    Histological and ultrastructural feature and nitrite production of caprine preantral follicles in vitro cultured in the presence or absence of serum

    Avaliou-se o efeito da adição de diferentes tipos e concentrações de soro sobre o desenvolvimento e a sobrevivência de folículos ovarianos pré-antrais (FOPA) caprinos in vitro. Além disso, verificou-se a relação entre as concentrações de nitrito presentes no meio de cultivo e a viabilidade folicular. Cada par ovariano foi dividido em 29 fragmentos, sendo um destinado ao controle. Os fragmentos foram cultivados por um ou sete dias em meio essencial mínimo suplementado (MEM+) ou MEM+ com diferentes concentrações (10 ou 20%) de soro fetal bovino (SFB), soro de cabra em estro (SCE) ou soro de cabra em diestro (SCD). Na análise morfológica após sete dias, apenas o tratamento com 10% de SFB apresentou percentual de FOPA normais similar ao MEM+ (P>0,05). A análise ultra-estrutural dos folículos cultivados por sete dias com MEM+ ou MEM+ com 10% de SFB mostrou danos oocitários, porém células da granulosa normais. A análise do meio de cultivo revelou correlação positiva entre a viabilidade folicular e a produção de nitrito. A suplementação com soro não melhorou a viabilidade de FOPA e a concentração de nitrito no meio de cultivo funcionou como um indicador da viabilidade das células da granulosa de FOPA caprinos cultivados in vitro. ______________________________________________________________________________________________________________ ABSTRACTThe effect of the addition of different types and concentrations of sera on the viability and development of caprine preantal follicles (PAF) in vitro cultured was analyzed. In addition, it was evaluated the correlation between nitrite concentrations in culture medium and folicular viability. Each ovarian pair was divided in 29 fragments and one was used as control. The fragments were cultured for one or seven days in minimal essential medium (MEM+) or MEM+ with different concentrations of (10 or 20%) bovine fetal serum (BFS), estrous goat serum (EGS), or diestrous goat serum (DGS). After seven days, the morphological analysis showed that only the treatment with 10% BFS maintained the percentage of normal PAF similar to MEM+ (P>0.05). The ultrastructural analysis of follicles cultured for seven days in MEM+ or MEM+ with 10% BFS showed some oocyte damage, although the granulosa cells were normal. Analysis of culture medium revealed a positive correlation between follicular viability and nitrite production. Supplementation with serum did not improve the viability of PAF and nitrite levels in culture medium served as an indicator of viability of granulose cells from caprine PAF in vitro cultured

    The protein obtained from the microorganisms is not only cheap but can be used as additive to provide a balanced nutrition for many animals feeding. The aim of this work was the single cell protein (SCP) production by Candida robusta URM5293 using sugarcane bagasse as substrate. The yeast C. robusta URM5293 was isolated from root sugarcane and was identified based on morphological and biochemical characteristics. Biomass production was done into Erlenmayers flasks (250 mL) containing culture medium supplemented of sugarcane bagasse hydrolyzate. Fermentations were carried varying culture conditions through the study of four different variables: temperature (25, 30, and 35°C), agitation intensity (110, 140 or 170 rpm), pH (6.0, 7.0, and 8.0) and production time (72, 96, and 120h), according to a 24-1 fractional factorial design. Results demonstrated that this yeast was able to ensure the highest level of biomass (141 g/L) when cultivated at 25°C, pH 6.0, 170 rpm of agitation intensity after 72h of cultivation using sugarcane bagasse. The present results demonstrate the potential of sugarcane bagasse hemicellulosic hydrolyzate as a substrate for the production of microbial protein by C. robusta URM5293

    Resumo O artigo tem como objetivo relacionar a geopaisagem e os sítios arqueológicos com pinturas rupestres dos municípios de Venturosa, Pedra e Buíque, a partir da análise e interpretação de dados geológicos, geomorfológicos, morfoestratigráficos e da distribuição espacial. A relação entre a paisagem dos espaços estudados, tendo em vista as transformações ocorridas ao longo do tempo com os sítios arqueológicos, permite ao pesquisador compreender a dinâmica da adaptação do homem ao meio ambiente e da escolha dos sítios, o que pode estar relacionada aos aspectos da paisagem. Esta análise busca expor a lógica de apropriação do meio natural pelos grupos humanos que habitaram aquelas regiões em tempos pretéritos

