    A Proteomic Study of the Bioactivity of Annona muricata Leaf Extracts in HT-1080 Fibrosarcoma Cells.

    Graviola (Annona muricata) is a tropical plant with many traditional ethnobotanic uses and pharmacologic applications. A metabolomic study of both aqueous and DMSO extracts from Annona muricata leaves recently allowed us to identify dozens of bioactive compounds. In the present study, we use a proteomic approach to detect altered patterns in proteins on both conditioned media and extracts of HT-1080 fibrosarcoma cells under treatment conditions, revealing new potential bioactivities of Annona muricata extracts. Our results reveal the complete sets of deregulated proteins after treatment with aqueous and DMSO extracts from Annona muricata leaves. Functional enrichment analysis of proteomic data suggests deregulation of cell cycle and iron metabolism, which are experimentally validated in vitro. Additional experimental data reveal that DMSO extracts protect HT-1080 fibrosarcoma cells and HMEC-1 endothelial cells from ferroptosis.     Study of humoral and cellular immunity in vaccinated with mRNA-1273

    This work was supported by "Investigacion y Desarrollo (I + D) del Sistema Andaluz de Salud (SAS)" and Instituto de Salud Carlos III (Proyecto FIS PI21/01708).The new vaccines against SARS-CoV-2 have raised a lot of expectations about their ability to induce immunity and the duration of this. This is the case of mRNA vaccines such as Moderna’s mRNA-1273. Therefore, it is necessary to study the humoral and cellular immunity generated by these vaccines. Our objectives are determining what is the normal response of antibody production, and what is the level of protective antibodies and monitoring patients in case of subsequent infection with COVID-19. We present the first results of a longitudinal study of the humoral response in 601 health workers vaccinated with Moderna. The results show a humoral immunity at 90 days after the second dose of 100%, with a strong decrease between the levels of circulating anti-S IgG antibodies between days 30 and 90 postvaccination. Observing a steeper decline in those who had higher titles at the beginning.     This is a communication in the format of a posterThe potential of marine organisms to yield bioactive molecules is vast and largely unexplored. Significantly, certain bioactive compounds derived from the marine environment have already received approval as anticancer drugs1. Among the multitude of bioactive compounds found in marine organisms, anthraquinones represent a notable class of molecules, with over 200 structurally related compounds having been isolated from diverse species of marine fungi2. Danthron (1,8-dihydroxy-9,10-anthraquinone) serves as an exemplary member of anthraquinones, possessing anti-tumoral and anti-angiogenic properties that have yet to be fully elucidated.     Antioxidant potential of an anthraquinone metabolite isolated from a marine fungus.

    Marine ecosystem is a fruitful source of many pharmacologically active. In particular, a number of marine anthraquinones and their derivatives, have been studied for their potential as anticancer and antimetastatic agents, although little is known about their antiangiogenic activities. In addition, there is some controversy regarding the poor antioxidant effect of these marine secondary metabolites. In this study, we describe for the first time the antiangiogenic activity of an anthraquinone, isolated from a marine fungus, which was evidenced in vitro by the inhibition of the activated endothelial cells survival, proliferation, and differentiation into tubelike structures. Furthermore, we have demonstrated its protective effect against reactive oxygen species (ROS), especially in presence of hydrogen peroxide, a potent prooxidant agent. These results were evidenced by a reduced ROS production and an increased redox capacity of cells after treatment with this anthraquinone, evidenced through the staining with DCFHDA and the sulfhydryl groups determination, respectively. The observation that this compound appears to present a more potent effect in tumor than in endothelial cells, suggests a putative selectivity. All these results highlight the need for further study of this molecule as a potential therapeutic agent for cancer and other angiogenesisdependent diseases.Universidad de Málaga.     Finite element modeling of temperature fields on the cutting edge in the dry high-speed turning of aisi 1045 steel

    High-speed turning is an advanced and emerging machining technique that, in contrast to the conventional machining, allows the manufacture of the workpiece with high accuracy, efficiency and quality, with lower production costs and with a considerable reduction in the machining times. The cutting tools used for the conventional machining cannot be employed for high-speed machining due to a high temperature induced in machining and a lower tool life. Therefore, it is necessary to study the influence of high cutting speeds on the temperature distribution in different typologies of cutting tools, with the aim of evaluating their behavior. In this paper, a finite element method modeling approach with arbitrary Lagrangian-Eulerian fully coupled thermal-stress analysis is employed. The research presents the results of different cutting tools (two coated carbide tools and uncoated cermet) effects on average surface temperature fields on the cutting edge in the dry high-speed turning of AISI 1045 steel. The numerical experiments were designed based on different cutting tools like input parameters and different temperature field zones like dependent variables in the dry high-speed turning of AISI 1045 steel. The results indicate that the dry high-speed turning of AISI 1045 steel does not influence significantly the temperature field zones when P10, P15 or P25 inserts are used. Therefore, the use of a dry high-speed turning method, which reduces the amount of lubricant and increases productivity, may represent an alternative to turning to the extent here described.Postprint (published version

