    Enneagram Collaborative Observations

    The way we interact is important so that we can have successful and efficient lifestyles. The way we treat people is important. We create different environments with the interactions we choose to make, and sometimes those interactions are often confusing to understand. Human connection boils down to open communication and honesty. Does knowing somebody’s personality type make it easier to collaborate with them? This project looks at the interactions between creatives based on their Enneagram Personality type. The Enneagram is broken down into 9 interconnecting personalities. These numbers represent more apparent personality traits as well as common-ground and compatibility factors between the numbers. When it comes to collaboration, especially in a creative field, working with others is inevitable, and empathy for others is crucial. This attitude engenders a healthy relationship between people, and it allows us to think more objectively with a basic understanding of the other person’s needs. By using my peers as a foundation for my artistic expression, I had the opportunity to learn more about each number in depth, and create displays that show interactivity between enneagram numbers to support the methods in creative collaborative learning

    A comparison of the effectiveness of teaching volleyball through the "fingertip" method and the "attack" method

    The purpose of this study was to compare the effectiveness of teaching volleyball to college women through the "Fingertip" and "Attack" methods. Effectiveness was considered in terms of the results of competitive play, the number of points scored and the reduction of ball-handling fouls in game situations, and the achievement of earlier, later, and continued success in playing as measured by Clifton's "Single Hit Volley Test for Women's Volleyball." The "Fingertip" method consisted primarily of the commonly recommended fingertip skills while the "Attack" method consisted primarily of the fisting skills recommended by Davis. Thirty-seven college women served as subjects for the study. The study consisted of fourteen class meetings. The subjects were divided into a control and an experimental group and were instructed by the "Fingertip" and "Attack" methods respectively. The groups which were equated according to skill by Pretest scores on the Clifton Test received separate classes of instruction and practice

    CORRECTION OF THE POST – NECKING TRUE STRESS – STRAIN DATA USING INSTRUMENTED NANOINDENTATION

    The study of large plastic deformations has been the focus of numerous studies particularly in the metal forming processes and fracture mechanics fields. A good understanding of the plastic flow properties of metallic alloys and the true stresses and true strains induced during plastic deformation is crucial to optimize the aforementioned processes, and to predict ductile failure in fracture mechanics analyzes. Knowledge of stresses and strains is extracted from the true stress-strain curve of the material from the uniaxial tensile test. In addition, stress triaxiality is manifested by the neck developed during the last stage of a tensile test performed on a ductile material. This necking phenomenon is the factor responsible for deviating from uniaxial state into a triaxial one, then, providing an inaccurate description of the material’s behavior after the onset of necking The research of this dissertation is aimed at the development of a correction method for the nonuniform plastic deformation (post-necking) portion of the true stress- strain curve. The correction proposed is based on the well-known relationship between hardness and flow (yield) stress, except that instrumented nanoindentation hardness is utilized rather than conventional macro or micro hardness. Three metals with different combinations of strain hardening behavior and crystal structure were subjected to quasi- static tensile tests: power-law strain hardening low carbon G10180 steel (BCC) and electrolytic tough pitch copper C11000 (FCC), and linear strain hardening austenitic stainless steel S30400 (FCC). Nanoindentation hardness values, measured on the broken tensile specimen, were converted into flow stress values by means of the constraint factor from Tabor’s, the representative plastic strain and the post-test true plastic strains measured. Micro Vickers hardness testing was carried out on the sample as well. The constraint factors were 5.5, 4.5 and 4.5 and the representative plastic strains were 0.028, 0.062 and 0.061 for G101800, C11000 and S30400 respectively. The established corrected curves relating post-necking flow stress to true plastic strain turned out to be well represented by a power-law function. Experimental results dictated that a unique single value for and for is not appropriate to describe materials with different plastic behaviors. Therefore, Tabor’s equation, along with the representative plastic strain concept, has been misused in the past. The studied materials exhibited different nanohardness and plastic strain distributions due to their inherently distinct elasto-plastic response. The proposed post- necking correction separates out the effect of triaxiality on the uniaxial true stress-strain curve provided that the nanohardness-flow stress relationship is based on uniaxial values of stress. Some type of size effect, due to the microvoids at the tip of the neck, influenced nanohardness measurements. The instrumented nanoindentation technique proved to be a very suitable method to probe elasto-plastic properties of materials such as nanohardness, elastic modulus, and quasi-static strain rate sensitivity among others. Care should be taken when converting nanohardness to Vickers and vice versa due to their different area definition used. Nanohardness to Vickers ratio oscillated between 1.01 and 1.1

