A microscopic Kapitza pendulum
Pyotr Kapitza studied in 1951 the unusual equilibrium features of a rigid pendulum when its point of suspension is under a high-frequency vertical vibration. A sufficiently fast vibration makes the top position stable, putting the pendulum in an inverted orientation that seemingly defies gravity. Kapitza’s analytical method, based on an asymptotic separation of fast and slow variables yielding a renormalized potential, has found application in many diverse areas. Here we study Kapitza’s pendulum going beyond its typical idealizations, by explicitly considering its finite stiffness and the dissipative interaction with the surrounding medium, and using similar theoretical methods as Kapitza. The pendulum is realized at the micrometre scale using a colloidal particle suspended in water and trapped by optical tweezers. Though the strong dissipation present at this scale prevents the inverted pendulum regime, new ones appear in which the equilibrium positions are displaced to the side, and with transitions between them determined either by the driving frequency or the friction coefficient. These new regimes could be exploited in applications aimed at particle separation at small scales
Stretching red blood cells with optical tweezers
In this work we report on experiments to measure the deformability of red blood cells (RBCs) when subject to stretching with optical tweezers. Individual red blood cells are trapped directly in a dual optical tweezers, and subject to a stretching protocol that is the same for all cells under test. Differences in the resulting extension are therefore indicative of differences in cell deformability. The targets for investigation are RBCs taken from patients with type 2 diabetes mellitus (T2DM) who exhibit diabetic retinopathy (DR), and from patients with birdshot chorioretinopathy (BCR). We find a statistically significant change in deformability for RBCs from DR patients compared to a control group, but no significant change for BCR patients. These results offer support to the importance of the role of RBC biomechanical properties in the progress of these conditions

Psychometric precision in phenotype definition is a useful step in molecular genetic investigation of psychiatric disorders.
Affective disorders are highly heritable, but few genetic risk variants have been consistently replicated in molecular genetic association studies. The common method of defining psychiatric phenotypes in molecular genetic research is either a summation of symptom scores or binary threshold score representing the risk of diagnosis. Psychometric latent variable methods can improve the precision of psychiatric phenotypes, especially when the data structure is not straightforward. Using data from the British 1946 birth cohort, we compared summary scores with psychometric modeling based on the General Health Questionnaire (GHQ-28) scale for affective symptoms in an association analysis of 27 candidate genes (249 single-nucleotide polymorphisms (SNPs)). The psychometric method utilized a bi-factor model that partitioned the phenotype variances into five orthogonal latent variable factors, in accordance with the multidimensional data structure of the GHQ-28 involving somatic, social, anxiety and depression domains. Results showed that, compared with the summation approach, the affective symptoms defined by the bi-factor psychometric model had a higher number of associated SNPs of larger effect sizes. These results suggest that psychometrically defined mental health phenotypes can reflect the dimensions of complex phenotypes better than summation scores, and therefore offer a useful approach in genetic association investigations.This work was supported by the Wellcome Trust [088869/Z/09/Z to M.R., P.B.J., D.G, and T. Sex-Specific Differences in Shoaling Affect Parasite Transmission in Guppies
Background: Individuals have to trade-off the costs and benefits of group membership during shoaling behaviour. Shoaling can increase the risk of parasite transmission, but this cost has rarely been quantified experimentally. Guppies (Poecilia reticulata) are a model system for behavioural studies, and they are commonly infected by gyrodactylid parasites, notorious fish pathogens that are directly transmitted between guppy hosts.
Methodology/Principal Findings:Parasite transmission in single sex shoals of male and female guppies were observed using an experimental infection of Gyrodactylus turnbulli. Parasite transmission was affected by sex-specific differences in host behaviour, and significantly more parasites were transmitted when fish had more frequent and more prolonged contact with each other. Females shoaled significantly more than males and had a four times higher risk to contract an infection.
Conclusions/Significance: Intersexual differences in host behaviours such as shoaling are driven by differences in natural and sexual selection experienced by both sexes. Here we show that the potential benefits of an increased shoaling tendency are traded off against increased risks of contracting an infectious parasite in a group-living species
Social pathways for Ebola Virus Disease in rural Sierra Leone, and some implications for containment
The current outbreak of Ebola Virus Disease in Upper West Africa is the largest ever recorded. Molecular evidence suggests spread has been almost exclusively through human-to-human contact. Social factors are thus clearly important to understand the epidemic and ways in which it might be stopped, but these factors have so far been little analyzed. The present paper focuses on Sierra Leone, and provides cross sectional data on the least understood part of the epidemic-the largely undocumented spread of Ebola in rural areas. Various forms of social networking in rural communities and their relevance for understanding pathways of transmission are described. Particular attention is paid to the relationship between marriage, funerals and land tenure. Funerals are known to be a high-risk factor for infection. It is suggested that more than a shift in awareness of risks will be needed to change local patterns of behavior, especially in regard to funerals, since these are central to the consolidation of community ties. A concluding discussion relates the information presented to plans for halting the disease. Local consultation and access are seen as major challenges to be addressed
Asymmetric mid-plane gas in ALMA images of HD 100546
In this paper we present new ALMA observations towards the proto-planet
hosting transitional disc of Herbig Ae/Be star HD 100546. This includes
resolved 1.3 mm continuum, CO and the first detection of CO in
this disc, which displays azimuthal asymmetry in regions spatially coincident
with structures previously identified in HST images related to spiral arms. The
lower limit on the mass of the dust disc is calculated to be
9.6x10M. A firm lower-limit on the total gas mass calculated
from optically thin, mid-plane tracing CO (2-1) emission is
0.018M assuming ISM abundances. These mass estimates provide an
estimate of gas-to-dust ratio in the disc of 19, the ratio will increase if
CO is relatively under-abundant in the disc compared to CO and H2.
