    Impact of Gene-Gender Effects of Adrenergic Polymorphisms on Hypothalamic-Pituitary-Adrenal Axis Activity in Depressed Patients

    Objective: There is overwhelming evidence that activation of the hypothalamic-pituitary-adrenal (HPA) system plays a major role in depression and cardiovascular disease in genetically susceptible individuals. We hypothesized that due to the multiple interactions between the sympathetic and the HPA systems via adrenoceptors, polymorphisms in these genes could have an impact on HPA axis activity in major depression. Methods: Using the dexamethasone/corticotrophin-releasing hormone (DEX/CRH) test, we investigated the association of alpha 2-adrenoceptor (ADRA2A -1291C -> G) and the beta 2-adrenoceptor gene (ADRB2 Arg16Gly) in 189 patients with major depression during the acute state of the disease and after remission. Results: Male ADRA2A -1291G allele homozygotes showed significant pretreatment HPA axis hyperactivity, with increased adrenocorticotropin (ACTH; F = 4.9, d.f. = 2, p = 0.009) and cortisol responses (F = 6.4, d.f. = 2, p = 0.003). In contrast, female ADRB2 Arg/Arg homozygotes had increased pretreatment ACTH (F = 7.17, d.f. = 2, p = 0.001) and cortisol (F = 8.95, d.f. = 2, p = 0.000) levels. Interestingly, in the respective genotypes, the stress hormones remained elevated in the second DEX/CRH test, despite a reduction in depressive symptoms. Conclusions: This study provides evidence that, depending on gender and polymorphisms, there is continuous HPA axis overdrive in a proportion of patients irrespective of the status of depression. Considering the importance of stress hormones for cardiovascular disorders, our data might suggest that these patients are at high risk of comorbidity between depression and cardiovascular disorders.     The chemokine CXCL13 in acute neuroborreliosis

    Objective Recent studies have suggested an important role of the B cell chemoattractant CXCL13 in acute neuroborreliosis (NB). Our aim was to confirm the diagnostic role of CXCL13 and to evaluate its relevance as a therapy response and disease activity marker in NB. Methods CXCL13 was measured in cerebrospinal fluid (CSF) and serum of patients with NB (n = 28), systemic borreliosis (SB, n = 9), Guillaine-Barre syndrome (GBS, n = 11), Bell's palsy (BP, n = 19), other cranial nerve palsies (CNP, n = 5), cephalgia (C, n = 20), bacterial CNS infections (B-CNS-I, n = 16) and viral CNS infections (V-CNS-I, n = 18). For follow-up studies, serial sample pairs were evaluated from 25 patients with NB (n = 56), 11 with B-CNS-I (n = 25) and 14 with V-CNS-I (n = 36). Results CSF-CXCL13 was significantly elevated in NB compared with other neurological diseases (p<0.001). Using receiver operating characteristic analysis, 337 ng/g was determined as a cut-off with a sensitivity of 96.4% and a specificity of 96.9%. Of all the parameters investigated, CSF CXCL13 showed the fastest response to antibiotic therapy, decreasing significantly (p = 0.008) within 1 week. In untreated patients, CSF CXCL13 was elevated in patients with a short duration of disease. Borrelia burgdorferi antibody index showed no significant (p = 0.356) change over follow-up. Conclusions The study confirms the relevance of CXCL13 as a diagnostic biomarker of NB and suggests that CSF CXCL13 in NB is linked to duration of disease and could be a marker of disease activity and response to antibiotic therapy

    Viewset Diffusion: (0-)image-conditioned 3D generative models from 2D data

    We present Viewset Diffusion, a diffusion-based generator that outputs 3D objects while only using multi-view 2D data for supervision. We note that there exists a one-to-one mapping between viewsets, i.e., collections of several 2D views of an object, and 3D models. Hence, we train a diffusion model to generate viewsets, but design the neural network generator to reconstruct internally corresponding 3D models, thus generating those too. We fit a diffusion model to a large number of viewsets for a given category of objects. The resulting generator can be conditioned on zero, one or more input views. Conditioned on a single view, it performs 3D reconstruction accounting for the ambiguity of the task and allowing to sample multiple solutions compatible with the input. The model performs reconstruction efficiently, in a feed-forward manner, and is trained using only rendering losses using as few as three views per viewset.     Bond Orientational Order, Molecular Motion and Free Energy of High Density DNA Mesophases

