    Public Perceptions of Wisconsin’s Pavements and Tradeoffs in Pavement Improvement

    Findings are reported from Phase II of a three-phase pooled-fund project in Wisconsin, Iowa, and Minnesota to determine perceptions of drivers regarding pavement of rural two-lane highways. Among the survey topics were drivers\u27 trust in the state department of transportation (DOT), pavement improvement trade-offs, and pavement evaluation. Results of the Wisconsin portion of the survey data are the focus of this study. The survey questionnaire was based in part on Phase I focus groups conducted to gauge beliefs about pavements as well as the language describing ruts, tining, and other pavement characteristics. Phase II entailed a statewide telephone survey of at least 400 randomly selected drivers in each of the three states. Although the focus here is on Wisconsin results, survey responses across the three states were very consistent. Included in the findings discussed are perceptions of pavement and the state DOT and pavement improvement options relating to construction, travel time, and delays. Results disclose key public perceptions of priorities with regard to spending limited funds. Also discussed are statistically significant relationships providing additional insights into public perceptions and pavement improvement on rural two-lane highways

    Differential SAGE analysis in Arabidopsis uncovers increased transcriptome complexity in response to low temperature

    Abstract Background Abiotic stress, including low temperature, limits the productivity and geographical distribution of plants, which has led to significant interest in understanding the complex processes that allow plants to adapt to such stresses. The wide range of physiological, biochemical and molecular changes that occur in plants exposed to low temperature require a robust global approach to studying the response. We have employed Serial Analysis of Gene Expression (SAGE) to uncover changes in the transcriptome of Arabidopsis thaliana over a time course of low temperature stress. Results Five SAGE libraries were generated from A. thaliana leaf tissue collected at time points ranging from 30 minutes to one week of low temperature treatment (4°C). Over 240,000 high quality SAGE tags, corresponding to 16,629 annotated genes, provided a comprehensive survey of changes in the transcriptome in response to low temperature, from perception of the stress to acquisition of freezing tolerance. Interpretation of these data was facilitated by representing the SAGE data by gene identifier, allowing more robust statistical analysis, cross-platform comparisons and the identification of genes sharing common expression profiles. Simultaneous statistical calculations across all five libraries identified 920 low temperature responsive genes, only 24% of which overlapped with previous global expression analysis performed using microarrays, although similar functional categories were affected. Clustering of the differentially regulated genes facilitated the identification of novel loci correlated with the development of freezing tolerance. Analysis of their promoter sequences revealed subsets of genes that were independent of CBF and ABA regulation and could provide a mechanism for elucidating complementary signalling pathways. The SAGE data emphasised the complexity of the plant response, with alternate pre-mRNA processing events increasing at low temperatures and antisense transcription being repressed. Conclusion Alternate transcript processing appears to play an important role in enhancing the plasticity of the stress induced transcriptome. Novel genes and cis-acting sequences have been identified as compelling targets to allow manipulation of the plant's ability to protect against low temperature stress. The analyses performed provide a contextual framework for the interpretation of quantitative sequence tag based transcriptome analysis which will prevail with the application of next generation sequencing technology.</p

    What children know about the source of their knowledge without reporting it as the source

    We argue that, amongst 3- to 5- year-olds, failure to report the source of knowledge recently acquired in answer to “How do you know…?” is due to a specific failure to make a causal inference, in line with source monitoring theory but not fuzzy trace theory. In three Experiments, children (N = 37; 30; 59) identified a hidden toy by seeing, feeling, or by being told, having had two modes of access on each trial, one informative (e.g. seeing a toy identified by colour) and the other uninformative (e.g. being told the toy’s colour by the Experimenter who had only felt it). Children who answered the know question wrongly nevertheless reported accurately who saw and who felt the toy, and what the well-informed player had said. They also realised when the Experimenter’s uninformative access implied their own knowledge was unreliable, suggesting precocious working understanding of knowledge sources

    Exercise Performance and Physiological Responses: The Potential Role of Redox Imbalance

