A statistical system management method to tackle data uncertainty when using key performance indicators of the balanced scorecard
[EN] This work is focused on the development of a graphical method using statistical non-parametric tests for randomness and parametric tests to detect significant trends and shifts in key performance indicators from balanced scorecards. It provides managers and executives with a tool to determine if processes are improving or decaying.
The method tackles the hitherto unresolved problem of data uncertainty due to sample size for key performance indicators on scorecards. Adsorption of human fibrinogen and albumin onto hydrophobic and hydrophilic Ti6Al4V powder
Adsorption of proteins on solid surfaces has been widely studied because of its importance in various biotechnological, medical and technical applications, such as medical implants or biosensors. One of the main problems is the adsorption-induced conformational changes because they often modify the biological activity of the proteins, which is believed to be a key factor on the subsequent cellular adhesion. The aim of this work is the study of the adsorption of human fibrinogen (Fg) and human serum albumin (HSA) onto Ti6Al4V particles, commercially available on different size, that are used to elaborate scaffolds to provide structural support to cell proliferation, promoting tissue development and bone regeneration among others. The study was done through the analysis of the adsorption isotherms and the electrical characterization of surfaces after adsorption in terms of the zeta potential (ζ). From this analysis it seems that Fg adsorbs preferentially vertically oriented (end-on) and HSA moves sequentially over the surface of the Ti6Al4V particles through dimmer formation, allowing adsorption progress over this initial bilayer. The zeta potential values of both proteins remain constant when the monolayer is formed. The study also extends the analysis of both adsorption behaviour and ζ potential characterization factors to the influence of the substrate hydrophobicity as this property can be modified for the Ti6Al4V by irradiating it with ultraviolet light (UV-C) without changes on its chemical composition [1], [2]. Effect of Co-Inoculation with Mycorrhiza and Rhizobia on the Nodule Trehalose Content of Different Bean Genotypes
Studies on Rhizobium-legume symbiosis show that trehalose content in nodules under drought stress correlates positively with an increase in plant tolerance to this stress. Fewer reports describe trehalose accumulation in mycorrhiza where, in contrast with rhizobia, there is no flux of carbohydrates from the microsymbiont to the plant. However, the trehalose dynamics in the Mycorrhiza-Rhizobium-Legume tripartite symbiosis is unknown. The present study explores the role of this tripartite symbiosis in the trehalose content of nodules grown under contrasting moisture conditions. Three wild genotypes (P. filiformis, P. acutifolis and P. vulgaris) and two commercial genotypes of Phaseolus vulgaris (Pinto villa and Flor de Mayo) were used. Co-inoculation treatments were conducted with Glomus intraradices and a mixture of seven native rhizobial strains, and trehalose content was determined by GC/MS. The results showed a negative effect of mycorrhizal inoculation on nodule development, as mycorrhized plants showed fewer nodules and lower nodule dry weight compared to plants inoculated only with Rhizobium. Mycorrhizal colonization was also higher in plants inoculated only with Glomus as compared to plants co-inoculated with both microsymbionts. In regard to trehalose, co-inoculation negatively affects its accumulation in the nodules of each genotype tested. However, the correlation analysis showed a significantly positive correlation between mycorrhizal colonization and nodule trehalose content
A personalized intervention to prevent depression in primary care: cost-effectiveness study nested into a clustered randomized trial
