    The photoinduced transition in magnetoresistive manganites: a comprehensive view

    We use femtosecond x-ray diffraction to study the structural response of charge and orbitally ordered Pr1x_{1-x}Cax_xMnO3_3 thin films across a phase transition induced by 800 nm laser pulses. By investigating the dynamics of both superlattice reflections and regular Bragg peaks, we disentangle the different structural contributions and analyze their relevant time-scales. The dynamics of the structural and charge order response are qualitatively different when excited above and below a critical fluence fcf_c. For excitations below fcf_c the charge order and the superlattice is only partially suppressed and the ground state recovers within a few tens of nanosecond via diffusive cooling. When exciting above the critical fluence the superlattice vanishes within approximately half a picosecond followed by a change of the unit cell parameters on a 10 picoseconds time-scale. At this point all memory from the symmetry breaking is lost and the recovery time increases by many order of magnitudes due to the first order character of the structural phase transition

    Suicide genes: Monitoring cells in patients with a safety switch

    Clinical trials increasingly incorporate suicide genes either as direct lytic agents for tumors or as safety switches in therapies based on genetically modified cells. Suicide genes can also be used as non-invasive reporters to monitor the biological consequences of administering genetically modified cells to patients and gather information relevant to patient safety. These genes can monitor therapeutic outcomes addressable by early clinical intervention. As an example, our recent clinical trial used 18F-9-(4-fluoro-3-hydroxymethylbutyl)guanine (18FHBG) and PET/CT scans to follow T cells transduced with herpes simplex virus thymidine kinase (TK) after administration to patients. Guided by preclinical data we ultimately hope to discern whether a particular pattern of transduced T cell migration within patients reflects early development of Graft vs. Host Disease (GvHD). Current difficulties in terms of choice of suicide gene, biodistribution of radiolabeled tracers in humans versus animal models, and threshold levels of genetically modified cells needed for detection by PET/CT are discussed. As alternative suicide genes are developed, additional radiolabel probes suitable for imaging in patients should be considered

    Discovery of a Wolf-Rayet Star Through Detection of its Photometric Variability

    We report the serendipitous discovery of a heavily reddened Wolf-Rayet star that we name WR142b. While photometrically monitoring a cataclysmic variable, we detected weak variability in a nearby field star. Low-resolution spectroscopy revealed a strong emission line at 7100 Ang., suggesting an unusual object and prompting further study. A spectrum taken with the Hobby-Eberly Telescope confirms strong HeII emission and a NIV 7112 Ang. line consistent with a nitrogen-rich Wolf-Rayet star of spectral class WN6. Analysis of the HeII line strengths reveals no detectable hydrogen in WR142b. A blue-sensitive spectrum obtained with the Large Binocular Telescope shows no evidence for a hot companion star. The continuum shape and emission line ratios imply a reddening of E(B-V)=2.2 to 2.5 mag.     Light curves and colours of the faint Uranian irregular satellites Sycorax, Prospero, Stephano, Setebos and Trinculo

    After the work of Gladman et al. (1998), it is now assessed that many irregular satellites are orbiting around Uranus. Despite many studies have been performed in past years, very few is know for the light-curves of these objects and inconsistencies are present between colours derived by different authors. This situation motivated our effort to improve both the knowledge of colours and light curves. We present and discuss time series observations of Sycorax, Prospero, Stephano, Setebos and Trinculo, five faint irregular satellites of Uranus, carried out at VLT, ESO Paranal (Chile) in the nights between 29 and 30 July, 2005 and 25 and 30 November, 2005. We derive light curves for Sycorax and Prospero and colours for all of these these bodies. For Sycorax we obtain colours B-V =0.839 +/- 0.014, V-R = 0.531 +/- 0.005 and a light curve which is suggestive of a periodical variation with period about 3.6 hours and amplitude about 0.067 +/- 0.004 mag. The periods and colours we derive for Sycorax are in agreement with our previous determination in 1999 using NTT. We derive also a light-curve for Prospero which suggests an amplitude of about 0.2 mag and a periodicity of about 4 hours. However, the sparseness of our data, prevents a more precise characterization of the light-curves, and we can not determine wether they are one-peaked or two-peaked. Hence, these periods and amplitudes have to be considered preliminary estimates. As for Setebos, Stephano and Trinculo the present data do not allow to derive any unambiguous periodicity, despite Setebos displays a significant variability with amplitude about as large as that of Prospero.     Regulation of releasable vesicle pool sizes by protein kinase A-dependent phosphorylation of SNAP-25

    AbstractProtein kinase A (PKA) is a key regulator of neurosecretion, but the molecular targets remain elusive. We combined pharmacological manipulations of kinase and phosphatase activities with mutational studies on the exocytotic machinery driving fusion of catecholamine-containing vesicles from chromaffin cells. We found that constitutive PKA activity was necessary to maintain a large number of vesicles in the release-ready, so-called primed, state, whereas calcineurin (protein phosphatase 2B) activity antagonized this effect. Overexpression of the SNARE protein SNAP-25a mutated in a PKA phosphorylation site (Thr-138) eliminated the effect of PKA inhibitors on the vesicle priming process. Another, unidentified, PKA target regulated the relative size of two different primed vesicle pools that are distinguished by their release kinetics. Overexpression of the SNAP-25b isoform increased the size of both primed vesicle pools by a factor of two, and mutations in the conserved Thr-138 site had similar effects as in the a isoform
