    Genomic and phenotypic characterization of an atypical Aeromonas salmonicida strain isolated from a lumpfish and producing unusual granular structures

    Aeromonas salmonicida strains are roughly classified into two categories, typical and atypical strains. The latter mainly regroup isolates that present unusual phenotypes or hosts, comparatively to the typical strains that belong to the salmonicida subspecies. This study focuses on an uncharacterized atypical strain, M18076-11, isolated from lumpfish (Cyclopterus lumpus) and not part of the four recognized Aeromonas salmonicida subspecies. This isolate presents an unreported phenotype in the A. salmonicida species: the formation of large granular aggregates. Granules are formed of a heterogeneous mix of live and dead cells, with live cells composing the majority of the population. Even if no mechanism was determined to cause cellular aggregation, small globular structures at the cell surface were observed, which might affect granular formation. Pan-genome phylogenetic analysis indicated that this strain groups alongside the masoucida subspecies. However, phenotypic tests showed that these strains have diverging phenotypes, suggesting that M18076-11 might belong to a new subspecies. Also, a pAsal1-like plasmid, which was only reported in strains of the subspecies salmonicida, was discovered in M18076-11. This study sheds light on unsuspected diversity in A. salmonicida subspecies and stresses the need of thorough identification when a new strain is encountered, as unique traits might be discovered

    La qualité de l'effluent des systèmes de biotraitement par boues activées est souvent limitée par la performance de l'unité de décantation. Cette étude analyse les causes possibles des difficultés d'opération rencontrées dans la décantation des boues évacuées aux décanteurs secondaires d'une raffinerie de pétrole. Les objectifs visent à déterminer et à quantifier l'effet des paramètres d'opération qui affectent le fonctionnement du décanteur, en les reliant si possible à son mauvais fonctionnement, à évaluer de l'ajout d'alun et à proposer des méthodes correctives pour améliorer l'efficacité de décantation.L'étude montre que :- l'efficacité des décanteurs sera améliorée si le niveau d'oxygène dissous et la concentration en matières totales en suspension (MTS) dans le bassin d'aération sont respectivement maintenus entre 0,9 à 1,3 ppm et 4 300 à 4 800 mg/L;- le pH de la liqueur mixte ne semble pas influencer la décantabitité;- l'addition d'alun doit être évitée ou maintenue inférieure à 50 mg/L;- l'agitation et les vitesses linéaires élevées dans les conduites d'alimentation des décanteurs doivent être évitées.The wastewater treatment systems built in the seventies for treating the effluents from oil refineries often fail to comply with new environmental standards. The present study focuses mainly on the problem of sludge settling in the clarifiers. The purpose of this work was to determine the effect of various parameters on the settling characteristics of the activated sludge, to evaluate the rote of alun as a flocculating agent and to characterize the operation of the clarifiers primarily as it relates to the settling problem. The influence of the quantity of the alun added and of the stirring speed on the settling characteristics of the sludge were determined in jar tests. The effects, on the performance of the clarifiers, of the level of dissolved oxygen in the bioreactor, of the pH and of the concentration in total suspended solids (TSS) maintained in the mixed liquor, were also investigated.The studies on the sludge settleability led to the following conclusions :- the concentration in dissolved oxygen and the concentration in TSS in the aeration tank are two parameters which have a critical effect on the effluent quality. The optimal operating intervals for these parameters are from 0.9 to 1.3 ppm for dissolved oxygen and from 4 300 to 4 800 mg/L for the TSS;- the pH of the mixed liquor is not a main factor and does not seem to have any effect on the sludge settleability;- the addition of alum in excess of 50 mg/L has an adverse effect on the sludge settling characteristics. Addition of alum below this level, however, improves the clarity of the residual liquor. Nevertheless, the addition of this flocculating agent is not recommended;- the high flowrates and violent mixing in the piping system used to feed the mixed liquor from the aeration tank to the clarifiers adversely affects the solid-liquid separation in the clarifiers.By simply stopping the addition of the flocculating agent, maintaining the dissolved oxygen concentration between 1 to 2 ppm and the TSS between 4 300 to 4 800 mg/L in the wastewater treatment unit, it was possible to improve substantially the quality of the effluent and thus meet the environmental standards

    Study of mesophilic Aeromonas salmonicida A527 strain sheds light on the species’ lifestyles and taxonomic dilemma

    The Gram-negative bacterium Aeromonas salmonicida contains five subspecies: salmonicida, smithia, achromogenes, masoucida and pectinolytica. Pectinolytica is a mesophilic subspecies with the ability to thrive at a wide range of temperatures, including 37◦C, while the four other subspecies are psychrophilic, restricted to lower temperatures. The psychrophilic subspecies are known to infect a wide range of fishes. However, there is no evidence of pathogenicity for the mesophilic subspecies pectinolytica. Study of the differences between the mesophilic and psychrophilic subspecies is hampered by the lack of completely sequenced and closed genomes from the mesophilic subspecies. A previous study reported that insertion sequences, which can induce genomic rearrangements at temperatures around 25◦C, could be one of the determinants explaining the differences in lifestyle (mesophilic or psychrophilic) between the subspecies. In this study, the genome of mesophilic strain A527 of A. salmonicida was sequenced, closed and analyzed to investigate the mesophilic–psychrophilic discrepancy. This reference genome supports the hypothesis that insertion sequences are major determinants of the lifestyle differences between the A. salmonicida subspecies. Moreover, the phylogenetic analysis performed to position strain A527 within the taxonomy raises an issue regarding the intraspecies structure of A. salmonicida