Through deprojection and azimuthal averaging of the image plane we detect 1.3
mm continuum emission out to 290+/-10 au,CO to 390+/-10 au and CO
to 300+/-10au. We measure a radially increasing millimetre spectral index
between wavelengths of 867m and 1.3 mm, which shows that grain sizes
increase towards the star, with solid particles growing to cm scales in the
inner disc
X-ray Absorption and Reflection in Active Galactic Nuclei
X-ray spectroscopy offers an opportunity to study the complex mixture of
emitting and absorbing components in the circumnuclear regions of active
galactic nuclei, and to learn about the accretion process that fuels AGN and
the feedback of material to their host galaxies. We describe the spectral
signatures that may be studied and review the X-ray spectra and spectral
variability of active galaxies, concentrating on progress from recent Chandra,
XMM-Newton and Suzaku data for local type 1 AGN. We describe the evidence for
absorption covering a wide range of column densities, ionization and dynamics,
and discuss the growing evidence for partial-covering absorption from data at
energies > 10 keV. Such absorption can also explain the observed X-ray spectral
curvature and variability in AGN at lower energies and is likely an important
factor in shaping the observed properties of this class of source.
Consideration of self-consistent models for local AGN indicates that X-ray
spectra likely comprise a combination of absorption and reflection effects from
material originating within a few light days of the black hole as well as on
larger scales. It is likely that AGN X-ray spectra may be strongly affected by
the presence of disk-wind outflows that are expected in systems with high
accretion rates, and we describe models that attempt to predict the effects of
radiative transfer through such winds, and discuss the prospects for new data

Planet formation in intermediate-separation binary systems
ABSTRACT
We report the first characterization of the individual discs in the intermediate-separation binary systems KK Oph and HD 144668 at millimetre wavelengths. In both systems, the circumprimary and the circumsecondary discs are detected in the millimetre continuum emission, but not in 13CO nor C18O lines. Even though the disc structure is only marginally resolved, we find indications of large-scale asymmetries in the outer regions of the primary discs, most likely due to perturbation by the companion. The derived dust masses are firmly above debris disc level for all stars. The primaries have about three times more dust in their discs than the secondaries. In the case of HD 144668, the opacity spectral index of the primary and secondary differ by a large margin of 0.69, which may be a consequence of the secondary disc being more compact. Upper limits on the gas masses imply less than 0.1 Mjup in any of these discs, meaning that giant planets can no longer form in them. Considering that there have been no massive gas discs identified to date in intermediate-separation binaries (i.e. binaries at a few hundred au separation), this opens space for speculation whether their binarity causes the removal of gas, with tidal interaction truncating the discs and hence shortening the accretion time-scale. More systematic studies in this respect are sorely needed.</jats:p
Genome-wide DNA methylation analysis for diabetic nephropathy in type 1 diabetes mellitus
BACKGROUND: Diabetic nephropathy is a serious complication of diabetes mellitus and is associated with considerable morbidity and high mortality. There is increasing evidence to suggest that dysregulation of the epigenome is involved in diabetic nephropathy. We assessed whether epigenetic modification of DNA methylation is associated with diabetic nephropathy in a case-control study of 192 Irish patients with type 1 diabetes mellitus (T1D). Cases had T1D and nephropathy whereas controls had T1D but no evidence of renal disease. METHODS: We performed DNA methylation profiling in bisulphite converted DNA from cases and controls using the recently developed Illumina Infinium(R) HumanMethylation27 BeadChip, that enables the direct investigation of 27,578 individual cytosines at CpG loci throughout the genome, which are focused on the promoter regions of 14,495 genes. RESULTS: Singular Value Decomposition (SVD) analysis indicated that significant components of DNA methylation variation correlated with patient age, time to onset of diabetic nephropathy, and sex. Adjusting for confounding factors using multivariate Cox-regression analyses, and with a false discovery rate (FDR) of 0.05, we observed 19 CpG sites that demonstrated correlations with time to development of diabetic nephropathy. Of note, this included one CpG site located 18 bp upstream of the transcription start site of UNC13B, a gene in which the first intronic SNP rs13293564 has recently been reported to be associated with diabetic nephropathy. CONCLUSION: This high throughput platform was able to successfully interrogate the methylation state of individual cytosines and identified 19 prospective CpG sites associated with risk of diabetic nephropathy.
