enabled the Suwannee River population to rebound substantially and naturally. Hatchery supplementation has not been necessary in this regard to date. Sturgeon are resilient and adaptable fishes with a geological history of 150 million years. Research undertaken since the 1970s has addressed many aspects of Gulf Sturgeon life history, reproduction, migration, population biology, habitat requirements, and other aspects of species biology. However, many knowledge gaps remain, prominently including the life history of early developmental stages in the first year of life. Natural population recovery is evident for the Suwannee River population, but seems promising as well for at least four other populations. The Pascagoula and Pearl River populations face a challenging future due a combination of natural and anthropogenic factors. These two populations, and perhaps the Escambi River population, are particularly vulnerable to periodic mass mortality due to major stochastic events including hurricanes, flooding, hypoxia, and toxic spills. The present manuscript provides a comprehensive synthesis of knowledge regarding the Gulf Sturgeon at the organismal and population levels, identifying knowledge gaps as priorities for future research. Topics not treated in the present synthesis include morphology, internal biology, physiology, and endocrinology.     A Comparison of Total Food Intake at a Personalised Buffet in People with Obesity, before and 24 Months after Roux-en-Y-Gastric Bypass Surgery

    Long-term reductions in the quantity of food consumed, and a shift in intake away from energy dense foods have both been implicated in the potent bariatric effects of Roux-en-Y gastric bypass (RYGB) surgery. We hypothesised that relative to pre-operative assessment, a stereotypical shift to lower intake would be observed at a personalised ad libitum buffet meal 24 months after RYGB, driven in part by decreased selection of high energy density items. At pre-operative baseline, participants (n = 14) rated their preference for 72 individual food items, each of these mapping to one of six categories encompassing high and low-fat choices in combination with sugar, complex carbohydrate or and protein. An 18-item buffet meal was created for each participant based on expressed preferences. Overall energy intake was reduced on average by 60% at the 24-month buffet meal. Reductions in intake were seen across all six food categories. Decreases in the overall intake of all individual macronutrient groups were marked and were generally proportional to reductions in total caloric intake. Patterns of preference and intake, both at baseline and at follow-up appear more idiosyncratic than has been previously suggested by verbal reporting. The data emphasise the consistency with which reductions in ad libitum food intake occur as a sequel of RYGB, this being maintained in the setting of a self-selected ad libitum buffet meal. Exploratory analysis of the data also supports prior reports of a possible relative increase in the proportional intake of protein after RYGB

    Crystallization and preliminary X-ray structural studies of a Melan-A pMHC-TCR complex

    Melanocytes are specialized pigmented cells that are found in all healthy skin tissue. In certain individuals, diseased melanocytes can form malignant tumours, melanomas, which cause the majority of skin-cancer-related deaths. The melanoma-associated antigenic peptides are presented on cell surfaces via the class I major histocompatibility complex (MHC). Among the melanoma-associated antigens, the melanoma self-antigen A/melanoma antigen recognized by T cells (Melan-A/MART-1) has attracted attention because of its wide expression in primary and metastatic melanomas. Here, a preliminary X-ray crystal structural study of a soluble cognate T-cell receptor (TCR) in complex with a pMHC presenting the Melan-A peptide (ELAGIGILTV) is reported. The TCR and pMHC were refolded, purified and mixed together to form complexes, which were crystallized using the sitting-drop vapour-diffusion method. Single TCR–pMHC complex crystals were cryocooled and used for data collection. Diffraction data showed that these crystals belonged to space group P4(1)/P4(3), with unit-cell parameters a = b = 120.4, c = 81.6 Å. A complete data set was collected to 3.1 Å and the structure is currently being analysed
