Analysis of the neutral fluxes in the divertor region of Wendelstein 7-X under attached and detached conditions using EMC3-EIRENE
This paper analyzes the neutral fluxes in the divertor region of the W7-X standard configuration for different input powers, both under attached and detached conditions. The performed analysis is conducted through EMC3-EIRENE simulations. They show the importance of the horizontal divertor to generate neutrals, and resolve the neutral plugging in the divertor region. Simulations of detached cases show a decrease in the number of generated neutrals compared to the attached simulations, in addition to a higher fraction of the ion flux arriving on the baffles during detachment. As the ionization takes place further inside the plasma during detachment, a larger percentage of the generated neutral particles leave the divertor as neutrals. The leakage in the poloidal and toroidal direction increases, just as the fraction of collected particles at the pumping gap. The fraction of pumped particles increases with a factor two, but stays below one percent. This demonstrates that detachment with the current target geometry, although it improves the power exhaust, is not yet leading to an increased particle exhaust
Bacillus subtilis MreB Orthologs Self-Organize into Filamentous Structures underneath the Cell Membrane in a Heterologous Cell System
Actin-like bacterial cytoskeletal element MreB has been shown to be essential for the maintenance of rod cell shape in many bacteria. MreB forms rapidly remodelling helical filaments underneath the cell membrane in Bacillus subtilis and in other bacterial cells, and co-localizes with its two paralogs, Mbl and MreBH. We show that MreB localizes as dynamic bundles of filaments underneath the cell membrane in Drosophila S2 Schneider cells, which become highly stable when the ATPase motif in MreB is modified. In agreement with ATP-dependent filament formation, the depletion of ATP in the cells lead to rapid dissociation of MreB filaments. Extended induction of MreB resulted in the formation of membrane protrusions, showing that like actin, MreB can exert force against the cell membrane. Mbl also formed membrane associated filaments, while MreBH formed filaments within the cytosol. When co-expressed, MreB, Mbl and MreBH built up mixed filaments underneath the cell membrane. Membrane protein RodZ localized to endosomes in S2 cells, but localized to the cell membrane when co-expressed with Mbl, showing that bacterial MreB/Mbl structures can recruit a protein to the cell membrane. Thus, MreB paralogs form a self-organizing and dynamic filamentous scaffold underneath the membrane that is able to recruit other proteins to the cell surface
Mutational processes contributing to the development of multiple myeloma.
To gain insight into multiple myeloma (MM) tumorigenesis, we analyzed the mutational signatures in 874 whole-exome and 850 whole-genome data from the CoMMpass Study. We identified that coding and non-coding regions are differentially dominated by distinct single-nucleotide variant (SNV) mutational signatures, as well as five de novo structural rearrangement signatures. Mutational signatures reflective of different principle mutational processes-aging, defective DNA repair, and apolipoprotein B editing complex (APOBEC)/activation-induced deaminase activity-characterize MM. These mutational signatures show evidence of subgroup specificity-APOBEC-attributed signatures associated with MAF translocation t(14;16) and t(14;20) MM; potentially DNA repair deficiency with t(11;14) and t(4;14); and aging with hyperdiploidy. Mutational signatures beyond that associated with APOBEC are independent of established prognostic markers and appear to have relevance to predicting high-risk MM
Parametrisation of target heat flux distribution and study of transport parameters for boundary modelling in W7-X
Modelling the scrape-off layer of a stellarator is challenging due to the
complex magnetic 3D geometry. The here presented study analyses simulations of
the scrape-off layer (SOL) of the stellarator Wendelstein 7-X (W7-X) using the
EMC3-EIRENE code for the magnetic standard configuration. Comparing with
experimental observations, the transport model is validated. Based on the
experimentally observed strike line width, the anomalous transport
coefficients, used as input to the code are determined to around
ms. This is however in disagreement with upstream measurements,
where such small cross-field transport leads to temperatures higher than
measured experimentally