Background: Depression is viewed as a major and increasing public health issue, as it causes high distress in the people experiencing it and considerable financial costs to society. Efforts are being made to reduce this burden by preventing depression. A critical component of this strategy is the ability to assess the individual level and profile of risk for the development of major depression. This paper presents the cost-effectiveness of a personalized intervention based on the risk of developing depression carried out in primary care, compared with usual care. Methods: Cost-effectiveness analyses are nested within a multicentre, clustered, randomized controlled trial of a personalized intervention to prevent depression. The study was carried out in 70 primary care centres from seven cities in Spain. Two general practitioners (GPs) were randomly sampled from those prepared to participate in each centre (i.e. 140 GPs), and 3326 participants consented and were eligible to participate. The intervention included the GP communicating to the patient his/her individual risk for depression and personal risk factors and the construction by both GPs and patients of a psychosocial programme tailored to prevent depression. In addition, GPs carried out measures to activate and empower the patients, who also received a leaflet about preventing depression. GPs were trained in a 10- to 15-h workshop. Costs were measured from a societal and National Health care perspective. Qualityadjustedlife years were assessed using the EuroQOL five dimensions questionnaire. The time horizon was 18 months. Results: With a willingness-to-pay threshold of (sic)10, 000 ((sic)8568) the probability of cost-effectiveness oscillated from 83% (societal perspective) to 89% (health perspective). If the threshold was increased to (sic)30, 000 ((sic)25, 704), the probability of being considered cost-effective was 94% (societal perspective) and 96%, respectively (health perspective). The sensitivity analysis confirmed these results. Conclusions: Compared with usual care, an intervention based on personal predictors of risk of depression implemented by GPs is a cost-effective strategy to prevent depression. This type of personalized intervention in primary care should be further developed and evaluated
Land and Peace in Colombia: FFP Methodology for Field Data Collection and Data Handling
Effective land administration is an essential step on the road to peace in Colombia. The Colombian government plans to have a complete nation-wide land tenure coverage within seven years. The traditional approach to land administration in Colombia is not up to this policy challenge: the pace is too slow, the costs too high, the procedures too complex. Fast and effective land administration is essential for the implementation of the Reforma Rural Integral of the Peace Agreements, and to maintain public confidence in the peace process. The Netherlands contributes to the implementation of the peace agreements by focussing on fast and effective land administration, together with the Colombian institutions. This paper presents a methodology for a project where fit-for-purpose Land Administration is tested at scale â after a successful field test. The test will be done in two pilot areas. These are participatory and integrated pilots, leading to cadastral maps and, more important, to land titles whenever the legal framework and institutional cooperation allows for land regularization and adjudication
Wavelength-switchable L-band fiber laser assisted by random reflectors
A wavelength-switchable L-band erbium-doped fiber laser (EDFL) assisted by an artificially controlled backscattering (ACB) fiber reflector is here presented. This random reflector was inscribed by femtosecond (fs) laser direct writing on the axial axis of a multimode fiber with 50Â ÎŒm core and 125Â ÎŒm cladding with a length of 17Â mm. This microstructure was placed inside a surgical syringe to be positioned in the center of a high-precision rotation mount to accurately control its angle of rotation. Only by rotating this mount, three different output spectra were obtained: a single wavelength lasing centered at 1574.75Â nm, a dual wavelength lasing centered at 1574.75Â nm and 1575.75Â nm, and a single wavelength lasing centered at 1575.5Â nm. All of them showed an optical signal-to-noise ratio (OSNR) of around 60Â dB when pumped at 300Â mW
Heterogeneity of melanoma cell responses to sleep apnea-derived plasma exosomes and to intermittent hypoxia
Obstructive sleep apnea (OSA) is associated with increased cutaneous melanoma incidence and adverse outcomes. Exosomes are secreted by most cells, and play a role in OSA-associated tumor progression and metastasis. We aimed to study the effects of plasma exosomes from OSA patients before and after adherent treatment with continuous positive airway pressure (CPAP) on melanoma cells lines, and also to identify exosomal miRNAs from melanoma cells exposed to intermittent hypoxia (IH) or normoxia. Plasma-derived exosomes were isolated from moderate-to-severe OSA patients before (V1) and after (V2) adherent CPAP treatment for one year. Exosomes were co-incubated with three3 different melanoma cell lines (CRL 1424; CRL 1619; CRL 1675) that are characterized by genotypes involving different mutations in