    The Infection of Vibrio Parahaemolyticus in Shrimp and Human

    Vibrio parahaemolyticus is an aquatic zoonotic agent that can threaten human and aquaculture animal health. Humans can be infected by consuming contaminated raw seafood or wound-related infections. Generally infection of V. parahemolyticus is orally transmitted and causes gastroenteritis in humans while in aquaculture animals especially shrimp can cause Acute Hepatopancreatic Necrosis Disease (AHPND) or Early Mortality Syndrome (EMS) with a very high mortality rate and cause economic losses. Shrimp species susceptible to infection are Litopenaeus vannamei, Penaeus monodon, and P. chinensis. V. parahaemolyticus produces several toxins in human disease such as thermostable direct hemolysin (TDH), TDH-related haemolysin (TRH), and thermolabile hemolysin (TLH). Meanwhile, Photorabdus insect-related (Pir) toxins consisting of PirAvp and PirBvp are the toxins associated with AHPND in shrimp. The genes that encode the toxin are used as targets to diagnose V. parahaemolyticus pathogens molecularly. Until now the treatment of V. parahaemolyticus infection is using antibiotics and fluid therapy, but there were V. parahaemolyticus isolates from aquaculture that have been resistant to antibiotics so that the use of antibiotics in aquaculture must be controlled and the use of alternative therapy are very important to be developed to control V. parahaemolyticus infection

    Tujuan dari penelitian adalah untuk mengetahui apakah aplikasi SIM-TPK berpengaruh terhadap kinerja karyawan pada PT. Pelabuhan Indonesia II (Persero) Cabang Palembang. Metode penelitian yang digunakan penulis untuk menganalisis pengaruh tersebut adalah metode regresi linear, dimana variabel yang digunakan adalah Usability sebagai variabel bebas (x) dan kinerja karyawan sebagai variabel terikat (y). Hasil dari penelitian ini didapatkan dari penyebaran data berupa kuesioner dengan responden karyawan/i PT. Pelabuhan Indonesia II (Persero) Cabang Palembang sebagai pengguna SIM-TPK. Data dianalisis dengan regresi linear sederhana menggunakan software statistik SPSS versi 19, yang menunjukkan bahwa faktor usability berpengaruh terhadap kinerja karyawan yang menggunakan aplikasi SIM-TPK, yang dapat dilihat dari hasil tingkat signifikan yaitu 0,000 yang berarti lebih kecil dari 0,05 sehingga H 1 diterima sedangkan H0 ditolak. Dengan faktor usability sebuah sistem yang baik, maka kinerja karyawan/i yang menggunakan aplikasi SIM-TPK pada PT. Pelabuhan Indonesia II (Persero) Cabang Palembang pun ikut meningkat

    De novo assembly of transcriptomes from a B73 maize line introgressed with a QTL for resistance to gray leaf spot disease reveals a candidate allele of a lectin receptor-like kinase

    Gray leaf spot (GLS) disease in maize, caused by the fungus Cercospora zeina, is a threat to maize production globally. Understanding the molecular basis for quantitative resistance to GLS is therefore important for food security. We developed a de novo assembly pipeline to identify candidate maize resistance genes. Near-isogenic maize lines with and without a QTL for GLS resistance on chromosome 10 from inbred CML444 were produced in the inbred B73 background. The B73-QTL line showed a 20% reduction in GLS disease symptoms compared to B73 in the field (p = 0.01). B73-QTL leaf samples from this field experiment conducted under GLS disease pressure were RNA sequenced. The reads that did not map to the B73 or C. zeina genomes were expected to contain novel defense genes and were de novo assembled. A total of 141 protein-coding sequences with B73-like or plant annotations were identified from the B73-QTL plants exposed to C. zeina. To determine whether candidate gene expression was induced by C. zeina, the RNAseq reads from C. zeina-challenged and control leaves were mapped to a master assembly of all of the B73-QTL reads, and differential gene expression analysis was conducted. Combining results from both bioinformatics approaches led to the identification of a likely candidate gene, which was a novel allele of a lectin receptor-like kinase named L-RLK-CML that (i) was induced by C. zeina, (ii) was positioned in the QTL region, and (iii) had functional domains for pathogen perception and defense signal transduction. The 817AA L-RLK-CML protein had 53 amino acid differences from its 818AA counterpart in B73. A second "B73-like" allele of L-RLK was expressed at a low level in B73-QTL. Gene copy-specific RT-qPCR confirmed that the l-rlk-cml transcript was the major product induced four-fold by C. zeina. Several other expressed defense-related candidates were identified, including a wall-associated kinase, two glutathione s-transferases, a chitinase, a glucan beta-glucosidase, a plasmodesmata callose-binding protein, several other receptor-like kinases, and components of calcium signaling, vesicular trafficking, and ethylene biosynthesis.     Karakteristik Tipologi Permukiman Kampung Jawa di Dusun Wanasari Denpasar, Bali

