    Leading-edge receptivity to a vortical freestream disturbance: A numerical analysis

    The receptivity to freestream vorticity of the boundary layer over a flat plate with an elliptic leading edge is investigated numerically. The flow is simulated by solving the incompressible Navier-Stokes system in general curvilinear coordinates with the vorticity and stream function as dependent variables. A finite-difference scheme which is second-order accurate in both space and time is used. As a first step, the steady basic-state solution is computed. Then a small amplitude vortical disturbance is introduced at the upstream boundary and the governing equations are solved time-accurately to evaluate the spatial and temporal growth of the perturbations leading to instability waves (Tollmien-Schlichting waves) inside the boundary layer. Preliminary results for a symmetric, 2-D disturbance reveal the presence of Tollmien-Schlichting waves aft of the flat-plate/ellipse juncture

    Effect of leading-edge geometry on boundary-layer receptivity to freestream sound

    The receptivity to freestream sound of the laminar boundary layer over a semi-infinite flat plate with an elliptic leading edge is simulated numerically. The incompressible flow past the flat plate is computed by solving the full Navier-Stokes equations in general curvilinear coordinates. A finite-difference method which is second-order accurate in space and time is used. Spatial and temporal developments of the Tollmien-Schlichting wave in the boundary layer, due to small-amplitude time-harmonic oscillations of the freestream velocity that closely simulate a sound wave travelling parallel to the plate, are observed. The effect of leading-edge curvature is studied by varying the aspect ratio of the ellipse. The boundary layer over the flat plate with a sharper leading edge is found to be less receptive. The relative contribution of the discontinuity in curvature at the ellipse-flat-plate juncture to receptivity is investigated by smoothing the juncture with a polynomial. Continuous curvature leads to less receptivity. A new geometry of the leading edge, a modified super ellipse, which provides continuous curvature at the juncture with the flat plate, is used to study the effect of continuous curvature and inherent pressure gradient on receptivity

    Numerical Studies of Boundary-Layer Receptivity

    Direct numerical simulations (DNS) of the acoustic receptivity process on a semi-infinite flat plate with a modified-super-elliptic (MSE) leading edge are performed. The incompressible Navier-Stokes equations are solved in stream-function/vorticity form in a general curvilinear coordinate system. The steady basic-state solution is found by solving the governing equations using an alternating direction implicit (ADI) procedure which takes advantage of the parallelism present in line-splitting techniques. Time-harmonic oscillations of the farfield velocity are applied as unsteady boundary conditions to the unsteady disturbance equations. An efficient time-harmonic scheme is used to produce the disturbance solutions. Buffer-zone techniques have been applied to eliminate wave reflection from the outflow boundary. The spatial evolution of Tollmien-Schlichting (T-S) waves is analyzed and compared with experiment and theory. The effects of nose-radius, frequency, Reynolds number, angle of attack, and amplitude of the acoustic wave are investigated. This work is being performed in conjunction with the experiments at the Arizona State University Unsteady Wind Tunnel under the direction of Professor William Saric. The simulations are of the same configuration and parameters used in the wind-tunnel experiments

Specifically, the crossflow instability and crossflow/Tollmien-Schlichting wave interactions are analyzed through the numerical solution of the full 3D Navier-Stokes equations including unsteadiness, curvature, and sweep. This approach is chosen because of the complexity of the problem and because it appears that linear stability theory is insufficient to explain the discrepancies between different experiments and between theory and experiment. The leading edge region of a swept wing is considered in a 3D spatial simulation with random disturbances as the initial conditions

The work has been closely coordinated with the experimental program of Professor William Saric, examining the same problem. Comparisons with NASA flight test data and the experiments at Arizona State University were a necessary and an important integral part of this work

    Knockdown of the DNA repair and redox signaling protein Ape1/ Ref-1 blocks ovarian cancer cell and tumor growth

    Apurinic endonuclease 1/redox effector factor-1 (Ape1/Ref-1 or Ape1) is an essential protein with two distinct functions. It is a DNA repair enzyme in the base excision repair (BER) pathway and a reduction–oxidation (redox) signaling factor maintaining transcription factors in an active reduced state. Our laboratory previously demonstrated that Ape1 is overexpressed in ovarian cancer and potentially contributes to resistance. Therefore, we utilized siRNA technology to knockdown protein levels of Ape1 in ovarian cancer cell line, SKOV-3x. Knocking Ape1 down had dramatic effects on cell growth in vitro but was not due to an increase in apoptosis and at least partially due to an extension in transit time through S-phase. Similarly, human ovarian tumor xenografts with reduced levels of Ape1 protein demonstrated a dramatic reduction in tumor volume (p < 0.01) and also statistically significant (p = 0.02) differences in 18F-fluorodeoxyglucose (FDG) uptake indicating reduced glucose metabolism and cellular proliferation. Ape1's role in DNA repair and redox signaling is important to our basic understanding of ovarian cancer cell growth and these findings strongly support Ape1 as a therapeutic target

    TIGIT Marks Exhausted T Cells, Correlates with Disease Progression, and Serves as a Target for Immune Restoration in HIV and SIV Infection.

    HIV infection induces phenotypic and functional changes to CD8+ T cells defined by the coordinated upregulation of a series of negative checkpoint receptors that eventually result in T cell exhaustion and failure to control viral replication. We report that effector CD8+ T cells during HIV infection in blood and SIV infection in lymphoid tissue exhibit higher levels of the negative checkpoint receptor TIGIT. Increased frequencies of TIGIT+ and TIGIT+ PD-1+ CD8+ T cells correlated with parameters of HIV and SIV disease progression. TIGIT remained elevated despite viral suppression in those with either pharmacological antiretroviral control or immunologically in elite controllers. HIV and SIV-specific CD8+ T cells were dysfunctional and expressed high levels of TIGIT and PD-1. Ex-vivo single or combinational antibody blockade of TIGIT and/or PD-L1 restored viral-specific CD8+ T cell effector responses. The frequency of TIGIT+ CD4+ T cells correlated with the CD4+ T cell total HIV DNA. These findings identify TIGIT as a novel marker of dysfunctional HIV-specific T cells and suggest TIGIT along with other checkpoint receptors may be novel curative HIV targets to reverse T cell exhaustion

    The development of a European fatal accident database

    A lack of representative European accident data to aid the development of safety policy, regulation and technological advancement is a major obstacle in the European Union. Data are needed to assess the performance of road and vehicle safety and is also needed to support the development of further actions by stakeholders. A recent analysis conducted by the European Transport Safety Council identified that there was no single system in place that could meet all of the needs and that there were major gaps including indepth crash causation information. This paper describes the process of developing a data collection and analysis system designed to partly fill these gaps. A project team with members from 7 countries was set up to devise appropriate variable lists to collect fatal crash data under the following topic levels: accident, road environment, vehicle, and road user, using retrospective detailed police reports (n=1,300). The typical level of detail recorded was a minimum of 150 variables for each accident. The project will enable multidisciplinary information on the circumstances of fatal crashes to be interpreted to provide information on a range of causal factors and events surrounding the collisions. This has major applications in the areas of active safety systems, infrastructure and road safety, as well as for tailoring behavioural interventions