BRAF, STK11, CDKN2A, and PTEN genes to assess the effect of exosomes on cell proliferation and migration, as well as on pAMK activity in the presence or absence of a chemical activator. Subsequently, CRL-1424 and CRL-1675 cells were exposed to intermittent hypoxia (IH) and normoxia, and exosomal miRNAs were identified followed by GO and KEG pathways and gene networks. The exosomes from these IH-exposed melanoma cells were also administered to THP1 macrophages to examine changes in M1 and M2 polarity markers. Plasma exosomes from V1 increased CRL-1424 melanoma cell proliferation and migration compared to V2, but not the other two cell lines. Exposure to CRL-1424 exosomes reduced pAMPK/tAMPK in V1 compared to V2, and treatment with AMPK activator reversed the effects. Unique exosomal miRNAs profiles were identified for CRL-1424 and CRL-1675 in IH compared to normoxia, with six miRNAs being regulated and several KEGG pathways were identified. Two M1 markers (CXCL10 and IL6) were significantly increased in monocytes when treated with exosomes from IH-exposed CRL-1424 and CRL-1625 cells. Our findings suggest that exosomes from untreated OSA patients increase CRL-1424 melanoma malignant properties, an effect that is not observed in two other melanoma cell lines. Exosomal cargo from CRL-1424 cells showed a unique miRNA signature compared to CRL-1675 cells after IH exposures, suggesting that melanoma cells are differentially susceptible to IH, even if they retain similar effects on immune cell polarity. It is postulated that mutations in STK-11 gene encoding for the serine/threonine kinase family that acts as a tumor suppressor may underlie susceptibility to IH-induced metabolic dysfunction, as illustrated by CRL-1424 cells. © 2021 by the authors. 18F-FDG metabolism in a rat model of chronic infarction: a 17-sector semiquantitative analysis
Strategies to establish the functional benefit of cell therapy in cardiac regeneration and the potential mechanism are needed. Aims: Development of a semi-quantitative method for non invasive assessment of cardiac viability and function in a rat model of myocardial infarction (MI) based on the use of microPET. Animals, methods: Ten rats were subjected to myocardial imaging 2, 7, 14, 30, 60 and 90 days after left coronary artery ligation. Intravenous 18F-fluoro-2-deoxy-2-D-glucose (18F-FDG) was administered and regional 18F activity concentrations per unit area were measured in 17 regions of interest (ROIs) drawn on cardiac polar maps. By comparing the differences in 18F uptake between baseline and each of the follow up time points, parametric polar maps of statistical significance (PPMSS) were calculated. Left ventricular ejection fraction (LVEF) was blindly assessed echocardiographically. All animals were sacrificed for histopathological analysis after 90 days. Results: The diagnostic quality of 18F-FDG microPET images was excellent. PPMSS demonstrated a statistically significant decrease in 18F concentrations as early as 48 hours after MI in 4 of the 17 ROIs (segments 7, 13, 16 and 17; p <0.05) that persisted throughout the study. Semi-quantitative analysis of 18F-FDG uptake correlated with echocardiographic decrease in LVEF (p <0.001). Conclusion: The use of PPMSS based on 18F-FDG-microPET provides valuable semi-quantitative information of heart glucose metabolism allowing for non-invasive follow up thus representing a useful strategy for assessment of novel therapies in cardiac regeneration
New MRI, 18F-DOPA and 11C-(+)-alpha-dihydrotetrabenazine templates for Macaca fascicularis neuroimaging: advantages to improve PET quantification
Normalization of neuroimaging studies to a stereotaxic space allows the utilization of standard volumes of
interest (VOIs) and voxel-based analysis (SPM). Such spatial normalization of PET and MRI studies requires a
high quality template image. The aim of this study was to create new MRI and PET templates of 18F-DOPA
and 11C-(+)-α-dihydrotetrabenazine (11C-DTBZ) of the Macaca fascicularis brain, an important animal
model of Parkinson's disease. MRI template was constructed as a smoothed average of the scans of 15
healthy animals, previously transformed into the space of one representative MRI. In order to create the PET
templates, 18F-DOPA and 11C-DTBZ PET of the same subjects were acquired in a dedicated small animal PET
scanner and transformed to the created MRI template space. To validate these templates for PET
quantification, parametric values obtained with a standard VOI-map applied after spatial normalization to
each template were statistically compared to results computed using individual VOIs drawn for each animal.