    A slum settlement constitutes a residential environmental condition with a quality far below the occupancy worthiness standard. Slum neighborhoods are characterized by high building density, low environmental quality, poor infrastructure, and social problems that occur. One area in Denpasar City that belongs to slum settlement is Wanasari subvillage, Dauh Puri Kaja, North Denpasar. Kampung Jawa settlement area is divided into 9 RT (Neighborhood Associations). This area is not fully qualified for a slum, which can be divided into typologies of mild slum, moderate slum, and heavy slum levels. In general, each RT's physical conditions have various characteristics. This study was at least to analyze the characteristics of each typology group of slums. The research method used was mixed methods, in which the research stages were started by identifying the characteristics of the slum settlements in each RT area having been categorized according to the existing slum levels. The process of classifying these categories was carried out by observing and identifying slum settlements by looking at the physical aspects of the environment, the socio-economic conditions of the community, environmental facilities and infrastructure, and the environmental hazard conditions. The results of the study obtained indicate that the basic factors influencing the characteristics of Kampung Jawa settlement are the level of education, the type of occupation, the level of the economy, and the locations of the RT. The lower the factors that are owned, the lower the quality of the physical environment, infrastructure, and the level of hazard formed in the area of Kampung Jawa settlement. Lingkungan permukiman kumuh ditandai dengan kepadatan bangunan yang tinggi, kualitas lingkungan yang rendah, infrastruktur yang tidak baik, serta permasalahan sosial yang terjadi di dalamnya. Salah satu kawasan di Kota Denpasar yang memiliki permukiman kumuh berada di Dusun Wanasari, Dauh Puri Kaja, Denpasar Utara. Kawasan permukiman Kampung Jawa terbagi atas 9 RT. Kawasan ini tidak sepenuhnya memiliki kualifikasi kumuh, dapat dibagi menjadi tipologi dengan tingkat kekumuhan ringan, kumuh sedang, dan kumuh berat. Secara umum, kondisi fisik masing – masing RT memiliki karakteristik yang beragam. Tujuan penelitian ini adalah untuk menganalisis karakteristik masing-masing kelompok tipologi kekumuhan tersebut. Metode penelitian yang digunakan metode campuran (mix methods), tahapan penelitian dimulai dengan mengidentifikasi karakteristik permukiman kumuh di masing – masing wilayah RT yang telah dikategorikan sesuai dengan tingkat kekumuhan yang ada. Proses mengelompokkan kategori ini dengan cara mengobservasi dan mengidentifikasi permukiman kumuh dengan melihat aspek fisik lingkungan, keadaan sosial – ekonomi masyarakatnya, sarana dan prasarana lingkungan, serta kondisi bahaya lingkungan tersebut. Hasil penelitian yang diperoleh menunjukkan bahwa faktor dasar yang mempengaruhi karakteristik permukiman di Kampung Jawa ini adalah tingkat pendidikan, jenis pekerjaan, dan tingkat perekonomian, dan letak lokasi RT. Semakin rendah faktor yang dimiliki semakin rendah pula kualitas lingkungan fisik, sarana prasarana, dan tingkat bahaya yang terbentuk di wilayah permukiman Kampung Jawa

    Knowing about the problem of Watershed is very important to manage the river environment. Oneof the indicator to know the watershed problem is concentration of suspend load sediment.Thisresearch conduct to observe about relationship between stream flow debit and Suspend Load ofSediment which influenced hydrologic condition on Padang Hilir sub Watershed. This researchconduct to from May until October 2013 at Padang Hilir Sub Watershed, use a survey method withrandom sampling. Water sampling is taken out for three times in a week at after rain or no rain forsix months so that total water sample as much as 82 sample. The result show that concentration ofload sediment at no rain measure have highest value in October as big as 256,67 mg/L (worse)and the lowest in July as big as 43,3 mg/L (good),meanwhile at after rain and no rain measurehave highest value at August for 416,67 mg/L (very worse) and the lowest in July as big as 25mg/L (good). At no and after rain relationship betwen concentration of suspend load sediment andstream flow showed by equation : Y = 74,721 + 0,895x with signification value was 0,023 anddetermination value (r2) was 0,014 ( very weak). Meanwhile at not rain relationship betwenconcentration of suspend load sediment on stream flow regression showed by equation :Y = 28,741 +0,030x with stream flow signification value as big as 0,046 and determination value(r2) was 0,065 (weak)
