    Robo-AO Discovery and Basic Characterization of Wide Multiple Star Systems in the Pleiades, Praesepe, and NGC 2264 Clusters

    We identify and roughly characterize 66 candidate binary star systems in the Pleiades, Praesepe, and NGC 2264 star clusters based on robotic adaptive optics imaging data obtained using Robo-AO at the Palomar 60" telescope. Only ∼\sim10% of our imaged pairs were previously known. We detect companions at red optical wavelengths having physical separations ranging from a few tens to a few thousand AU. A 3-sigma contrast curve generated for each final image provides upper limits to the brightness ratios for any undetected putative companions. The observations are sensitive to companions with maximum contrast ∼\sim6m^m at larger separations. At smaller separations, the mean (best) raw contrast at 2 arcsec is 3.8m^m (6m^m), at 1 arcsec is 3.0m^m (4.5m^m), and at 0.5 arcsec is 1.9m^m (3m^m). PSF subtraction can recover close to the full contrast in to the closer separations. For detected candidate binary pairs, we report separations, position angles, and relative magnitudes. Theoretical isochrones appropriate to the Pleiades and Praesepe clusters are then used to determine the corresponding binary mass ratios, which range from 0.2-0.9 in q=m2/m1q=m_2/m_1. For our sample of roughly solar-mass (FGK type) stars in NGC 2264 and sub-solar-mass (K and early M-type) primaries in the Pleiades and Praesepe, the overall binary frequency is measured at ∼\sim15.5% ±\pm 2%. However, this value should be considered a lower limit to the true binary fraction within the specified separation and mass ratio ranges in these clusters, given that complex and uncertain corrections for sensitivity and completeness have not been applied.Comment: Accepted to A

    Evaluation of novel ß-ribosidase substrates for the differentiation of Gram-negative bacteria

    Aims:  To synthesize novel substrates for the detection of β-ribosidase and assess their potential for the differentiation of Gram-negative bacteria. Methods and Results:  Two novel chromogenic substrates, 3′,4′-dihydroxyflavone-4′-β-D-ribofuranoside (DHF-riboside) and 5-bromo-4-chloro-3-indolyl-β-D-ribofuranoside (X-riboside) were evaluated along with a known fluorogenic substrate, 4-methylumbelliferyl-β-D-ribofuranoside (4MU-riboside). A total of 543 Gram-negative bacilli were cultured on media containing either DHF-riboside or X-riboside. Hydrolysis of DHF-riboside or X-riboside resulted in the formation of clearly distinguishable black or blue-green colonies, respectively. Hydrolysis of 4MU-riboside was evaluated in a liquid medium in microtiter trays and yielded blue fluorescence on hydrolysis which was measured using fluorimetry. β-Ribosidase activity was widespread with 75% of strains, including 85·6% of Enterobacteriaceae, showing activity with at least one substrate. Genera that demonstrated β-ribosidase activity included Aeromonas, Citrobacter, Enterobacter, Escherichia, Hafnia, Klebsiella, Morganella, Providencia, Pseudomonas, Salmonella and Shigella. In contrast, strains of Proteus spp., Acinetobacter spp., Yersinia enterocolitica, Vibrio cholerae and Vibrio parahaemolyticus generally failed to demonstrate β-ribosidase activity. Conclusions:  The novel substrates DHF-riboside and X-riboside are effective for the detection of β-ribosidase in agar-based media and may be useful for the differentiation and identification of Gram-negative bacteria. Significance and Impact of the Study:  This is the first report describing the application and utility of chromogenic substrates for β-ribosidase. These substrates could be applied in chromogenic media for differentiation of Gram-negative bacteria

    Association of Sperm-Associated Antigen 5 and Treatment Response in Patients With Estrogen Receptor-Positive Breast Cancer