The high correlation between both procedures validated the utilization of all the templates, improving the
reproducibility of PET analysis. To prove the utility of the templates for voxel-based quantification, dopamine
striatal depletion in a representative monkey treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine
(MPTP) was assessed by SPM analysis of 11C-DTBZ PET. A symmetric reduction in striatal 11C-DTBZ uptake
was detected in accordance with the induced lesion. In conclusion, templates of M. fascicularis brain have
been constructed and validated for reproducible and automated PET quantification. All templates are
electronically available via the internet
Effectiveness of an mHealth intervention combining a smartphone app and smart band on body composition in an overweight and obese population: Randomized controlled trial (EVIDENT 3 study)
Background: Mobile health (mHealth) is currently among the supporting elements that may contribute to an improvement in health markers by helping people adopt healthier lifestyles. mHealth interventions have been widely reported to achieve greater weight loss than other approaches, but their effect on body composition remains unclear.
Objective: This study aimed to assess the short-term (3 months) effectiveness of a mobile app and a smart band for losing weight and changing body composition in sedentary Spanish adults who are overweight or obese.
Methods: A randomized controlled, multicenter clinical trial was conducted involving the participation of 440 subjects from primary care centers, with 231 subjects in the intervention group (IG; counselling with smartphone app and smart band) and 209 in the control group (CG; counselling only). Both groups were counselled about healthy diet and physical activity. For the 3-month intervention period, the IG was trained to use a smartphone app that involved self-monitoring and tailored feedback, as well as a smart band that recorded daily physical activity (Mi Band 2, Xiaomi). Body composition was measured using the InBody 230 bioimpedance device (InBody Co., Ltd), and physical activity was measured using the International Physical Activity Questionnaire.
Results: The mHealth intervention produced a greater loss of body weight (â1.97 kg, 95% CI â2.39 to â1.54) relative to standard counselling at 3 months (â1.13 kg, 95% CI â1.56 to â0.69). Comparing groups, the IG achieved a weight loss of 0.84 kg more than the CG at 3 months. The IG showed a decrease in body fat mass (BFM; â1.84 kg, 95% CI â2.48 to â1.20), percentage of body fat (PBF; â1.22%, 95% CI â1.82% to 0.62%), and BMI (â0.77 kg/m2, 95% CI â0.96 to 0.57). No significant changes were observed in any of these parameters in men; among women, there was a significant decrease in BMI in the IG compared with the CG. When subjects were grouped according to baseline BMI, the overweight group experienced a change in BFM of â1.18 kg (95% CI â2.30 to â0.06) and BMI of â0.47 kg/m2 (95% CI â0.80 to â0.13), whereas the obese group only experienced a change in BMI of â0.53 kg/m2 (95% CI â0.86 to â0.19). When the data were analyzed according to physical activity, the moderate-vigorous physical activity group showed significant changes in BFM of â1.03 kg (95% CI â1.74 to â0.33), PBF of â0.76% (95% CI â1.32% to â0.20%), and BMI of â0.5 kg/m2 (95% CI â0.83 to â0.19).
Conclusions: The results from this multicenter, randomized controlled clinical trial study show that compared with standard counselling alone, adding a self-reported app and a smart band obtained beneficial results in terms of weight loss and a reduction in BFM and PBF in female subjects with a BMI less than 30 kg/m2 and a moderate-vigorous physical activity level. Nevertheless, further studies are needed to ensure that this profile benefits more than others from this intervention and to investigate modifications of this intervention to achieve a global effect