    Increases in oxidative stress or decreases in antioxidant capacity, or redox imbalance, are known to alter physiological function and has been suggested to influence performance. To date, no study has sought to manipulate this balance in the same participants and observe the impact on physiological function and performance. Using a single-blind, placebo-controlled, and counterbalanced design, this study examined the effects of increasing free radicals, via hyperoxic exposure (FiO2 = 1.0), and/or increasing antioxidant capacity, through consuming an antioxidant cocktail (AOC; vitamin-C, vitamin-E, α-lipoic acid), on 5-kilometer (km) cycling time-trial performance, and the physiological and fatigue responses in healthy college-aged males. Hyperoxic exposure prior to the 5 km TT had no effect on performance, fatigue, or the physiological responses to exercise. The AOC significantly reduced average power output (222 ± 11 vs. 214 ± 12 W), increased 5 km time (516 ± 17 vs. 533 ± 18 sec), suppressed ventilation (VE; 116 ± 5 vs. 109 ± 13 L/min), despite similar oxygen consumption (VO2; 43.1 ± 0.8 vs. 44.9 ± 0.2 mL/kg per min), decreased VE/VO2 (35.9 ± 2.0 vs. 32.3 ± 1.5 L/min), reduced economy (VO2/W; 0.20 ± 0.01 vs. 0.22 ± 0.01), increased blood lactate (10 ± 0.7 vs. 11 ± 0.7 mmol), and perception of fatigue (RPE; 7.39 ± 0.4 vs. 7.60 ± 0.3) at the end of the TT, as compared to placebo (main effect, placebo vs. AOC, respectively). Our data demonstrate that prior to exercise, ingesting an AOC, but not exposure to hyperoxia, likely disrupts the delicate balance between pro- and antioxidant forces, which negatively impacts ventilation, blood lactate, economy, perception of fatigue, and performance (power output and 5 km time) in young healthy males. Thus, caution is warranted in athletes taking excess exogenous antioxidants

    Phosphorylation of Sli15 by Ipl1 is important for proper CPC localization and chromosome stability in <em>Saccharomyces cerevisiae</em>

    The chromosomal passenger complex (CPC) is a key regulator of eukaryotic cell division, consisting of the protein kinase Aurora B/Ipl1 in association with its activator (INCENP/Sli15) and two additional proteins (Survivin/Bir1 and Borealin/Nbl1). Here we have identified multiple sites of CPC autophosphorylation on yeast Sli15 that are located within its central microtubule-binding domain and examined the functional significance of their phosphorylation by Ipl1 through mutation of these sites, either to non-phosphorylatable alanine (sli15-20A) or to acidic residues to mimic constitutive phosphorylation (sli15-20D). Both mutant sli15 alleles confer chromosome instability, but this is mediated neither by changes in the capacity of Sli15 to activate Ipl1 kinase nor by decreased efficiency of chromosome biorientation, a key process in cell division that requires CPC function. Instead, we find that mimicking constitutive phosphorylation of Sli15 on the Ipl1 phosphorylation sites causes delocalization of the CPC in metaphase, whereas blocking phosphorylation of Sli15 on the Ipl1 sites drives excessive localization of Sli15 to the mitotic spindle in pre-anaphase cells. Consistent with these results, direct interaction of Sli15 with microtubules in vitro is greatly reduced either following phosphorylation by Ipl1 or when constitutive phosphorylation at the Ipl1-dependent phosphorylation sites is mimicked by aspartate or glutamate substitutions. Furthermore, we find that mimicking Ipl1 phosphorylation of Sli15 interferes with the 'tension checkpoint'--the CPC-dependent mechanism through which cells activate the spindle assembly checkpoint to delay anaphase in the absence of tension on kinetochore-microtubule attachments. Ipl1-dependent phosphorylation of Sli15 therefore inhibits its association with microtubules both in vivo and in vitro and may negatively regulate the tension checkpoint mechanism

    To investigate sporadic human cryptosporidiosis trends in the United Kingdom, we tested 3,030 Cryptosporidium spp.–positive fecal samples, submitted for routine typing in 2007–2008, for C. cuniculus. C. cuniculus prevalence was 1.2%; cases were mostly indigenous and occurred across all age groups. Most occurred during August–October and may be linked to exposure opportunities

The quasilinear elliptic operator is assumed to be variational and is such that 0 acts like an isolated eigenvalue with a corresponding eigenfunction which does not change sign. The driving force is further assumed to be in one-sided resonance around the eigenvalue 0, and a solvability condition of potential type is imposed. Variational methods are used to obtain existence. Our results significantly improve earlier results of the authors

    Students’ Acquisition of Agricultural and Entrepreneurship (Agripreneurship) Knowledge and Skills: Does Instructional Approach and their Sex Matter?