    RACIONAL: O transplante hepático é o único tratamento efetivo para as hepatopatias crônicas terminais e a taxa de sobrevida tem aumentado nas últimas décadas. No entanto, as complicações biliares têm alta incidência e permanecem como o "calcanhar de Aquiles" do transplante de fígado. OBJETIVO: Avaliar retrospectivamente os resultados do tratamento endoscópico das complicações biliares em pacientes submetidos à transplante hepático. MÉTODOS: Foram avaliados pacientes transplantados hepáticos para realização de colangiopancreatografia retrógrada endoscópica por suspeita de complicação biliar. RESULTADOS: Quinze pacientes (11 homens, média de idade de 49,57 anos) foram estudados. Nesse período foram realizadas 36 colangiopancreatografias retrógradas endoscópicas (2,4/paciente). Neste grupo, 100% receberam órgão de doador falecido. Estenose da anastomose coledococoledocociana foi diagnosticada em 13 pacientes e o sucesso da terapêutica endoscópica foi de 53,84% (38,46% ainda em tratamento). Fístula biliar foi diagnosticada em um paciente, sendo resolvida pelo tratamento endoscópico. Disfunção da âmpola hepatopancreaticobiliar com coledocolitíase foi diagnosticada em um paciente, também resolvida pela terapêutica endoscópica. CONCLUSÕES: As complicações biliares pós-transplante hepático são relativamente comuns, com predominância de estenoses. O tratamento endoscópico foi eficaz na maioria dos pacientes desta série.<br>BACKGROUND: Liver transplantation is the only effective treatment for chronic liver diseases and terminal survival rate has increased in recent decades. However, biliary complications have high incidence and remain as the "Achilles heel" for liver transplantation. AIM: To evaluate retrospectively endoscopic treatment outcomes of biliary complications in post-liver transplantations. METHODS: The sample consisted of post-liver transplantation patients for endoscopic retrograde cholangiopancreatography due to suspected biliary complications. RESULTS: Fifteen patients were included (10 male, mean age of 49.57 years) and 36 endoscopic retrograde cholangiopancreatographies were undertaken (2.4/patient). Biliary stricture was diagnosed in 13 patients and endoscopic treatment was successful in 56% (38,46% still in treatment). Biliary leaks were found in one patient and dysfunction of the hepatobilliary ampulla with choledocholithiasis was diagnosed in one patient, both cured by endoscopic treatment. CONCLUSIONS: Post-liver transplantation biliary complications are relatively common and endoscopic treatment may result in satisfactory outcome. Stenosis was the more frequent complication in this series

    Arterial bullet embolism after thoracic gunshot wound

    Abstract Bullet embolization of the arterial or venous systems is a rare complication of penetrating gunshot injuries. A 29-year-old man presented at the emergency department with a gunshot wound to the left arm, which had transfixed the arm and entered the thorax, with no exit wound. Initial radiographies showed a projectile in the upper left thigh. Contrast-enhanced tomography showed a pseudo-aneurysm of the descending thoracic aorta and the bullet inside the proximal left superficial femoral artery. Physical examination found diminished left pedal pulses, and the patient complained of left toe numbness. Endovascular thoracic aortic pseudoaneurysm repair was performed, sealing the descending aortic orifice with an endograft, and thromboembolectomy/bullet retrieval was carried out via a left femoral incision, both successfully. Considering that diagnosis of missile emboli depends on a high degree of suspicion, physicians who manage gunshot wound patients must be acutely aware of the possibility of intravascular bullet embolism

    Impact of pericardiectomy on exercise capacity and sleep of patients with chronic constrictive pericarditis.

    BackgroundHeart failure is associated with exercise intolerance and sleep- disordered breathing; however, studies in patients with chronic constrictive pericarditis are scarce. The purpose of our study was to assess exercise capacity and sleep in patients with chronic constrictive pericarditis (CCP) undergoing a pericardiectomy.MethodsWe studied consecutive patients scheduled for pericardiectomy due to symptomatic CCP. Were performed quality of life (Minnesota Living with Heart Failure Questionnaire-MLHFQ) and sleep questionnaires (Epworth, Pittsburgh Sleep Quality Index-PSQI), serum B-type natriuretic peptide (BNP), serum C-reactive protein, transthoracic echocardiography, cardiopulmonary exercise test and overnight polysomnography immediately before and six months after pericardiectomy.ResultsTwenty-five patients (76% males, age: 45.5±13.8 years, body mass index: 24.9±3.7 kg/m2, left ventricular ejection fraction: 60±6%) with CCP (76% idiopathic, 12% tuberculosis) were studied. As compared to the preoperative period, pericardiectomy resulted in reduction in BNP (143 (83.5-209.5) vs 76 (40-117.5) pg/mL, p = 0.011), improvement in VO2 peak (18.7±5.6 vs. 25.2±6.3 mL/kg/min, pConclusionPatients with symptomatic CCP showed reduced exercise capacity and sleep-disordered breathing. After pericardiectomy, there was improvement in exercise capacity and neutral effect on sleep-disordered breathing