    T-cell subpopulations αβ and γδ in cord blood of very preterm infants : The influence of intrauterine infection

Immune response mechanisms in this group of patients and factors that influence cord blood mononuclear cell populations remain poorly understood and are considered insufficient. However, competent immune functions of the cord blood mononuclear cells are also described. The aim of this work was to evaluate the T-cell population (CD3+) with its subpopulations bearing T-cell receptor (TCR) αβ or TCR γδ in the cord blood of preterm infants born before 32 weeks of gestation by mothers with or without an intrauterine infection. Being a pilot study, it also aimed at feasibility check and assessment of an expected effect size. The cord blood samples of 46 infants age were subjected to direct immunofluorescent staining with monoclonal antibodies and then analyzed by flow cytometry. The percentage of CD3+ cells in neonates born by mothers with diagnosis of intrauterine infection was significantly lower than in neonates born by mothers without infection (p = 0.005; Mann-Whitney U test). The number of cells did not differ between groups. Infection present in the mother did not have an influence on the TCR αβ or TCR γδ subpopulations. Our study contributes to a better understanding of preterm infants' immune mechanisms, and sets the stage for further investigations.Peer reviewedFinal Published versio

    A Comparison of Red Fluorescent Proteins to Model DNA Vaccine Expression by Whole Animal In Vivo Imaging

    DNA vaccines can be manufactured cheaply, easily and rapidly and have performed well in pre-clinical animal studies. However, clinical trials have so far been disappointing, failing to evoke a strong immune response, possibly due to poor antigen expression. To improve antigen expression, improved technology to monitor DNA vaccine transfection efficiency is required. In the current study, we compared plasmid encoded tdTomato, mCherry, Katushka, tdKatushka2 and luciferase as reporter proteins for whole animal in vivo imaging. The intramuscular, subcutaneous and tattooing routes were compared and electroporation was used to enhance expression. We observed that overall, fluorescent proteins were not a good tool to assess expression from DNA plasmids, with a highly heterogeneous response between animals. Of the proteins used, intramuscular delivery of DNA encoding either tdTomato or luciferase gave the clearest signal, with some Katushka and tdKatushka2 signal observed. Subcutaneous delivery was weakly visible and nothing was observed following DNA tattooing. DNA encoding haemagglutinin was used to determine whether immune responses mirrored visible expression levels. A protective immune response against H1N1 influenza was induced by all routes, even after a single dose of DNA, though qualitative differences were observed, with tattooing leading to high antibody responses and subcutaneous DNA leading to high CD8 responses. We conclude that of the reporter proteins used, expression from DNA plasmids can best be assessed using tdTomato or luciferase. But, the disconnect between visible expression level and immunogenicity suggests that in vivo whole animal imaging of fluorescent proteins has limited utility for predicting DNA vaccine efficacy

    Altered splicing of the BIN1 muscle-specific exon in humans and dogs with highly progressive centronuclear myopathy

    Amphiphysin 2, encoded by BIN1, is a key factor for membrane sensing and remodelling in different cell types. Homozygous BIN1 mutations in ubiquitously expressed exons are associated with autosomal recessive centronuclear myopathy (CNM), a mildly progressive muscle disorder typically showing abnormal nuclear centralization on biopsies. In addition, misregulation of BIN1 splicing partially accounts for the muscle defects in myotonic dystrophy (DM). However, the muscle-specific function of amphiphysin 2 and its pathogenicity in both muscle disorders are not well understood. In this study we identified and characterized the first mutation affecting the splicing of the muscle-specific BIN1 exon 11 in a consanguineous family with rapidly progressive and ultimately fatal centronuclear myopathy. In parallel, we discovered a mutation in the same BIN1 exon 11 acceptor splice site as the genetic cause of the canine Inherited Myopathy of Great Danes (IMGD). Analysis of RNA from patient muscle demonstrated complete skipping of exon 11 and BIN1 constructs without exon 11 were unable to promote membrane tubulation in differentiated myotubes. Comparative immunofluorescence and ultrastructural analyses of patient and canine biopsies revealed common structural defects, emphasizing the importance of amphiphysin 2 in membrane remodelling and maintenance of the skeletal muscle triad. Our data demonstrate that the alteration of the muscle-specific function of amphiphysin 2 is a common pathomechanism for centronuclear myopathy, myotonic dystrophy, and IMGD. The IMGD dog is the first faithful model for human BIN1-related CNM and represents a mammalian model available for preclinical trials of potential therapies

    Health workers' perspectives on asthma care coordination between primary and specialised healthcare in the COVID-19 pandemic: a protocol for a qualitative study in Ecuador and Brazil.

