    In vitro cell culture of Charybdis congesta for enhanced production of secondary metabolites: Proscillaridin A, Scillaren A and Scilliroside

    Callus cultures of Charybdis congesta were initiated in vitro and the effect of growth regulators was tested on callus growth and secondary metabolite production. Among several standard media formulated for use in the present study, MS and B5 were found to be potentially active and facilitated the calculation of callus induction frequency (CIF). The CIF was higher in both MS (70%) and B5 (63%) media supplemented with 1-naphthalene acetic acid (NAA) (9.0 μM) and benzyl amino purine (BAP) (0.9 μM). However, with indole-3-butyric acid (IBA) (9.0 μM) and BAP (0.8 μM), less amount (22.6%) of CIF was observed in MS medium but no callus formation was noticed in B5 medium. Rapid high performance thin layer chromatography (HPTLC) screening of callus extracts revealed that the callus established in MS medium supplemented with 4.5 μM NAA and 0.46 μM BAP produced the highest yield of Proscillaridin A (4.51 mg/g DW), Scilliroside (3.3 mg/g DW), Scillaren A (2.35 mg/g DW) and desacetylscilliroside (8.62 mg/g DW), which was higher than from the intact plants. The results obtained indicate that the in vitro cultures of C. congesta might be an excellent source of secondary metabolites and further metabolic profiling may provide insights into up scaling of the compounds which lead to greater commercial interest and continuous supply of cultures.Keywords: Squill, Charybdis congesta, bufadienolides, callus cultures, reflectance scanning densitometryAfrican Journal of Biotechnology Vol. 12(15), pp. 1754-17

    Benzyl Isothiocyanate Suppresses Pancreatic Tumor Angiogenesis and Invasion by Inhibiting HIF-α/VEGF/Rho-GTPases: Pivotal Role of STAT-3

    Our previous studies have shown that benzyl isothiocyanate (BITC) suppresses pancreatic tumor growth by inhibiting STAT-3; however, the exact mechanism of tumor growth suppression was not clear. Here we evaluated the effects and mechanism of BITC on pancreatic tumor angiogenesis. Our results reveal that BITC significantly inhibits neovasularization on rat aorta and Chicken-Chorioallantoic membrane. Furthermore, BITC blocks the migration and invasion of BxPC-3 and PanC-1 pancreatic cancer cells in a dose dependant manner. Moreover, secretion of VEGF and MMP-2 in normoxic and hypoxic BxPC-3 and PanC-1 cells was significantly suppressed by BITC. Both VEGF and MMP-2 play a critical role in angiogenesis and metastasis. Our results reveal that BITC significantly suppresses the phosphorylation of VEGFR-2 (Tyr-1175), and expression of HIF-α. Rho-GTPases, which are regulated by VEGF play a crucial role in pancreatic cancer progression. BITC treatment reduced the expression of RhoC whereas up-regulated the expression of tumor suppressor RhoB. STAT-3 over-expression or IL-6 treatment significantly induced HIF-1α and VEGF expression; however, BITC substantially suppressed STAT-3 as well as STAT-3-induced HIF-1α and VEGF expression. Finally, in vivo tumor growth and matrigel-plug assay show reduced tumor growth and substantial reduction of hemoglobin content in the matrigel plugs and tumors of mice treated orally with 12 µmol BITC, indicating reduced tumor angiogenesis. Immunoblotting of BITC treated tumors show reduced expression of STAT-3 phosphorylation (Tyr-705), HIF-α, VEGFR-2, VEGF, MMP-2, CD31 and RhoC. Taken together, our results suggest that BITC suppresses pancreatic tumor growth by inhibiting tumor angiogenesis through STAT-3-dependant pathway

    IN VITRO HEPATOPROTECTIVE EFFECT OF ECHINOCHLOA COLONA ON ETHANOL-INDUCED OXIDATIVE DAMAGE IN HEPG2 CELLS

      Objective: The current study was aimed to evaluate the methanolic extract of caryopses of Echinochloa colona (ECME) for its in vitro hepatoprotective activity against ethanol in HepG2 cell lines.Methods: In this regard, the cytotoxicity studies were conducted for the extract, ECME using 3-(4,5-dimethythiazol- 2-yl)-2,5-diphenyl tetrazolium bromide assay to determine the inhibitory concentration 50% value based on which, the doses 50, 100, and 200 μg/ml were selected for the hepatoprotective studies in HepG2 cell lines. The toxicity was induced using ethanol (100 mM). The in vitro hepatoprotective activity of the extract was assessed based on the changes in the level of biochemical parameters such as aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase.Results: The extract, ECME has shown a dose-dependent cytoprotective activity with maximum protection at 200 μg/ml. The percentage cell viability of the extract, ECME at 200 μg/ml was more, i.e., 69.33% which was well comparable to that of standard drug, silymarin (100 μg/ml).Conclusion: The study revealed that the extract had shown significant hepatoprotective activity at all the test doses against ethanol-induced cytotoxity assay

