    Discovery of a Visual T-Dwarf Triple System and Binarity at the L/T Transition

    We present new high contrast imaging of 8 L/T transition brown dwarfs using the NIRC2 camera on the Keck II telescope. One of our targets, the T3.5 dwarf 2MASS J08381155 + 1511155, was resolved into a hierarchal triple with projected separations of 2.5+/-0.5 AU and 27+/-5 AU for the BC and A(BC) components respectively. Resolved OSIRIS spectroscopy of the A(BC) components confirm that all system members are T dwarfs. The system therefore constitutes the first triple T-dwarf system ever reported. Using resolved photometry to model the integrated-light spectrum, we infer spectral types of T3, T3, and T4.5 for the A, B, and C components respectively. The uniformly brighter primary has a bluer J-Ks color than the next faintest component, which may reflect a sensitive dependence of the L/T transition temperature on gravity, or alternatively divergent cloud properties amongst components. Relying on empirical trends and evolutionary models we infer a total system mass of 0.034-0.104 Msun for the BC components at ages of 0.3-3 Gyr, which would imply a period of 12-21 yr assuming the system semi-major axis to be similar to its projection. We also infer differences in effective temperatures and surface gravities between components of no more than ~150 K and ~0.1 dex. Given the similar physical properties of the components, the 2M0838+15 system provides a controlled sample for constraining the relative roles of effective temperature, surface gravity, and dust clouds in the poorly understood L/T transition regime. Combining our imaging survey results with previous work we find an observed binary fraction of 4/18 or 22_{-8}^{+10}% for unresolved spectral types of L9-T4 at separations >~0.1 arcsec. This translates into a volume-corrected frequency of 13^{-6}_{+7}%, which is similar to values of ~9-12% reported outside the transition.     While the green building movement is common practice in the commercial realm, it is not yet widely popular with residential buildings. We considered the question “How can residents of western Montana adopt eco-friendly housing practices that are locally appropriate and relevant?” There is an opportunity to increase green living practices among renters and homeowners in western Montana through retrofitting, gardening, composting, and similar behaviors. By considering climatic factors relevant to the region, suggestions for relevant eco-friendly practices can be made available to homeowners and renters alike. We will research green living practices used in other countries with similar climatic factors as western Montana. We will then make a website to help streamline locally relevant information catered to help residents take action towards their sustainability goals. We will survey a sample of residents throughout western Montana to inform the materials provided on the website. For example, these materials may include, but are not limited to, sustainability project demonstration videos, links to local builders, history, blogs, global initiatives, and links to other resources. We expect this website to be relevant and increase accessibility to western Montana renters and homeowners

    Design a small modular reactor that is easily transportable for use in disaster relief as well as remote military outposts. A rail shippable reactor gives quick and easy transportation from one part of a country to another. The reactor must have a three MWe production capacity to ensure the reactor has the performance to power larger government facilities, such as hospitals and water treatment plants. The reactor must have enough fuel for a six-month minimum fuel cycle. Atmospheric cooling only provides the ability to reject heat to the atmosphere, minimizing the weight requirements. Uranium fuel will have a maximum of 19.75% enrichment, to minimize proliferation concerns with the reactor

    Molecular architecture of softwood revealed by solid-state NMR

    Economically important softwood from conifers is mainly composed of the polysaccharides cellulose, galactoglucomannan and xylan, and the phenolic polymer, lignin. The interactions between these polymers lead to wood mechanical strength and must be overcome in biorefining. Here, we use 13C multidimensional solid-state NMR to analyse the polymer interactions in never-dried cell walls of the softwood, spruce. In contrast to some earlier softwood cell wall models, most of the xylan binds to cellulose in the two-fold screw conformation. Moreover, galactoglucomannan alters its conformation by intimately binding to the surface of cellulose microfibrils in a semi-crystalline fashion. Some galactoglucomannan and xylan bind to the same cellulose microfibrils, and lignin is associated with both of these cellulose-bound polysaccharides. We propose a model of softwood molecular architecture which explains the origin of the different cellulose environments observed in the NMR experiments. Our model will assist strategies for improving wood usage in a sustainable bioeconomy

    Rapid detection, cloning and molecular cytogenetic characterisation of sequences from an MRP-encoding amplicon by chromosome microdissection.

    Chromosome microdissection was utilised for the analysis of cytogenetic markers of gene amplification [homogeneously staining regions (hsrs) and double minutes (dmins)] in two doxorubicin-resistant cell lines, fibrosarcoma HT1080/DR4 and small-cell lung cancer H69AR. Microdissection products from the hsr(7)(p12p15) of HT1080/DR4 were amplified and used for fluorescent in situ hybridisation (micro-FISH) analysis of drug-sensitive HT1080, resistant HT1080/DR4 and normal lymphocytes. The results demonstrated that the hsr contains a domain of DNA amplification of complex origin including sequences derived from 16p11.2-16p13.1, 2q11.2, 7q32-7q34 and 10q22. The amplification was confirmed by converting the micro-dissected probe into a microclone library for probing HT1080 and HT1080/DR4 Southerns. A micro-FISH probe from normal band region 16p11-16p13 further demonstrated amplification of 16p sequences in both HT1080/DR4 and H69AR. During the course of this analysis, Cole et al. (1992) (Science, 258, 1650-1653) published the amplification of the MRP gene in H69AR cells, which maps to chromosome 16p13.1. Our results corroborate the finding of MRP amplification in these doxorubicin-resistant cell lines, but, importantly, they provide information on the composition of the complex amplicon contributions from four different chromosomes. This study demonstrates the potential utility of chromosome microdissection for the rapid recovery of sequences from amplified regions in drug-resistant cells

    Hydroxybiphenylamide GroEL/ES Inhibitors Are Potent Antibacterials against Planktonic and Biofilm Forms of Staphylococcus aureus

    We recently reported the identification of a GroEL/ES inhibitor (1, N-(4-(benzo[d]thiazol-2-ylthio)-3-chlorophenyl)-3,5-dibromo-2-hydroxybenzamide) that exhibited in vitro antibacterial effects against Staphylococcus aureus comparable to vancomycin, an antibiotic of last resort. To follow up, we have synthesized 43 compound 1 analogs to determine the most effective functional groups of the scaffold for inhibiting GroEL/ES and killing bacteria. Our results identified that the benzothiazole and hydroxyl groups are important for inhibiting GroEL/ES-mediated folding functions, with the hydroxyl essential for antibacterial effects. Several analogs exhibited >50-fold selectivity indices between antibacterial efficacy and cytotoxicity to human liver and kidney cells in cell culture. We found that MRSA was not able to easily generate acute resistance to lead inhibitors in a gain-of-resistance assay and that lead inhibitors were able to permeate through established S. aureus biofilms and maintain their bactericidal effects