    There is no proven test that can guide the optimal treatment, either endocrine therapy or chemotherapy, for estrogen receptor-positive breast cancer. Objective: To investigate the associations of sperm-associated antigen 5 (SPAG5) transcript and SPAG5 protein expressions with treatment response in systemic therapy for estrogen receptor-positive breast cancer. Design, Settings, and Participants: This retrospective cohort study included patients with estrogen receptor-positive breast cancer who received 5 years of adjuvant endocrine therapy with or without neoadjuvant anthracycline-based combination chemotherapy (NACT) derived from 11 cohorts from December 1, 1986, to November 28, 2019. The associations of SPAG5 transcript and SPAG5 protein expression with pathological complete response to NACT were evaluated, as was the association of SPAG5 mRNA expression with response to neoadjuvant endocrine therapy. The associations of distal relapse-free survival with SPAG5 transcript or SPAG5 protein expressions were analyzed. Data were analyzed from September 9, 2015, to November 28, 2019. Main Outcomes and Measures: The primary outcomes were breast cancer-specific survival, distal relapse-free survival, pathological complete response, and clinical response. Outcomes were examined using Kaplan-Meier, multivariable logistic, and Cox regression models. Results: This study included 12 720 women aged 24 to 78 years (mean [SD] age, 58.46 [12.45] years) with estrogen receptor-positive breast cancer, including 1073 women with SPAG5 transcript expression and 361 women with SPAG5 protein expression of locally advanced disease stage IIA through IIIC. Women with SPAG5 transcript and SPAG5 protein expressions achieved higher pathological complete response compared with those without SPAG5 transcript or SPAG5 protein expressions (transcript: odds ratio, 2.45 [95% CI, 1.71-3.51]; P < .001; protein: odds ratio, 7.32 [95% CI, 3.33-16.22]; P < .001). Adding adjuvant anthracycline chemotherapy to adjuvant endocrine therapy for SPAG5 mRNA expression in estrogen receptor-positive breast cancer was associated with prolonged 5-year distal relapse-free survival in patients without lymph node involvement (hazard ratio, 0.34 [95% CI, 0.14-0.87]; P = .03) and patients with lymph node involvement (hazard ratio, 0.35 [95% CI, 0.18-0.68]; P = .002) compared with receiving 5-year endocrine therapy alone. Mean (SD) SPAG5 transcript was found to be downregulated after 2 weeks of neoadjuvant endocrine therapy compared with pretreatment levels in 68 of 92 patients (74%) (0.23 [0.18] vs 0.34 [0.24]; P < .001). Conclusions and Relevance: These findings suggest that SPAG5 transcript and SPAG5 protein expressions could be used to guide the optimal therapies for estrogen receptor-positive breast cancer. Retrospective and prospective clinical trials are warranted

    Cataclysmic Variables in the First Year of the Zwicky Transient Facility

    Using selection criteria based on amplitude, time, and color, we have identified 329 objects as known or candidate cataclysmic variables (CVs) during the first year of testing and operation of the Zwicky Transient Facility. Of these, 90 are previously confirmed CVs, 218 are strong candidates based on the shape and color of their light curves obtained during 3–562 days of observation, and the remaining 21 are possible CVs but with too few data points to be listed as good candidates. Almost half of the strong candidates are within 10 deg of the galactic plane, in contrast to most other large surveys that have avoided crowded fields. The available Gaia parallaxes are consistent with sampling the low mass transfer CVs, as predicted by population models. Our follow-up spectra have confirmed Balmer/helium emission lines in 27 objects, with four showing high-excitation He ii emission, including candidates for an AM CVn, a polar, and an intermediate polar. Our results demonstrate that a complete survey of the Galactic plane is needed to accomplish an accurate determination of the number of CVs existing in the Milky Way

    Characteristics of enrolment in an intensive home-visiting programme among eligible first-time adolescent mothers in England: a linked administrative data cohort study

    Background: Intensive home visiting for adolescent mothers may help reduce health disparities. Given limited resources, such interventions need to be effectively targeted. We evaluated which mothers were enrolled in the Family Nurse Partnership (FNP), an intensive home-visiting service for first-time young mothers commissioned in >130 local authorities in England since 2007. Methods: We created a population-based cohort of first-time mothers aged 13–19 years giving birth in English National Health Service hospitals between 1 April 2010 and 31 March 2017, using administrative hospital data linked with FNP programme, educational and social care data. Mothers living in a local authority with an active FNP site were eligible. We described variation in enrolment rates across sites, and identified maternal and FNP site characteristics associated with enrolment. Results: Of 110 520 eligible mothers, 25 680 (23.2% (95% CI: 23.0% to 23.5%)) were enrolled. Enrolment rates varied substantially across 122 sites (range: 11%–68%), and areas with greater numbers of first-time adolescent mothers achieved lower enrolment rates. Mothers aged 13–15 years were most likely to be enrolled (52%). However, only 26% of adolescent mothers with markers of vulnerability (including living in the most deprived areas and ever having been looked after as a child) were enrolled. Conclusion: A substantial proportion of first-time adolescent mothers with vulnerability markers were not enrolled in FNP. Variation in enrolment across sites indicates insufficient commissioning of places that is not proportional to level of need, with mothers in areas with large numbers of other adolescent mothers least likely to receive support

    Cataclysmic Variables in the Second Year of the Zwicky Transient Facility

    Using a filter in the GROWTH Marshal based on color and the amplitude and the timescale of variability, we have identified 372 objects as known or candidate cataclysmic variables (CVs) during the second year of operation of the Zwicky Transient Facility (ZTF). From the available difference imaging data, we found that 93 are previously confirmed CVs, and 279 are strong candidates. Spectra of four of the candidates confirm them as CVs by the presence of Balmer emission lines, while one of the four has prominent HeII lines indicative of containing a magnetic white dwarf. Gaia EDR3 parallaxes are available for 154 of these systems, resulting in distances from 108-2096 pc and absolute magnitudes in the range of 7.5-15.0, with the largest number of candidates between 10.5-12.5. The total numbers are 21% higher than from the previous year of the survey with a greater number of distances available but a smaller percentage of systems close to the Galactic plane. Comparison of these findings with a machine learning method of searching all the light curves reveals large differences in each dataset related to the parameters involved in the search process.Comment: Accepted in AJ, 24 pages, 2 tables, 7 figure