    An improved continuous compositional-spread technique based on pulsed-laser deposition and applicable to large substrate areas

    A new method for continuous compositional-spread (CCS) thin-film fabrication based on pulsed-laser deposition (PLD) is introduced. This approach is based on a translation of the substrate heater and the synchronized firing of the excimer laser, with the deposition occurring through a slit-shaped aperture. Alloying is achieved during film growth (possible at elevated temperature) by the repeated sequential deposition of sub-monolayer amounts. Our approach overcomes serious shortcomings in previous in-situ implementations of CCS based on sputtering or PLD, in particular the variations of thickness across the compositional spread and the differing deposition energetics as function of position. While moving-shutter techniques are appropriate for PLD-approaches yielding complete spreads on small substrates (i.e. small as compared to distances over which the deposition parameters in PLD vary, typically about 1 cm), our method can be used to fabricate samples that are large enough for individual compositions to be analyzed by conventional techniques, including temperature-dependent measurements of resistivity and dielectric and magnetic and properties (i.e. SQUID magnetometry).     Efficacy of sacubitril/valsartan relative to a prior decompensation: the PARADIGM-HF trial

    Objectives: This study assessed whether the benefit of sacubtril/valsartan therapy varied with clinical stability. Background: Despite the benefit of sacubitril/valsartan therapy shown in the PARADIGM-HF (Prospective Comparison of ARNI with ACEI to Determine Impact on Global Mortality and Morbidity in Heart Failure) trial, it has been suggested that switching from an angiotensin-converting enzyme inhibitor or an angiotensin receptor blocker should be delayed until occurrence of clinical decompensation. Methods: Outcomes were compared among patients who had prior hospitalization within 3 months of screening (n = 1,611 [19%]), between 3 and 6 months (n = 1,009 [12%]), between 6 and 12 months (n = 886 [11%]), >12 months (n = 1,746 [21%]), or who had never been hospitalized (n = 3,125 [37%]). Results: Twenty percent of patients without prior HF hospitalization experienced a primary endpoint of cardiovascular death or heart failure (HF) hospitalization during the course of the trial. Despite the increased risk associated with more recent hospitalization, the efficacy of sacubitril/valsartan therapy did not differ from that of enalapril according to the occurrence of or time from hospitalization for HF before screening, with respect to the primary endpoint or with respect to cardiovascular or all-cause mortality. Conclusions: Patients with recent HF decompensation requiring hospitalization were more likely to experience cardiovascular death or HF hospitalization than those who had never been hospitalized. Patients who were clinically stable, as shown by a remote HF hospitalization (>3 months prior to screening) or by lack of any prior HF hospitalization, were as likely to benefit from sacubitril/valsartan therapy as more recently hospitalized patients.     Enrollment Decision-Making by Students in Forestry and Related Natural Resource Degree Programmes Globally

    A survey of 396 undergraduate and graduate students from 51 countries on 5 continents currently enrolled in Forestry or Related Natural Resource (FRNR) degree programmes was conducted of attendees to the International Union of Forest Research Organizations\u27 (IUFRO) conference in Salt Lake City, Utah, 2014. These perspectives come from some of the most active students in their respective fields. We explored the motivating reasons for enrolling in their current FRNR programme, and conversely why they may have been hesitant to do so. Results indicate that enjoyment of nature was the most important factor on average driving the decision to enroll, closely followed by job satisfaction, concern for the environment, enjoyment of outdoor recreation, being outdoors, and an interest in subject material. Hesitancy factors included earning potential, availability of funding/scholarships, and politically contentious issues. A number of significant differences were found across demographic categories. Of particular note was the greater hesitancy on the part of women and people of color to enroll in FRNR degree programmes compared to their white male counterparts. We discuss the limitations of our study arising from its international scope and imbalance of responses among countries and regions. HIGHLIGHTS Forestry and Related Natural Resources (FRNR) students from 51 countries report that enjoyment of nature was the most important factor driving their decision to enroll. Decision factors that caused hesitation included earning potential, availability of funding, and political issues. Importance factors differed significantly between genders, race/ethnicity, academic standing, world region, and social background (i.e. urban vs rural). Women and people of color from multiple world regions had a greater hesitancy to enroll in an FRNR programme than their white male counterparts. Implications for recruitment and retention include the need for continual diversity and inclusion efforts and a balance between personal preferences and employability

