    Sperm Lipid Composition in Early Diverged Fish Species: Internal vs. External Mode of Fertilization

    The lipid composition of sperm membranes is crucial for fertilization and differs among species. As the evolution of internal fertilization modes in fishes is not understood, a comparative study of the sperm lipid composition in freshwater representatives of externally and internally fertilizing fishes is needed for a better understanding of taxa-specific relationships between the lipid composition of the sperm membrane and the sperm physiology. The lipidomes of spermatozoa from stingray, a representative of cartilaginous fishes possessing internal fertilization, and sterlet, a representative of chondrostean fishes with external fertilization, have been studied by means of nuclear magnetic resonance (NMR), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS), electrospray MS, gas chromatography-(GC) MS, and thin-layer chromatography (TLC). NMR experiments revealed higher cholesterol content and the presence of phosphatidylserine in stingray compared to sterlet sperm. Unknown MS signals could be assigned to different glycosphingolipids in sterlet (neutral glycosphingolipid Gal-Cer(d18:1/16:0)) and stingray (acidic glycosphingolipid sulpho-Gal-Cer(d18:1/16:0)). Free fatty acids in sterlet sperm indicate internal energy storage. GC-MS experiments indicated a significant amount of adrenic acid, but only a low amount of docosahexaenoic acid in stingray sperm. In a nutshell, this study provides novel data on sperm lipid composition for freshwater stingray and sterlet possessing different modes of fertilization

    Visando o desenvolvimento das técnicas de reprodução em peixes, foi realizada a caracterização e a criopreservação do sêmen de Brycon cephalus. As características seminais observadas foram: sêmen de coloração leitosa quase transparente, com um volume médio de 4mL e concentração espermática de 9.617±1.630 x 10(6) espermatozóides/mm³. Os espermatozóides (comprimento = 31,288±4,47 µm) são do tipo aquasperm, apresentando uma pequena cabeça arredondada (comprimento= 1,752±0,18 µm; largura = 1,752±0,17 µm) sem vesícula acrosomal, com um núcleo com cromatina altamente compactada, formando grumos grosseiros e um complexo centriolar localizado na fossa nuclear; uma peça intermediária (comprimento = 2,561±0,44 µm) que se estreita no sentido antero-posterior, um canal citoplasmático e um flagelo (comprimento = 29,521±4,37 µm). Os testes de fertilização com sêmen descongelado demonstraram um efeito significante (alfa = 0,05) no aumento da fertilidade quando utilizado o diluente tipo B, para ambos os tipos de palhetas, 0,5mL e 4,0mL. Não foi constatado efeito significativo (alfa = 0,05) entre a utilização de palhetas de 0,5mL ou de 4,0mL em relação à porcentagem de eclosão.Aiming to improve fish reproduction techniques, the characterization and cryopreservation of semen of Brycon cephalus were performed. The seminal characteristics observed were: an almost transparent, milky semen with a mean volume of 4mL, and sperm concentration of 9.617±1.630 x 10(6) spermatozoa/mm³. Spermatozoa (length = 31.288 ±4.47µm) were of the aquasperm type and displayed a small, round head (length = 1.727 ±0.18 µm; width = 1.752 ±0.17µm) without acrosomal vesicle, nucleus with highly condensed chromatin forming coarse clots and centriolar complex located in the nuclear fossa; a midpiece (length = 2.561±0.44µm), narrowed rearward, with a cytoplasmic canal; and a flagellum (length = 29.521 ±4.37µm). Fertilization tests with thawed semen demonstrated a significant effect (alpha= 0.05) on the increase of thawed semen fertility with diluent type B in both 0.5mL and 4.0 mL straws.     Reproductive cycle of the Neotropical cichlid yellow peacock bass Cichla kelberi: A novel pattern of testicular development

    The present study describes the testicular maturation phases (associating the germ cells development and the morphological changes suffered by the germinal epithelium along the whole year), and the testicular morphology in the yellow peacock bass Cichla kelberi, relating it to other species. For this purpose, 78 specimens were studied according conventional techniques of light microscope. The testes in C. kelberi were classified as unrestricted spermatogonial lobular, an apomorphic characteristic in the recent groups of Teleost. Furthermore, were defined five testicular maturation phases: Preparatory phase; Early Germinal Epithelium Development; Mid Germinal Epithelium Development; Late Germinal Epithelium Development and; Regression. Similar classifications were described to other species indicating that the testicular classifications based on this propose, can be applied to lots of fishes. However, besides it similarity, the testicular reproductive cycle of C. kelberifollows a different pattern in the Regression phase, on which the gonadal restructuration and the spermatogonial proliferation gathers at the same time. So, the testes in C. kelberi never return to the Preparatory phase to start a new reproductive cycle, being this one present only at the first reproductive cycle in this species. This fact also explains the absence of individuals totally spent after their first reproductive cycle

    Effects of cryoprotectants and low temperatures on hatching and abnormal embryo development of Prochilodus lineatus (Characiformes: Prochilodontidae)

    ABSTRACT This study evaluated the effect of the cryoprotectants and the low temperatures on the embryonic development of Prochilodus lineatus, describing their main morphological alterations. On chilling sensitivity test, the survival rates at the twenty somites stage (20S) were 53.6% at 0ºC, and 100% in 5ºC. To test toxicity, the embryos were exposed to a graded series of 1,2-Propanediol (PROP), dimethyl sulfoxide (Me2SO4) and glycerol (GLY), terminating in a solution of high osmolarity. There was no significant difference in the embryos survival of toxicity test between series of PROP and Me2SO4 in the 6S and 20S. In the cooling protocols, were evaluated the effects of low temperature associated with cryoprotectants. At 5ºC, PROP showed survival rates above 75% in the gastrula stage (G) and above 90% in the 6S and 20S stages. High rates of abnormalities were observed, and the most recurrent were: small bodies, fins presenting uncontrolled cell growth, membrane rupture, and retraction. These results demonstrate the need to use cryoprotectant solutions, even when there is no ice nucleation, and, on the other hand, shows that high cryoprotectant concentrations promote numerous morphological lesions, compromising normal embryonic development

    Oogenesis in Laetacara araguaiae (Ottoni and Costa, 2009) (Labriformes: Cichlidae)

    We aimed to analyze the oogenesis of adult females of the cichlid fish Laetacara araguaiae. The specimens' gonads were removed and processed for light and transmission electron microscopy. Oogenesis in L. araguaiae showed the following characteristics: a germinal epithelium with three types of oogonia (A-undifferentiated, A-differentiated and B-oogonia), oocytes at meiotic prophase stage and ovarian follicle formation. Oocytes showing primary growth with pre-vitellogenic and cortical alveolus were observed. Similar to data for other cichlids, oocytes in secondary growth or vitellogenesis were characterized by the initial deposition of yolk microgranules. The event that characterizes the maturation stage is nucleolus migration, also called the germinal vesicle, to the oocyte periphery in the direction of the micropyle. The follicular complex undergoes several changes throughout the oocyte stages. To the best of our knowledge this study is the first to describe L. araguaiae oogenesis. Moreover, this study is the first step to better understand the reproductive biology of this species, which shows great potential for use as an ornamental fish.Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP
