Interferon Beta 2/Interleukin-6 and Interleukin-3 Synergize in Stimulating Proliferation of Human Early Hematopoietic Progenitor Cells.
Early 4-hydroxyperoxycyclophosphamide (4-HC) resistant hematopoietic progenitor cells (pre-colony-forming units, pre-CFU) were evaluated by a two-step liquid culture system, (earlier progenitors), pre-CFU, as well as by the conventional semi-solid mixed colony assay (later progenitors) for their growth response to interleukin-6 (IL-6), interleukin-3 (IL-3), and a combination of both factors. The effect of the IL-6/IL-3 combination was compared to that of IL-1/IL-3. IL-3 alone proved less effective in supporting earlier pre-CFU cells than later progenitor cells. In a previous work IL-6 promoted the growth of early multipotential progenitor cells circulating in hairy cell leukemia (HCL) patients. IL-6 alone did not stimulate growth of either early or later normal progenitor cells. However, a significant synergistic effect was obtained when IL-6 and IL-3 were added together (p \u3c0.05). IL-6/IL-3 synergism was more potent than IL-1/IL-3 in promoting growth of colonies. The previously described synergistic effect of IL-1/IL-3 seems to be independent of IL-6. Thus, our results suggest that the multifunctional cytokine IL-6, may be of use in shortening the engraftment time in bone marrow transplantation
Assessment of Domestic Well-Being: From Perception to Measurement
Nowadays, there are plenty of sensing devices that enable the measurement of physiological, environmental, and behavioral parameters of people 24 hours a day, seven days a week and provide huge quantities of different data. Data and signals coming from sensing devices, installed in indoor or outdoor environments or often worn by the users, generate heterogeneous and complex structured datasets, most of the time not uniformly structured. The artificial intelligence (AI) algorithms applied to these sets of data have demonstrated capabilities to infer indices related to a subject's status and well-being [1]. Well-being is a key parameter in the World Health Organization (WHO) definition of health, considering its physical, mental, and social spheres. Quantitatively assessing a subject's well-being is of paramount importance if we want to assess the whole status of a person, which is particularly useful in the case of ageing people living alone. Assessment allows for continuous remote monitoring to improve people's quality of life (QoL) according to their perceptions, needs, and preferences. Technology undoubtedly plays a pivotal role in this regard, providing us new tools to support the objective evaluation of a subject's status, including her/his perception of the living environment. Its potential is huge, also in terms of support to the healthcare system and ageing people; however, there are several engineering challenges to consider, especially in terms of sensors integrability, connectivity, and metrological performance, in order to obtain reliable and accurate measurement systems
Altruism during predation in an assassin bug
Zelus annulosus is an assassin bug species mostly noted on Hirtella physophora, a myrmecophyte specifically associated with the ant Allomerus decemarticulatus known to build traps on host tree twigs to ambush insect preys. The Z. annulosus females lay egg clutches protected by a sticky substance. To avoid being trapped, the first three instars of nymphs remain grouped in a clutch beneath the leaves on which they hatched, yet from time to time, they climb onto the upper side to group ambush preys. Long-distance prey detection permits these bugs to capture flying or jumping insects that alight on their leaves. Like some other Zelus species, the sticky substance of the sundew setae on their forelegs aids in prey capture. Group ambushing permits early instars to capture insects that they then share or not depending on prey size and the hunger of the successful nymphs. Fourth and fifth instars, with greater needs, rather ambush solitarily on different host tree leaves, but attract siblings to share large preys. Communal feeding permits faster prey consumption, enabling small nymphs to return sooner to the shelter of their leaves. By improving the regularity of feeding for each nymph, it likely regulates nymphal development, synchronizing molting and subsequently limiting cannibalism
Interleukin-6 and interleukin-6 receptor secretion by chronic lymphatic leukaemia and normal B lymphocytes: effect of PMA and PWM
Interleukin-6 (IL-6) and soluble interleukin-6 receptor (sIL-6R) were detected in supernatants of cultures of B chronic lymphatic leukaemia (CLL) lymphocytes. Phorbol-12-myristate 13 acetate (PMA) caused a decrease in the levels of IL-6 in 14 out of 16 cultures and an increase in levels of sIL6R in all 15 cases. The effect of pokeweed mitogen (PWM) was variable and not significant. The levels of IL-6 were below the detection limit (60 pg/ml) in sera of 13 CLL patients whereas sIL-6R was detected (13 ng/ml to 97 ng/ml) in the 13 sera. IL6 was not detected in cultures of unstimulated or stimulated with PMA or PWM normal human B cells. Levels of sIL-6R were minimal in cultures of normal B lymphocytes and were increased in PMA stimulated cultures. The results are consistent with the view that B-CLL cells produce spontaneously IL-6 which could act in an autocrine fashion to cause shedding of surface IL-6R and account for the correlation found between serum levels of sIL-6R and B-CLL lymphocyte numbers. The fall in levels of IL-6 in PMA stimulated CLL cultures might express masking or degradation of IL-6 after combination with the receptor
On the electron energy distribution function in the high power impulse magnetron sputtering discharge
We apply the Ionization Region Model (IRM) and the Orsay Boltzmann equation
for ELectrons coupled with Ionization and eXcited states kinetics (OBELIX)
model to study the electron kinetics of a high power impulse magnetron
sputtering (HiPIMS) discharge. In the IRM the bulk (cold) electrons are assumed
to exhibit a Maxwellian energy distribution and the secondary (hot) electrons,
emitted from the target surface upon ion bombardment, are treated as a high
energy tail, while in the OBELIX the electron energy distribution is calculated
self-consistently using an isotropic Boltzmann equation. The two models are
merged in the sense that the output from the IRM is used as an input for
OBELIX. The temporal evolutions of the particle densities are found to agree
very well between the two models. Furthermore, a very good agreement is
demonstrated between the bi-Maxwellian electron energy distribution assumed by
the IRM and the electron energy distribution calculated by the OBELIX model. It
can therefore be concluded that assuming a bi-Maxwellian electron energy
distribution, constituting a cold bulk electron group and a hot secondary
electron group, is a good approximation for modeling the HiPIMS discharge

Repression of the Hox gene abd-A by ELAV-mediated Transcriptional Interference
Intergenic transcription is a common feature of eukaryotic genomes and performs important and diverse cellular functions. Here, we investigate the iab-8 ncRNA from the Drosophila Bithorax Complex and show that this RNA is able to repress the transcription of genes located at its 3â end by a sequence-independent, transcriptional interference mechanism. Although this RNA is expressed in the early epidermis and CNS, we find that its repressive activity is limited to the CNS, where, in wild-type embryos, it acts on the Hox gene, abd-A, located immediately downstream of it. The CNS specificity is achieved through a 3â extension of the transcript, mediated by the neuronal-specific, RNA-binding protein, ELAV. Loss of ELAV activity eliminates the 3â extension and results in the ectopic activation of abd-A. Thus, a tissue-specific change in the length of a ncRNA is used to generate a precise pattern of gene expression in a higher eukaryote
North western Alps Holocene paleohydrology recorded by flooding activity in Lake Le Bourget, France and possible relations with Mont-Blanc glaciers fluctuations
International audienceA 14-m long piston core was retrieved from Lake Le Bourget, NWAlps (France), in order to provide a continuous record of flooding events of the Rhone River during the Holocene. The selection of the coring site was based on high resolution seismic profiling, in an area with limited mass wasting deposits and accumulated proximal Rhone River inter-and underflow deposits. The age-depth model of this core is based on (i) 14 AMS radiocarbon dates, (ii)radionuclide dating(137Cs) and (iii) the identification of historical data (flood events, eutrophication of the lake).The sedimentary record dates back to 9400 cal BP, and includes a thin mass wasting event deposited around 4500 cal BP. A multi-proxy approach was used to track the evolution and origin of clastic sedimentation during the Holocene, in order to identify periods of higher hydrologic al activity in the catchment area. Spectrophotometry was used to detect fluctuations in clastic supply and the study of clay minerals (especially the Illite crystallinity index) allowed locating the main source area of fine grained clastic particles settling at the lake after flood events. This dataset highlights up to 12 periods of more intense flooding events over the last 9400 years in Lake Le Bourget and shows that the main source area of clastic particles during this period is the upper part of the Arve River drainage basin. This part of the catchment area drains several large glaciers from the Mont-Blanc Massif, and fluctuations in Rhone River flood supply in Lake Le Bourget is interpreted as resulting essentially from Mont-Blanc Glacier activity during the Holocene.The comparison of clastic sedimentationin Lake Le Bourget with periods of increasing land use and periods of Alpine glacier and mid-European lake level fluctuations, suggest that the core LDB04 clastic record in Lake Le Bourget is a continuous proxy of the Holocene hydrologic al history of the NW Alps