    Failure to communicate: Inefficiencies in voluntary incentive programs for private forest owners in Michigan

    Coordinating forest management across thousands of nonindustrial private forest (NIPF) owners is a difficult yet necessary task for state land management agencies. Voluntary Incentive Programs (VIPs) can coordinate the decentralized activities of these owners in return for services or financial incentives. However, many VIPs typically have low enrollment. Our study investigates the implementation of VIPs to increase forest management coordination among NIPFs in Michigan. We present findings from 20 semi-structured interviews with leaders of state and local land management organizations, and government officials at state natural resource agencies, and contrast their answers with those recorded from 37 interviews of NIPF owners regarding their knowledge and attitudes toward VIPs. Our interviews highlight a critical disconnect between NIPF owner motivations and VIP incentives, as well as misallocated resources for VIP promotion by state agencies, driving low enrollment. At the core, low enrollment in VIPs is generated by inadequate communication between NIPF owners and program managers, along with distrust of government agency objectives. Viewing managers as “street level bureaucrats”, civil servants whose job discretion is impacted heavily by available resources, may increase our understanding of the issues plaguing VIPs and help identify improvements to VIP design and implementation

    Influence of Sacubitril/Valsartan (LCZ696) on 30-day readmission after heart failure hospitalization

    Background: Patients with heart failure (HF) are at high risk for hospital readmission in the first 30 days following HF hospitalization. Objectives: This study sought to determine if treatment with sacubitril/valsartan (LCZ696) reduces rates of hospital readmission at 30-days following HF hospitalization compared with enalapril. Methods: We assessed the risk of 30-day readmission for any cause following investigator-reported hospitalizations for HF in the PARADIGM-HF trial, which randomized 8,399 participants with HF and reduced ejection fraction to treatment with LCZ696 or enalapril. Results: Accounting for multiple hospitalizations per patient, there were 2,383 investigator-reported HF hospitalizations, of which 1,076 (45.2%) occurred in subjects assigned to LCZ696 and 1,307 (54.8%) occurred in subjects assigned to enalapril. Rates of readmission for any cause at 30 days were 17.8% in LCZ696-assigned subjects and 21.0% in enalapril-assigned subjects (odds ratio: 0.74; 95% confidence interval: 0.56 to 0.97; p = 0.031). Rates of readmission for HF at 30-days were also lower in subjects assigned to LCZ696 (9.7% vs. 13.4%; odds ratio: 0.62; 95% confidence interval: 0.45 to 0.87; p = 0.006). The reduction in both all-cause and HF readmissions with LCZ696 was maintained when the time window from discharge was extended to 60 days and in sensitivity analyses restricted to adjudicated HF hospitalizations. Conclusions: Compared with enalapril, treatment with LCZ696 reduces 30-day readmissions for any cause following discharge from HF hospitalization

    Effects of sacubitril/valsartan in the PARADIGM-HF Trial (Prospective Comparison of ARNI with ACEI to Determine Impact on Global Mortality and Morbidity in Heart Failure) according to background therapy

    Background—In the PARADIGM-HF trial (Prospective Comparison of ARNI with ACEI to Determine Impact on Global Mortality and Morbidity in Heart Failure), the angiotensin receptor neprilysin inhibitor sacubitril/valsartan was more effective than the angiotensin-converting enzyme inhibitor enalapril in patients with heart failure and reduced ejection fraction. We examined whether this benefit was consistent irrespective of background therapy. Methods and Results—We examined the effect of study treatment in the following subgroups: diuretics (yes/no), digitalis glycoside (yes/no), mineralocorticoid receptor antagonist (yes/no), and defibrillating device (implanted defibrillating device, yes/no). We also examined the effect of study drug according to β-blocker dose (≥50% and <50% of target dose) and according to whether patients had undergone previous coronary revascularization. We analyzed the primary composite end point of cardiovascular death or heart failure hospitalization, as well as cardiovascular death. Most randomized patients (n=8399) were treated with a diuretic (80%) and β-blocker (93%); 47% of those taking a β-blocker were treated with ≥50% of the recommended dose. In addition, 4671 (56%) were treated with a mineralocorticoid receptor antagonist, 2539 (30%) with digoxin, and 1243 (15%) had a defibrillating device; 2640 (31%) had undergone coronary revascularization. Overall, the sacubitril/valsartan versus enalapril hazard ratio for the primary composite end point was 0.80 (95% confidence interval, 0.73–0.87; P<0.001) and for cardiovascular death was 0.80 (0.71–0.89; P<0.001). The effect of sacubitril/valsartan was consistent across all subgroups examined. The hazard ratio for primary end point ranged from 0.74 to 0.85 and for cardiovascular death ranged from 0.75 to 0.89, with no treatment-by-subgroup interaction. Conclusions—The benefit of sacubitril/valsartan, over an angiotensin-converting enzyme inhibitor, was consistent regardless of background therapy and irrespective of previous coronary revascularization or β-blocker dose