    Danthron, an Anthraquinone Isolated from a Marine Fungus, Is a New Inhibitor of Angiogenesis Exhibiting Interesting Antitumor and Antioxidant Properties

    The role played by a sustained angiogenesis in cancer and other diseases stimulates the interest in the search for new antiangiogenic drugs. In this manuscript, we provide evidence that 1,8- dihydroxy-9,10-anthraquinone (danthron), isolated from the fermentation broth of the marine fungus Chromolaenicola sp. (HL-114-33-R04), is a new inhibitor of angiogenesis. The results obtained with the in vivo CAM assay indicate that danthron is a potent antiangiogenic compound. In vitro studies with human umbilical endothelial cells (HUVEC) reveal that this anthraquinone inhibits certain key functions of activated endothelial cells, including proliferation, proteolytic and invasive capabilities and tube formation. In vitro studies with human breast carcinoma MDA-MB231 and fibrosarcoma HT1080 cell lines suggest a moderate antitumor and antimetastatic activity of this compound. Antioxidant properties of danthron are evidenced by the observation that it reduces the intracellular reactive oxygen species production and increases the amount of intracellular sulfhydryl groups in endothelial and tumor cells. These results support a putative role of danthron as a new antiangiogenic drug with potential application in the treatment and angioprevention of cancer and other angiogenesis-dependent diseases.Partial funding for open access charge: Universidad de Málag

    Antiangiogenic potential of an olive oil extract: insights from a proteomic study

    Extra virgin olive oil (EVOO), a staple of the Mediterranean diet, is rich in phenolic compounds recognized for their potent bioactive effects, including anticancer and anti-inflammatory properties. However, its effects on vascular health remain relatively unexplored. In this study, we examined the impact of a “picual” EVOO extract from Jaén, Spain, on endothelial cells. Proteomic analysis revealed the modulation of angiogenesis-related processes. In subsequent in vitro experiments, the EVOO extract inhibited endothelial cell migration, adhesion, invasion, ECM degradation, and tube formation while inducing apoptosis. These results provide robust evidence of the extract’s antiangiogenic potential. Our findings highlight the potential of EVOO extracts in mitigating angiogenesis-related pathologies, such as cancer, macular degeneration, and diabetic retinopathy.Funding for open access charge: Universidad de Málaga / CBU

    En la presente investigación se muestra mediante el uso del conjunto de proyecciones climáticas de reducción de escala estadística pertenecientes al NEX-GDDP, los cambios a largo plazo en precipitación y temperatura que ocurrirían en América Central y el Caribe a finales del siglo XXI, esto a través del cálculo de las diferencias de los ensambles de los multi-modelos de los dos escenarios correspondientes a las rutas de concentración representativas (RCP 4.5 y RCP 8.5) con un período de 2070-2099 menos el escenario histórico que abarca de 1970-1999. Vislumbrando como un acelerado aumento en las concentraciones de gases de efecto invernadero debido a la dependencia del uso excesivo de combustibles fósiles, traería consigo cambios en esta región, tales como condiciones más secas en países como Guatemala, Belice, Honduras, El Salvador, Nicaragua y Costa Rica, y de manera contraria condiciones más húmedas en la costa Pacífica de países como Colombia, Ecuador y Perú.The long-term changes in precipitation and temperature that will occur in Central America and the Caribbean at the end of the 21st century are shown using the NEX-GDDP statistical downscaled projections. We used this high resolution dataset to compute the differences between the multi-model ensembles of the two prospective scenarios corresponding to the representative concentration pathways (RCP 4.5 and RCP 8.5) with a period of 2070-2099 and the retrospective baseline scenario covering 1970-1999.     Malnutrition is considered one of the main components to achieve an optimal state of health and ensure a good quality of life. Nutritional conditions are increasingly common in the world. FAO in 2017, states the information on the prevalence of malnutrition, also provides global figures for several regions of the world, approximately one in five people in the developing world have chronic undernutrition, 192 million children suffer from protein-energy malnutrition and More than 2,000 million have micronutrient deficiencies. This study was carried out with the objective of disseminating a topic to be consulted by health professionals and students. Our Ministry of Public Health dedicates important promotion and dissemination actions to reduce the effects of malnutrition on human beings.La malnutrición se considera uno de los principales componentes para alcanzar un estado de salud óptimo y garantizar una buena calidad de vida. Las afecciones nutricionales son cada vez más frecuentes en el mundo. La FAO en el año 2017, enuncia la información sobre la prevalencia de la malnutrición, además aporta cifras globales para varias regiones del mundo, aproximadamente una de cada cinco personas del mundo en desarrollo presentan subnutrición crónica, 192 millones de niños sufren de malnutrición proteinoenergética y más de 2.000 millones tienen carencias de micronutrientes.
