    Effect of Ionizing Radiation on Human Myeloperoxidase: Reaction With Hydrated Electrons

    Financiado para publicación en acceso aberto: Universidade da Coruña/CISUG[Abstract] Myeloperoxidase (MPO) is a myeloid-lineage restricted enzyme largely expressed in the azurophilic granules of neutrophils. It catalyses the formation of reactive oxygen species, mainly hypochlorous acid, contributing to anti-pathogenic defense. Disorders in the production or regulation of MPO may lead to a variety of health conditions, mainly of inflammatory origin, including autoimmune inflammation. We have studied the effect of ionizing radiation on the activity of MPO, as measured by the capacity retained by the enzyme to produce hypochlorous acid as reactive oxygen species after exposure to successive doses of solvated electrons, the strongest possible one-e− reducing agent in water. Chlorination activity was still present after a very high irradiation dose, indicating that radiation damage does not take place at the active site, hindered in the core of MPO structure. Decay kinetics show a dependence on the wavelength, supporting that the process must occur at peripheral functional groups situated on external and readily accessible locations of the enzyme.     Chemical and mechanical stability of air annealed cathodic arc evaporated CrAlON coatings

    This study reports the synthesis and characterization of ternary Cr-Al-O and quaternary Cr-Al-O-N coatings deposited by cathodic arc physical vapour deposition, for various nitrogen and oxygen mass flow ratios during the growth process. The composition, microstructure, indentation hardness and modulus of the films have been characterized by scanning electron microscopy, electron probe micro-analysis, X-ray diffraction, and nanoindentation techniques. The evolution of the microstructure and mechanical properties of the coatings after ambient air annealing from 800 °C up to 1100 °C have been investigated. As the oxygen to nitrogen mass flow increases, the as-deposited coatings exhibit lower hardness, higher roughness, lower crystallinity and a more marked columnar structure. At oxygen to nitrogen mass flow ratios bigger than 10/90, the coatings exhibit a stoichiometry of the type (CrAl)2+εO3−ε. Only the coatings with an oxygen to nitrogen mass flow ratio smaller than 10/90 retained nitrogen in their compositions. In all cases, the coatings developed a cubic fcc lattice structure. After annealing at 1100 °C the resulting microstructure showed a clear dependency upon the initial composition of the films. The evolution of the microstructure during the high temperature tests, as well as the analysis of the nanoindentation hardness, composition and thickness also provided valuable information about the combined effects of the thermal stability and the oxidation of the deposited coatings

    Molecular and in vivo studies of a glutamate-class prolyl-endopeptidase for coeliac disease therapy

    The digestion of gluten generates toxic peptides, among which a highly immunogenic proline-rich 33-mer from wheat α-gliadin, that trigger coeliac disease. Neprosin from the pitcher plant is a reported prolyl endopeptidase. Here, we produce recombinant neprosin and its mutants, and find that full-length neprosin is a zymogen, which is self-activated at gastric pH by the release of an all-β pro-domain via a pH-switch mechanism featuring a lysine plug. The catalytic domain is an atypical 7+8-stranded β-sandwich with an extended active-site cleft containing an unprecedented pair of catalytic glutamates. Neprosin efficiently degrades both gliadin and the 33-mer in vitro under gastric conditions and is reversibly inactivated at pH > 5. Moreover, co-administration of gliadin and the neprosin zymogen at the ratio 500:1 reduces the abundance of the 33-mer in the small intestine of mice by up to 90%.     Root Structure and Growth in Diverse Soils

    Unlike most biofuel species, Jatropha curcas has promise for use in marginal lands, but it may serve an additional role by stabilizing soils. We evaluated the growth and structural responsiveness of young J. curcas plants to diverse soil conditions. Soils included a sand, a sandy-loam, and a clay-loam from eastern Mexico. Growth and structural parameters were analyzed for shoots and roots, although the focus was the plasticity of the primary root system architecture (the taproot and four lateral roots). The sandy soil reduced the growth of both shoot and root systems significantly more than sandy-loam or clay-loam soils; there was particularly high plasticity in root and shoot thickness, as well as shoot length. However, the architecture of the primary root system did not vary with soil type; the departure of the primary root system from an index of perfect symmetry was 14 ± 5% (mean ± standard deviation). Although J. curcas developed more extensively in the sandy-loam and clay-loam soils than in sandy soil, it maintained a consistent root to shoot ratio and root system architecture across all types of soil. This strong genetic determination would make the species useful for soil stabilization purposes, even while being cultivated primarily for seed oil

    New Palaeolithic sites from the Depresión de Monforte de Lemos (Lugo, Galicia, Spain)

    Este artículo presenta el descubrimiento de nuevos yacimientos paleolíticos en la Depresión de Monforte de Lemos (Noroeste de la Península Ibérica). Esta zona fue rellenada con depósitos cuaternarios que han proporcionado más de 30 yacimientos al aire libre. Como consecuencia del descubrimiento casual de artefactos paleolíticos, se ha desarrollado un proyecto de intervención arqueológica sistemática, que ha demostrado el potencial de esta área. Muchos de los artefactos líticos han sido hallados en superficie, pero además contamos con dos yacimientos que tienen materiales en contexto estratigráfico (O Regueiral y Áspera). En la Depresión de Monforte hemos atestiguado poblamiento humano a lo largo del Pleistoceno medio y superior, con implementos líticos clasificados como Modo 2, Modo 3 y Modo 4. Los yacimientos más importantes son As Lamas, Chao de Fabeiro (ambos con industria lítica de Modo 2, que incluye bifaces y hendedores), O Regueiral (Modo 3) y Valverde (Modo 4). Por consiguiente, la Depresión de Monforte reúne las condiciones apropiadas para el desarrollo de un estudio diacrónico sobre la evolución del comportamiento técnico durante el Paleolítico, y su relación con la evolución del medioambiente a lo largo del PleistocenoThis paper reports the discovery of new Paleolithic sites in the Depresión de Monforte de Lemos (Monforte Basin, NW of the Iberian Peninsula). This zone was filled with Quaternary deposits that have yielded 30 open air sites. Due to the casual discovery of Paleolithic artifacts a systematic archaeological work was carried out, assessing the archaeological potential of this area. Most of these lithic artifacts were found on surface, but also we have located two sites with archaeological materials in stratigraphic context (O Regueiral and Áspera). In the basin of Monforte we have recognized a human settlement along the Middle and Upper Pleistocene, with lithic implements classified as Mode 2, Mode 3 and Mode 4. The most important sites are As Lamas, Chao de Fabeiro (both with Mode 2 lithic industry, that includes handaxes and cleavers), O Regueiral (Mode 3), and Valverde (Mode 4).     A GIS-supported Multidisciplinary Database for the Management of UNESCO Global Geoparks: the Courel Mountains Geopark (Spain)

