    Why are biotic iron pools uniform across high- and low-iron pelagic ecosystems?

    Dissolved iron supply is pivotal in setting global phytoplankton productivity and pelagic ecosystem structure. However, most studies of the role of iron have focussed on carbon biogeochemistry within pelagic ecosystems, with less effort to quantify the iron biogeochemical cycle. Here we compare mixed-layer biotic iron inventories from a low-iron (~0.06nmol L-1) subantarctic (FeCycle study) and a seasonally high-iron (~0.6nmol L-1) subtropical (FeCycle II study) site. Both studies were quasi-Lagrangian, and had multi-day occupation, common sampling protocols, and indirect estimates of biotic iron (from a limited range of available published biovolume/carbon/iron quotas). Biotic iron pools were comparable (~100Β±30pmol L-1) for low- and high-iron waters, despite a tenfold difference in dissolved iron concentrations. Consistency in biotic iron inventories (~80Β±24pmol L-1, largely estimated using a limited range of available quotas) was also conspicuous for three Southern Ocean polar sites. Insights into the extent to which uniformity in biotic iron inventories was driven by the need to apply common iron quotas obtained from laboratory cultures were provided from FeCycle II. The observed twofold to threefold range of iron quotas during the evolution of FeCycle II subtropical bloom was much less than reported from laboratory monocultures. Furthermore, the iron recycling efficiency varied by fourfold during FeCycle II, increasing as stocks of new iron were depleted, suggesting that quotas and iron recycling efficiencies together set biotic iron pools. Hence, site-specific differences in iron recycling efficiencies (which provide 20-50% and 90% of total iron supply in high- and low-iron waters, respectively) help offset the differences in new iron inputs between low- and high-iron sites. Future parameterization of iron in biogeochemical models must focus on the drivers of biotic iron inventories, including the differing iron requirements of the resident biota, and the subsequent fate (retention/export/recycling) of the biotic iron

    Work and Welfare in the American States: Analyzing the Effects of the JOBS Program

    This research seeks to determine whether the Job Opportunities and Basic Skills GOBS) program (established under the 1988 Family Support Act) was successful in reducing the number of welfare recipients among U.S. states for the period 1984 to 1996. Within the context of two theoretical perspectives-developmental and rational choice-we assess the impact of JOBS on AFDC participation rates using a pooled time-series design. At best, JOBS had a minimal effect. We estimate that states with higher proportions of their AFDC populations enrolled in JOBS programs had only slightly lower rates of participation in AFDC. Other forces were far more influential in reducing welfare participation. In particular, states with higher per capita income, lower female unemployment rates, lower poverty rates, and higher wages for low-paying jobs had the lowest welfare recipiency The AFDC participation rates of neighboring states had a significant effect, as well.     MYC Cooperates with AKT in Prostate Tumorigenesis and Alters Sensitivity to mTOR Inhibitors

    MYC and phosphoinositide 3-kinase (PI3K)-pathway deregulation are common in human prostate cancer. Through examination of 194 human prostate tumors, we observed statistically significant co-occurrence of MYC amplification and PI3K-pathway alteration, raising the possibility that these two lesions cooperate in prostate cancer progression. To investigate this, we generated bigenic mice in which both activated human AKT1 and human MYC are expressed in the prostate (MPAKT/Hi-MYC model). In contrast to mice expressing AKT1 alone (MPAKT model) or MYC alone (Hi-MYC model), the bigenic phenotype demonstrates accelerated progression of mouse prostate intraepithelial neoplasia (mPIN) to microinvasive disease with disruption of basement membrane, significant stromal remodeling and infiltration of macrophages, B- and T-lymphocytes, similar to inflammation observed in human prostate tumors. In contrast to the reversibility of mPIN lesions in young MPAKT mice after treatment with mTOR inhibitors, Hi-MYC and bigenic MPAKT/Hi-MYC mice were resistant. Additionally, older MPAKT mice showed reduced sensitivity to mTOR inhibition, suggesting that additional genetic events may dampen mTOR dependence. Since increased MYC expression is an early feature of many human prostate cancers, these data have implications for treatment of human prostate cancers with PI3K-pathway alterations using mTOR inhibitors

    Research Exploring Physical Activity in Care Homes (REACH): study protocol for a randomised controlled trial

    Background: As life expectancy increases and the number of older people, particularly those aged 85 years and over, expands there is an increase in demand for long-term care. A large proportion of people in a care home setting spend most of their time sedentary, and this is one of the leading preventable causes of death. Encouraging residents to engage in more physical activity could deliver benefits in terms of physical and psychological health, and quality of life. This study is the final stage of a programme of research to develop and preliminarily test an evidence-based intervention designed to enhance opportunities for movement amongst care home residents, thereby increasing levels of physical activity. Methods/design: This is a cluster randomised feasibility trial, aiming to recruit at least 8–12 residents at each of 12 residential care homes across Yorkshire, UK. Care homes will be randomly allocated on a 1:1 basis to receive either the intervention alongside usual care, or to continue to provide usual care alone. Assessment will be undertaken with participating residents at baseline (prior to care home randomisation) and at 3, 6, and 9 months post-randomisation. Data relating to changes in physical activity, physical function, level of cognitive impairment, mood, perceived health and wellbeing, and quality of life will be collected. Data at the level of the home will also be collected and will include staff experience of care, and changes in the numbers and types of adverse events residents experience (for example, hospital admissions, falls). Details of National Health Service (NHS) usage will be collected to inform the economic analysis. An embedded process evaluation will obtain information to test out the theory of change underpinning the intervention and its acceptability to staff and residents. Discussion: This feasibility trial with embedded process evaluation and collection of health economic data will allow us to undertake detailed feasibility work to inform a future large-scale trial. It will provide valuable information to inform research procedures in this important but challenging area

    Comprehensive annotation of the Parastagonospora nodorum reference genome using next-generation genomics, transcriptomics and proteogenomics

    Parastagonospora nodorum, the causal agent of Septoria nodorum blotch (SNB), is an economically important pathogen of wheat (Triticum spp.), and a model for the study of necrotrophic pathology and genome evolution. The reference P. nodorum strain SN15 was the first Dothideomycete with a published genome sequence, and has been used as the basis for comparison within and between species. Here we present an updated reference genome assembly with corrections of SNP and indel errors in the underlying genome assembly from deep resequencing data as well as extensive manual annotation of gene models using transcriptomic and proteomic sources of evidence (https://github.com/robsyme/Parastagonospora_nodorum_SN15). The updated assembly and annotation includes 8,366 genes with modified protein sequence and 866 new genes. This study shows the benefits of using a wide variety of experimental methods allied to expert curation to generate a reliable set of gene models