    Agricultural and Entrepreneurship education are interdisciplinary due to amalgamating the natural sciences and social sciences. These disciplines have gained the interest of those looking to equip youth with skills for self-reliance. Teachers employ various instructional approaches, including student-centric approaches such as project-based learning (PjBL) and teacher-centric methods, for example, the lecture method, to facilitate learning. Existing research, however, suggests that students’ learning can be influenced by other factors, for example, learning styles, socio-cultural norms, sex stereotypes, and the instructional approach(es) used. We examined the impact of using the lecture method (counterfactual group) versus PjBL (treatment group) approaches on student acquisition of agricultural knowledge in the context of poultry science and their intentions to become agripreneurs. A statistically significant disordinal interaction with a medium effect size was found at p \u3c .05 between groups and student sex for poultry science knowledge. The female students performed better under the PjBL, while the males did so under lecture-based instruction. We also found a statistically significant and positive (p \u3c .05) association between students’ sex and their intent to become agripreneurs for the treatment group. More female students than males in the treatment group indicated they were either likely or highly likely to become agripreneurs in the future. These findings imply that females in the treatment group benefited more from the intervention, PjBL, than their male peers. Additional research should be conducted to measure the long-term impact of using various teaching approaches on students’ learning of agriculture and entrepreneurship content depending on their sex

    Microheterogeneity of Type II cAMP-Dependent Protein Kinase in Various Mammalian Species and Tissues

    Excluding autophosphorylated species, at least six forms of the regulatory subunit of type II cAMP-dependent protein kinase (R(II)) from various mammalian tissues were identified by sodium dodecyl sulfate (SDS) gel electrophoresis of purified samples and of crude preparations photoaffinity labeled with 8-azido[32P] cAMP and by gel filtration. After autophosphorylation some heart R(II) forms termed type IIA (bovine, porcine, equine, and dog) shifted to a more slowly migrating band on SDS gels while others termed type IIB (rat, guinea pig, rabbit, and monkey) did not detectably shift. Both subclasses of R(II) exhibited variation in apparent M(r) on SDS gels. Bovine and porcine heart nonautophosphorylated R(II) had M(r) 56,000 and the autophosphorylated R(II) had M(r) 58,000, while dog and equine heart R(II) had M(r) 54,000 and 56,000 for these bands, respectively. Rat heart R(II) had M(r) 56,000 while rabbit and guinea pig heart R(II) had M(r) 52,000. More than one R(II) was found in different tissues of the same species. Rabbit skeletal muscle contained a M(r) 56,000 IIB form. Bovine lung contained almost equal amounts of a IIA form apparently identical to that of bovine heart and a M(r) 52,000 IIB form similar to that which predominated in bovine brain. Rat adipose tissue, brain, and monkey heart contained predominantly a M(r) 51,000 IIB form. The rat liver M(r) 56,000 IIB form chromatographed differently from all other R(II) tested by gel filtration. Several lines of evidence indicated that the various forms of R(II) were not derived from one another through proteolysis or other processes. Each of the type II forms rapidly incorporated 0.3-1.0 mol of 32P per mol of subunit when incubated with [γ-32P]ATP and C subunit. Four of the forms tested were similar in the cAMP concentration dependence for activation of their corresponding holoenzymes and inhibited C subunit about equally. Each exhibited two components of [3H]cAMP dissociation, indicating two intrachain cAMP-binding sites, and the dissociation rates for the respective sites, and the dissociation rates for the respective sites were similar