    By equilibrating condensed DNA arrays against reservoirs of known osmotic stress and examining them with several structural probes, it has been possible to achieve a detailed thermodynamic and structural characterization of the change between two distinct regions on the liquid crystalline phase digram: a higher-density hexagonally packed region with long-range bond orientational order in the plane perpendicular to the average molecular direction; and a lower-density cholesteric region with fluid-like positional order. X-rays scattering on highly ordered DNA arrays at high density and with the helical axis oriented parallel to the incoming beam showed a six-fold azimuthal modulation of the first order diffraction peak that reflects the macroscopic bond-orientational order. Transition to the less-dense cholesteric phase through osmotically controlled swelling shows the loss of this bond orientational order that had been expected from the change in optical birefringence patterns and that is consistent with a rapid onset of molecular positional disorder. This change in motion was previously inferred from intermolecular force measurements and is now confirmed by 31P\rm ^{31}P NMR. Controlled reversible swelling and compaction under osmotic stress, spanning a range of densities between 120\sim 120 mg/ml to 600\sim 600 mg/ml, allows measurement of the free energy changes throughout each phase and at the phase transition, essential information for theories of liquid-crystalline states.Comment: 14 pages, 3 figures in gif format available at http://abulafia.mgsl.dcrt.nih.gov/pics.html E-mail: [email protected]

    Splatter image: ultra-fast single-view 3D reconstruction

    We introduce the Splatter Image, an ultra-efficient approach for monocular 3D object reconstruction. Splatter Image is based on Gaussian Splatting, which allows fast and high-quality reconstruction of 3D scenes from multiple images. We apply Gaussian Splatting to monocular reconstruction by learning a neural network that, at test time, performs reconstruction in a feed-forward manner, at 38 FPS. Our main innovation is the surprisingly straightforward design of this network, which, using 2D operators, maps the input image to one 3D Gaussian per pixel. The resulting set of Gaussians thus has the form an image, the Splatter Image. We further extend the method take several images as input via cross-view attention. Owning to the speed of the renderer (588 FPS), we use a single GPU for training while generating entire images at each iteration to optimize perceptual metrics like LPIPS. On several synthetic, real, multi-category and large-scale benchmark datasets, we achieve better results in terms of PSNR, LPIPS, and other metrics while training and evaluating much faster than prior works.     Unsupervised learning of probably symmetric deformable 3D objects from images in the wild (extended abstract)

    We propose a method to learn 3D deformable object categories from raw single-view images, without external supervision. The method is based on an autoencoder that factors each input image into depth, albedo, viewpoint and illumination. In order to disentangle these components without supervision, we use the fact that many object categories have, at least approximately, a symmetric structure. We show that reasoning about illumination allows us to exploit the underlying object symmetry even if the appearance is not symmetric due to shading. Furthermore, we model objects that are probably, but not certainly, symmetric by predicting a symmetry probability map, learned end-to-end with the other components of the model. Our experiments show that this method can recover very accurately the 3D shape of human faces, cat faces and cars from single-view images, without any supervision or a prior shape model.     Transmission dynamics and prospects for the elimination of canine rabies

    Rabies has been eliminated from domestic dog populations in Western Europe and North America, but continues to kill many thousands of people throughout Africa and Asia every year. A quantitative understanding of transmission dynamics in domestic dog populations provides critical information to assess whether global elimination of canine rabies is possible. We report extensive observations of individual rabid animals in Tanzania and generate a uniquely detailed analysis of transmission biology, which explains important epidemiological features, including the level of variation in epidemic trajectories. We found that the basic reproductive number for rabies, R&lt;sub&gt;0&lt;/sub&gt;, is very low in our study area in rural Africa (&#8764;1.2) and throughout its historic global range (&#60;2). This finding provides strong support for the feasibility of controlling endemic canine rabies by vaccination, even near wildlife areas with large wild carnivore populations. However, we show that rapid turnover of domestic dog populations has been a major obstacle to successful control in developing countries, thus regular pulse vaccinations will be required to maintain population-level immunity between campaigns. Nonetheless our analyses suggest that with sustained, international commitment, global elimination of rabies from domestic dog populations, the most dangerous vector to humans, is a realistic goal
