    Exploratory design studies conducted to establish the configuration of an augmentor wing vruise blowing (valveless) system in a 150-passenger STOL airplane were reported in NASA CR-114570. Those studies have been updated to incorporate the results of static rig, flow duct, and wind tunnel tests. Minor adjustments in duct flow velocity, flap length, and blowing nozzle geometry were incorporated to provide airplane characteristics that minimize takeoff gross weight and achieve sideline noise objectives for an advanced commercial STOL airplane

    Characteristic differences between the promoters of intron-containing and intronless ribosomal protein genes in yeast

    <p>Abstract</p> <p>Background</p> <p>More than two thirds of the highly expressed ribosomal protein (RP) genes in <it>Saccharomyces cerevisiae </it>contain introns, which is in sharp contrast to the genome-wide five percent intron-containing genes. It is well established that introns carry regulatory sequences and that the transcription of RP genes is extensively and coordinately regulated. Here we test the hypotheses that introns are innately associated with heavily transcribed genes and that introns of RP genes contribute regulatory TF binding sequences. Moreover, we investigate whether promoter features are significantly different between intron-containing and intronless RP genes.</p> <p>Results</p> <p>We find that directly measured transcription rates tend to be lower for intron-containing compared to intronless RP genes. We do not observe any specifically enriched sequence motifs in the introns of RP genes other than those of the branch point and the two splice sites. Comparing the promoters of intron-containing and intronless RP genes, we detect differences in number and position of Rap1-binding and IFHL motifs. Moreover, the analysis of the length distribution and the folding free energies suggest that, at least in a sub-population of RP genes, the 5' untranslated sequences are optimized for regulatory function.</p> <p>Conclusion</p> <p>Our results argue against the direct involvement of introns in the regulation of transcription of highly expressed genes. Moreover, systematic differences in motif distributions suggest that RP transcription factors may act differently on intron-containing and intronless gene promoters. Thus, our findings contribute to the decoding of the RP promoter architecture and may fuel the discussion on the evolution of introns.</p

    Gene expression trees in lymphoid development

    <p>Abstract</p> <p>Background</p> <p>The regulatory processes that govern cell proliferation and differentiation are central to developmental biology. Particularly well studied in this respect is the lymphoid system due to its importance for basic biology and for clinical applications. Gene expression measured in lymphoid cells in several distinguishable developmental stages helps in the elucidation of underlying molecular processes, which change gradually over time and lock cells in either the B cell, T cell or Natural Killer cell lineages. Large-scale analysis of these <it>gene expression trees </it>requires computational support for tasks ranging from visualization, querying, and finding clusters of similar genes, to answering detailed questions about the functional roles of individual genes.</p> <p>Results</p> <p>We present the first statistical framework designed to analyze gene expression data as it is collected in the course of lymphoid development through clusters of co-expressed genes and additional heterogeneous data. We introduce dependence trees for continuous variates, which model the inherent dependencies during the differentiation process naturally as gene expression trees. Several trees are combined in a mixture model to allow inference of potentially overlapping clusters of co-expressed genes. Additionally, we predict microRNA targets.</p> <p>Conclusion</p> <p>Computational results for several data sets from the lymphoid system demonstrate the relevance of our framework. We recover well-known biological facts and identify promising novel regulatory elements of genes and their functional assignments.     SVC: structured visualization of evolutionary sequence conservation

    We have developed a web application for the detailed analysis and visualization of evolutionary sequence conservation in complex vertebrate genes. Given a pair of orthologous genes, the protein-coding sequences are aligned. When these sequences are mapped back onto their encoding exons in the genomes, a scaffold of the conserved gene structure naturally emerges. Sequence similarity between exons and introns is analysed and embedded into the gene structure scaffold. The visualization on the SVC server provides detailed information about evolutionarily conserved features of these genes. It further allows concise representation of complex splice patterns in the context of evolutionary conservation. A particular application of our tool arises from the fact that around mRNA editing sites both exonic and intronic sequences are highly conserved. This aids in delineation of these sites.     T-Reg Comparator: an analysis tool for the comparison of position weight matrices

    T-Reg Comparator is a novel software tool designed to support research into transcriptional regulation. Sequence motifs representing transcription factor binding sites are usually encoded as position weight matrices. The user inputs a set of such weight matrices or binding site sequences and our program matches them against the T-Reg database, which is presently built on data from the Transfac [E. Wingender (2004) In Silico Biol., 4, 55–61] and Jaspar [A. Sandelin, W. Alkema, P. Engstrom, W. W. Wasserman and B. Lenhard (2004) Nucleic Acids Res., 32, D91–D94]. Our tool delivers a detailed report on similarities between user-supplied motifs and motifs in the database. Apart from simple one-to-one relationships, T-Reg Comparator is also able to detect similarities between submatrices. In addition, we provide a user interface to a program for sequence scanning with weight matrices. Typical areas of application for T-Reg Comparator are motif and regulatory module finding and annotation of regulatory genomic regions. T-Reg Comparator is available at

    Performance of the 19XB 10-Stage Axial-Flow Compressor with Altered Blade Angles

    Previous performance data of the 19XB axial-flow compressor indicated that the outlet guide vanes and possibly the inlet guide vanes were stalling. Calculations were made to determine if these adverse conditions could be eliminated and if the manufacturer's design specifications could be more nearly approached by altering the blade angles of the first few compression stages as well as the outlet guide vanes. With the blade angles altered, experimental data were taken at compressor speeds of 8500 to 17,000 rpm with inlet-air conditions of 7.4 inches of mercury absolute and 59 0 F. The temperature-rise efficiency increased with speed from 0.70 at 8500 rpm to 0.74 at 13,600 rpm and dropped gradually to 0.70 at 17,000 rpm. At the design speed of 17,000 rpm, the pressure ratio at the peak efficiency point was 3.63. The maximum pressure ratio at design speed was 4.15 at an equivalent weight flow of 29.8 pounds per second. The altered compressor operated very .near the design specifications of pressure ratio and equivalent weight flow. At the high speeds, the peak adiabatic temperature-rise efficiency was increased 0.02 to 0,06 by altering the blade angles. The peak pressure ratio was increased 0.29 at design speed (17,000 rpm) and 0.05 and 0.13 at 11,900 and 13,600 rpm, respectively. The equivalent weight flow through the altered compressor was reduced 2 pounds per second at 15,300 and 17,000 rpm, as was expected from the design calculations. As extreme caution was taken not to surge the compressor violently, the point of minimum air flow may not have been reached in the present investigation and in a previous investigation. A true comparison of the pressure ratios obtained at the high speeds therefore cannot be made