    INTRODUCTION: Asthma is a common long-term disorder and strategies to improve asthma control are still a challenge. Integrated delivery of health systems is critical for effective asthma care: there is limited information on experiences of care coordination for asthma from Latin America, especially on perspectives of health personnel and in the context of the COVID-19 pandemic. METHODS AND ANALYSIS: This protocol details a qualitative approach to analyse health workers' perspectives of healthcare coordination for asthma control during COVID-19 pandemic in Ecuador and Brazil, at primary and specialised levels, through in-depth semistructured interviews using a video communications platform. The analysis will identify knowledge and perspectives based on coordination of clinical information, clinical management and administrative coordination. Theoretical sampling will be used to obtain approximately equal numbers of women and men within each level of healthcare; data saturation will be used to determine sample size. Transcripts will be analysed using content-coding procedures to mark quotations related to major topics and subthemes included in the interview guide, and narrative analysis will be based on a theoretical framework for healthcare coordination to identify new themes and subthemes. ETHICS AND DISSEMINATION: Ethical approval was obtained from the ethics committees of Hospital General Docente CalderĂłn, Quito, Ecuador; and Universidade Federal da Bahia, Salvador, Brazil. The findings of this study will be disseminated through peer-reviewed articles, conference presentations and condensed summaries for key stakeholders and partners

    The Association between Intrauterine Inflammation and Spontaneous Vaginal Delivery at Term: A Cross-Sectional Study

    BACKGROUND:Different factors contribute to the onset of labor at term. In animal models onset of labor is characterized by an inflammatory response. The role of intrauterine inflammation, although implicated in preterm birth, is not yet established in human term labor. We hypothesized that intrauterine inflammation at term is associated with spontaneous onset of labor. METHODS/RESULTS:In two large urban hospitals in the Netherlands, a cross-sectional study of spontaneous onset term vaginal deliveries and elective caesarean sections (CS), without signs of labor, was carried out. Placentas and amniotic fluid samples were collected during labor and/or at delivery. Histological signs of placenta inflammation were determined. Amniotic fluid proinflammatory cytokine concentrations were measured using ELISA. A total of 375 women were included. In term vaginal deliveries, more signs of intrauterine inflammation were found than in elective CS: the prevalence of chorioamnionitis was higher (18 vs 4%, p = 0.02) and amniotic fluid concentration of IL-6 was higher (3.1 vs 0.37 ng/mL, p<0.001). Similar results were obtained for IL-8 (10.93 vs 0.96 ng/mL, p<0.001) and percentage of detectable TNF-alpha (50 vs 4%, p<0.001). CONCLUSIONS:This large cross-sectional study shows that spontaneous term delivery is characterized by histopathological signs of placenta inflammation and increased amniotic fluid proinflammatory cytokines

    Coping (together) with hate: Strategies used by Mexican-origin families in response to racial–ethnic discrimination

    Coping in the context of racial–ethnic discrimination is often framed as an individualistic process, where the focus is on how the individual deals with the racialized stressor to mitigate its negative effects. However, individuals exist within social contexts including the family and coping processes may operate interdependently as well. Further, racialized stressors have the potential to disrupt the entire family system, regardless of whether the experience in that moment is shared among all its members. Despite these realities, few studies have considered how Latinx youth and their parents may cope together in the face of racial–ethnic discrimination. To address this gap, we analyzed focus group data from Mexican-origin adolescents (n = 17; Mage = 12.8; 71% girls) and their parents (n = 17; Mage = 42.8; 82% mothers) to explore the coping strategies used in response to racial–ethnic discrimination. An inductive thematic analysis identified a broad range of coping strategies representing both individualistic and interdependent approaches to deal with racial–ethnic discrimination. Strategies included (a) reframing (with pride) and ignoring an encounter, (b) standing up for oneself, (c) talking issues out, (d) problem-solving together, and (e) protection tactics. These findings provide evidence for the ways in which Mexican-origin families help adolescents cope with racial–ethnic discrimination and offer a glimpse as to how adolescents may help their families cope as well. Future research is needed to further explore the interdependent nature of coping as Latinx family members protect and support one another in the face of pervasive racialized stressors