    [Abstract] The management of a UNESCO Global Geopark (UGGp) requires a vast wealth of miscellaneous scientific knowledge that can be successfully organised using a Geographical Information System (GIS). This paper presents a pragmatic GIS database to assist in the suitable governance of the Courel Mountains UGGp (2017) in Northwest Spain. The database is structured in 66 coverages compiled from public sources and previous works or produced through traditional mapping (combining fieldwork and photointerpretation) and GIS tools. The acquired data was later homogenised and validated by a multidisciplinary team and archived in independent coverages. Forty thematic maps illustrate the broad range of cartographic information included in the GIS database. Among them, 25 basic maps provide an overview of the UGGp and 15 new maps focus on crosscutting and technical issues. All maps illustrate the huge potential of GIS to create new resources combining coverages and adapting the legend according to their purpose and audience. The database facilitates the suitable publishing of consistent outputs (e.g., brochures, books, panels, webpages, web serves), as well as the elaboration of technical data to assist the park management. The database furnishes information on the design of education actions, touristic routes, activities and Geopark facilities. The GIS database is also a supportive tool for scientific research and provides the necessary knowledge to conduct geoconservation actions based on land use, geological hazards and the occurrence of natural and cultural heritages. Altogether, the GIS database constitutes a powerful instrument for policy-making, facilitating the identification and evaluation of alternative strategy plans.This work was developed in the framework of the Scientific Program of the Courel Mountains UGGp with the cooperation of tourism agents (A.M.     Lack of replication of interactions between polymorphisms in rheumatoid arthritis susceptibility: case–control study

    [Abstract] INTRODUCTION: Approximately 100 loci have been definitively associated with rheumatoid arthritis (RA) susceptibility. However, they explain only a fraction of RA heritability. Interactions between polymorphisms could explain part of the remaining heritability. Multiple interactions have been reported, but only the shared epitope (SE) × protein tyrosine phosphatase nonreceptor type 22 (PTPN22) interaction has been replicated convincingly. Two recent studies deserve attention because of their quality, including their replication in a second sample collection. In one of them, researchers identified interactions between PTPN22 and seven single-nucleotide polymorphisms (SNPs). The other showed interactions between the SE and the null genotype of glutathione S-transferase Mu 1 (GSTM1) in the anti-cyclic citrullinated peptide-positive (anti-CCP+) patients. In the present study, we aimed to replicate association with RA susceptibility of interactions described in these two high-quality studies. METHODS: A total of 1,744 patients with RA and 1,650 healthy controls of Spanish ancestry were studied. Polymorphisms were genotyped by single-base extension. SE genotypes of 736 patients were available from previous studies. Interaction analysis was done using multiple methods, including those originally reported and the most powerful methods described. RESULTS: Genotypes of one of the SNPs (rs4695888) failed quality control tests. The call rate for the other eight polymorphisms was 99.9%. The frequencies of the polymorphisms were similar in RA patients and controls, except for PTPN22 SNP. None of the interactions between PTPN22 SNPs and the six SNPs that met quality control tests was replicated as a significant interaction term--the originally reported finding--or with any of the other methods. Nor was the interaction between GSTM1 and the SE replicated as a departure from additivity in anti-CCP+ patients or with any of the other methods. CONCLUSIONS: None of the interactions tested were replicated in spite of sufficient power and assessment with different assays.     Leaf water relations in Diospyros kaki during a mild water deficit exposure

    The resistance mechanisms (stress avoidance and stress tolerance) developed by persimmon plants (Diospyros kaki L. f. grafted on Diospyros lotus L.) in response to mild water stress and the sensitivity of continuously (on a whole-day basis) and discretely (at predawn and midday) measured indicators of the plant water status were investigated in 3-year old ‘Rojo Brillante’ persimmon plants. Control (T0) plants were drip irrigated in order to maintain soil water content at levels slightly above soil field capacity (102.3% of soil field capacity) and T1 plants were drip irrigated for 33 days in order to maintain the soil water content at around 80% of soil field capacity. The results indicated persimmon plants confront a mild water stress situation by gradually developing stomata control (stress avoidance mechanism) and exhibiting some xeromorphic characteristic such as high leaf relative apoplastic water content, which could contribute to the retention of water at low leaf water potentials. In addition, sap flow measurements made by the heat-pulse technique were seen to be the most suitable method for estimating persimmon water status, because it provided the highest signal intensity (actual value/reference value):noise (coefficient of variation) ratio in almost all intervals of time considered and provides continuous and automated registers of the persimmon water status in real time. © 2019 The AuthorsWe are grateful to the Arnau family from Explotaciones Ecológicas Harisa S.L. and Mr. J.
