    Seasonal variation of size-resolved aerosol fluxes in a Peri-urban deciduous broadleaved forest

    Eddy covariance measurements of aerosol fluxes were performed above an oak-hornbeam forest in the Po Plain (Northern Italy), from February to May and from September to December 2019. Measurements aimed at assessing the influence of forest phenology and leaf presence/absence on the seasonal evolution of size-segregated aerosol fluxes. The size-resolved aerosol concentration in the range 0.006-10 μm was sampled with a 14-stage impactor (ELPI+, Dekati, FI), and the filters exposed in May were subjected to chemical analysis. Over the whole sampling period, the forest removed from the atmosphere an average of 3.12 mg of aerosol m−2 d−1. The direction and the intensity of the aerosol fluxes were not constant through the year, as a strong seasonal and size-dependent variability emerged. In particular, leaf-presence drove a net deposition of the accumulation mode aerosol (100 nm< particle diameter Dp<1000 nm) and an emission of the Aitken (10 nm< Dp<100 nm) and coarse mode (Dp>1000 nm) aerosols. On the contrary, in absence of leaves all the sub-micrometer aerosol size-classes showed net daily upward fluxes, while coarse mode aerosol fluxes were prevalently downward. Monthly averages of deposition velocities of Aitken and accumulation mode aerosols correlated with the Leaf Area Index (LAI) seasonal trend, thus indicating an important role of the amount of the leaf surface area on the deposition and emission of these size-classes. Furthermore, an influence of the stomatic activity was suggested for the Aitken mode aerosol, since its deposition velocity followed the same diel course of the stomatal conductance to water. The analysis of the influence of meteorological parameters on aerosol deposition velocities highlighted that dynamic and convective turbulence (described by friction velocity, u* and Deardorff velocity, w*) enhanced the vertical aerosol exchanges, both upward and downward, while the approaching of condensing conditions reduced the flux intensities

    Cardiovascular risk profile and lifestyle habits in a cohort of Italian cardiologists. Results of the SOCRATES survey

    Objectives. To offer a snapshot of the personal health habits of Italian cardiologists, the Survey on Cardiac Risk Profile and Lifestyle Habits in a Cohort of Italian Cardiologists (SOCRATES) study was undertaken. Background. Cardiologists' cardiovascular profile and lifestyle habits are poorly known worldwide. Methods. A Web-based electronic self-reported survey, accessible through a dedicated website, was used for data entry, and data were transferred via the web to a central database. The survey was divided in 4 sections: baseline characteristics, medical illnesses and traditional cardiovascular risk factors, lifestyle habits and selected medication use. The e-mail databases of three national scientific societies were used to survey a large and representative sample of Italian cardiologists. Results. During the 3-month period of the survey, 1770 out of the 5240 cardiologists contacted (33.7%) completed and returned one or more sections of the questionnaire. More than 49% of the participants had 1 out of 5 classical risk factors (e.g. hypertension, hypercholesterolemia, active smoking, diabetes and previous vascular events). More than 28% of respondents had 2 to 5 risk factors and only 22.1% had none and therefore, according to age and sex, could be considered at low-intermediate risk. Despite the reported risk factors, more than 90% of cardiologists had a self-reported risk perception quantified as mild, such as low or intermediate. Furthermore, overweight/obesity, physical inactivity and stress at work or at home were commonly reported, as well as a limited use of cardiovascular drugs, such as statins or aspirin. Conclusions. The average cardiovascular profile of Italian cardiologist is unlikely to be considered ideal or even favorable according to recent statements and guidelines regarding cardiovascular risk. Thus, there is a large room for improvement and a need for education and intervention

    Stem cells isolated from human dental pulp and amniotic fluid improve skeletal muscle histopathology in mdx/SCID mice