    Application of MFFC and Edge Detection for Remote Driven Vehicles Through Matlab

    Speech recognition is a rapidly emerging technology in Human Computer Interaction HCI. It has many applications as we use it from search engines to the device control it serves many areas we interact every day from dawn to dusk. Along with the uses we have many limitations in speech processing such as Language barrier, Accent and Noise, so to implement the speech processing we have many challenges. To enable the advantages of this speech processing most of the leading software companies like Apple, Microsoft and Google are continuously evolving their speech enabled applications. The speech processing eases the physically challenged people's interaction with the devices and makes them productive. The Idea of the automatically driven cars is introduced by Google and Audi, but they are not acceptable in most of the cases because of lacking trust in current technology. Thus we here worked on the remote driven vehicle in a more secured method using the Mel Frequency cepstral Coefficient. The secure driving of the vehicle can be ensured by the remote driver. This technique is very rapid and more reliable for the speech detection thus the remote driver can use the MFCC and the video from the vehicle needed to be broadcasted to the remote driver that can be done an IP camera running on a data network. And the instructions can from the remote driver can be sent to the vehicle by an app created with python that connected to a micro controller. To minimize the limitations in the remote drive the vehicle must be enabled with automatic braking when obstacle approaches which can be done by the Ultrasonic sensors that do the distance estimation. The remote drivers usually have a very limited view of road they drive and they must get the precise edges of the road this can be achieved by processing the stream of images to calculate the edge in Matlab

    A rare case of indirect inguinal hernia with ovary as content at a hospital of Visakhapatnam

    A hernia is the protrusion of the small intestines or omentum or other organs through a defect in the abdominal wall. Inguinal herniain females is very rare and occurs in <5% of women. Here, we report the case of a 25-year-old female who presented with swellingin the left groin associated with dragging pain in the left iliac fossa. She was investigated and diagnosed as a left indirect inguinalhernia. Hernioplasty was planned and intraoperatively ovary along with fimbria was identified as content. An inguinal ovary mayoccur if the gubernaculum fails to attach to the uterus in fetal life or if the canal of Nuck remains open after birth

    Effect of feeding maize fiber in wet, dry and silage form with cotton cake supplementation on intake, nutrient utilization and performance in Nellore Brown sheep

    Maize fiber was evaluated in wet, dry and silage form with 200 g cotton cake supplementation in growing Nellore Brown ram lambs (24.8±0.96) using six sheep per treatment in a growth-cummetabolism trial of 90 days with collection of feed, leftover, feces and urine samples during the last ten days. Average daily gain (g), nutrient digestibility (OM, CP, NDF, ADF) tended to be higher (P = 0.07 to 0.09) and intake of OM, DOM, CP (gld) and ME (MJ!d) and nitrogen retention were significantly (P = 0. 0002 to 0. 002) higher in lambs when fed maize fiber in silage rather than in wet and dry form. Depending on input such as labor required ensilaging or drying of maize fiber seems an economically more beneficial and from a food security point of view a safer way than feeding wet maize fiber

    Comparative Analysis of common Edge Detection Algorithms using Pre-processing Technique

    Edge detection is the process of segmenting an image by detecting discontinuities in brightness. So far, several standard segmentation methods have been widely used for edge detection. However, due to inherent quality of images, these methods prove ineffective if they are applied without any preprocessing. In this paper, an image preprocessing approach has been adopted in order to get certain parameters that are useful to perform better edge detection with the standard edge detection methods. The proposed preprocessing approach involves median filtering to reduce the noise in image and then Edge Detection technique is carried out. And atlast Standard edge detection methods can be applied to the resultant preprocessing image and its Simulation results are show that our preprocessed approach when used with a standard edge detection method enhances its performance