    Introduction: Duchenne muscular dystrophy (DMD), caused by a lack of the functional structural protein dystrophin, leads to severe muscle degeneration where the patients are typically wheelchair-bound and die in their mid-twenties from cardiac or respiratory failure or both. The aim of this study was to investigate the potential of human dental pulp stem cells (hDPSCs) and human amniotic fluid stem cells (hAFSCs) to differentiate toward a skeletal myogenic lineage using several different protocols in order to determine the optimal conditions for achieving myogenic commitment and to subsequently evaluate their contribution in the improvement of the pathological features associated with dystrophic skeletal muscle when intramuscularly injected into mdx/SCID mice, an immune-compromised animal model of DMD. Methods: Human DPSCs and AFSCs were differentiated toward myogenic lineage in vitro through the direct co-culture with a myogenic cell line (C2C12 cells) and through a preliminary demethylation treatment with 5-Aza-2\u2032-deoxycytidine (5-Aza), respectively. The commitment and differentiation of both hDPSCs and hAFSCs were evaluated by immunofluorescence and Western blot analysis. Subsequently, hDPSCs and hAFSCs, preliminarily demethylated and pre-differentiated toward a myogenic lineage for 2 weeks, were injected into the dystrophic gastrocnemius muscles of mdx/SCID mice. After 1, 2, and 4 weeks, the gastrocnemius muscles were taken for immunofluorescence and histological analyses. Results: Both populations of cells engrafted within the host muscle of mdx/SCID mice and through a paracrine effect promoted angiogenesis and reduced fibrosis, which eventually led to an improvement of the histopathology of the dystrophic muscle. Conclusion: This study shows that hAFSCs and hDPSCs represent potential sources of stem cells for translational strategies to improve the histopathology and potentially alleviate the muscle weakness in patients with DMD

    Evaluation of site effects in the Aterno river valley (Central Italy) from aftershocks of the 2009 L'Aquila earthquake

    A temporary network of 33 seismic stations was deployed in the area struck by the 6th April 2009, Mw 6.3, L’Aquila earthquake (central Italy), with the aim to investigate the site amplification within the Aterno river Valley. The seismograms of 18 earthquakes recorded by 14 of the 33 stations were used to evaluate the average horizontal to vertical spectral ratio (HVSR) for each site and the standard horizontal spectral ratio (SSR) between a site and a reference station. The obtained results have been compared to the geological and geophysical information in order to explain the resonance frequencies and the amplification levels with respect to surface geology of the valley. The result indicate that there is no uniform pattern of amplification, due to the complex geologic setting, as the thickness and degree of cementation of the deposits is highly variable. As consequence, a large number of the local site response is observed, therefore it is very difficult to elaborate a unique model that can explain such a variability of the amplification.Published697-7154.1.     Evaluation of the local site effects in the upper and middle Aterno valley

    In the months following the April 6th, 2009, L’Aquila earthquake, several Italian and foreign research institutions installed dozens of seismic stations to monitor more than 100 localities with the aim of studying the local site effects in the epicentral area (upper and middle Aterno valley). The stations (accelerometers and velocimeters) have been deployed inside or very close to the inhabited areas. Among the investigated sites there are Onna, where almost the totality of the buildings collapsed, and the historic centre of L’Aquila, both towns suffering many casualties. The preliminary results for the examined sites show an extreme variability of ground motion and significant amplification for the most damaged localities.In press4.1.     Candidate biomarkers from the integration of methylation and gene expression in discordant autistic sibling pairs

    While the genetics of autism spectrum disorders (ASD) has been intensively studied, resulting in the identification of over 100 putative risk genes, the epigenetics of ASD has received less attention, and results have been inconsistent across studies. We aimed to investigate the contribution of DNA methylation (DNAm) to the risk of ASD and identify candidate biomarkers arising from the interaction of epigenetic mechanisms with genotype, gene expression, and cellular proportions. We performed DNAm differential analysis using whole blood samples from 75 discordant sibling pairs of the Italian Autism Network collection and estimated their cellular composition. We studied the correlation between DNAm and gene expression accounting for the potential effects of different genotypes on DNAm. We showed that the proportion of NK cells was significantly reduced in ASD siblings suggesting an imbalance in their immune system. We identified differentially methylated regions (DMRs) involved in neurogenesis and synaptic organization. Among candidate loci for ASD, we detected a DMR mapping to CLEC11A (neighboring SHANK1) where DNAm and gene expression were significantly and negatively correlated, independently from genotype effects. As reported in previous studies, we confirmed the involvement of immune functions in the pathophysiology of ASD. Notwithstanding the complexity of the disorder, suitable biomarkers such as CLEC11A and its neighbor SHANK1 can be discovered using integrative analyses even with peripheral tissues
