    OBJECTIVE: to analyze alcohol consumption among adolescents of public schools in the city of Recife-PE.METHOD: it is a descriptive, exploratory, quantitative approach, developed in three state public schools in the city of Recife-PE, with the population composed of schoolchildren of both sexes, between 12 and 17 years of age.RESULTS: the study identified risk factors that influence alcohol consumption among schoolchildren interviewed, highlighting the variables age, school disapproval and number of people with whom.CONCLUSION: it is suggested that there be strengthened health education practices through partnerships between the school and the local Family Health Unit through the School Health Program (PSE), which stimulate health-friendly practices.OBJETIVO: analizar el consumo de alcohol entre adolescentes de las escuelas públicas en la ciudad de Recife-PE.MÉTODO: se trata de una pesquisa descriptiva, exploratoria de abordaje cuantitativo, desarrollada en tres escuelas públicas estatales de la ciudad de Recife-PE, con la población compuesta por escolares de ambos sexos, en el grupo de edad entre 12 a 17 años.RESULTADOS: el estudio identificó factores de riesgo que influencian el consumo de alcohol entre los escolares entrevistados, destacándose las variables edad, reprobación escolar y número de personas con quienes.CONCLUSIÓN: se sugiere que haya el fortalecimiento de prácticas de educación en salud a través de alianzas entre escuela y Unidad de Salud de la Familia local a través del Programa de Salud en la Escuela (PSE), que estimulan prácticas favorables a la salud.OBJETIVO: analisar o consumo de álcool entre adolescentes das escolas públicas na cidade do Recife-PE.MÉTODO: trata-se de uma pesquissa descritiva, exploratória de abordagem quantitativa, desenvolvida em três escolas públicas estaduais da cidade do Recife-PE, com a população composta por escolares de ambos os sexos, na faixa etária entre 12 a 17 anos.RESULTADOS: o estudo identificou fatores de risco que influenciam o consumo de álcool entre os escolares entrevistados, destacando-se as variáveis idade, reprovação escolar e número de pessoas com quem mora.CONCLUSÃO: sugere-se que haja o fortalecimento de práticas de educação em saúde através de parcerias entre escola e Unidade de Saúde da Família local por meio do Programa de Saúde na Escola (PSE), que estimulem práticas favoráveis à saúde

    A Controladoria e Ouvidoria Municipal do Crato é um órgão instituído através de lei municipal e considerado de staff, pois no organograma da entidade se encontra logo após o gestor, e tem autonomia para desempenhar suas atividades. O objetivo geral deste estudo foi analisar a fase de institucionalização em que se encontra a controladoria no âmbito município do Crato (CE).  A metodologia tem abordagem qualitativa e a pesquisa se enquadra como bibliográfica e de levantamento. Para obtenção dos dados foi aplicado um questionário ao controlador do respectivo município. Dentre as atividades desempenhadas pela controladoria estão: planejar e otimizar os sistemas de informações; realizar uuditorias; executar a atividade de ouvidoria; orientar os gestores em processos administrativos e elaborar a prestação de contas do chefe do Executivo. A mesma não desempenha a atividade de orientação do orçamento, nem o assessoramento no planejamento e execução das atividades da contabilidade. Contudo tem otimizado os resultados da prefeitura como um todo. 

    Purification of a lectin from Cratylia mollis crude extract seed by a single step PEG/phosphate aqueous two-phase system

    The partitioning and purification of lectins from the crude extract of Cratylia mollis seeds (Cramoll 1,4) was investigated in aqueous two-phase systems (ATPS). A factorial design model (24) was used to evaluate the influence of polyethylene glycol (PEG) molar mass (15008000g/mol), PEG concentration (12.517.5% w/w), phosphate (1015% w/w) concentration, and pH (68) on the differential partitioning, purification factor, and yield of the lectin. Polymer and salt concentration were the most important variables affecting partition of lectin and used to find optimum purification factor by experimental BoxBehnken design together with the response surface methodology (RSM). ATPS showed best conditions composed by 13.9% PEG1500, 15.3% phosphate buffer at pH 6, which ensured purification factor of 4.70. Sodium dodecyl sulfatepolyacrylamide gel electrophoresis showed a single band of protein with 26.1kDa. Furthermore, results demonstrated a thermostable lectin presenting activity until 60°C and lost hemagglutinating activity at 80°C.     Screening, production and biochemical characterization of a new fibrinolytic enzyme produced by Streptomyces sp. (Streptomycetaceae) isolated from Amazonian lichens

    Thrombosis is a pathophysiological disorder caused by accumulation of fibrin in the blood. Fibrinolytic proteases with potent thrombolytic activity have been produced by diverse microbial sources. Considering the microbial biodiversity of the Amazon region, this study aimed at the screening, production and biochemical characterization of a fibrinolytic enzyme produced by Streptomyces sp. isolated from Amazonian lichens. The strain Streptomyces DPUA1576 showed the highest fibrinolytic activity, which was 283 mm2. Three variables at two levels were used to assess their effects on the fibrinolytic production. The parameters studied were agitation (0.28 - 1.12 g), temperature (28 - 36 ºC) and pH (6.0 - 8.0); all of them had significant effects on the fibrinolytic production. The maximum fibrinolytic activity (304 mm2) was observed at 1.12 g, 28 ºC, and pH of 8.0. The crude extract of the fermentation broth was used to assess the biochemical properties of the enzyme. Protease and fibrinolytic activities were stable during 6 h, at a pH ranging from 6.8 to 8.4 and 5.8 to 9.2, respectively. Optimum temperature for protease activity ranged between 35 and 55 °C, while the highest fibrinolytic activity was observed at 45 ºC. Proteolytic activity was inhibited by Cu2+ and Co2+ ions, phenylmethylsulfonyl fluoride (PMSF) and pepstatin A, which suggests that the enzyme is a serine protease. Enzymatic extract cleaved fibrinogen at the subunits A-chain, A-chain, and -chain. The results indicated that Streptomyces sp.     Rationale, study design, and analysis plan of the Alveolar Recruitment for ARDS Trial (ART): Study protocol for a randomized controlled trial

    Background: Acute respiratory distress syndrome (ARDS) is associated with high in-hospital mortality. Alveolar recruitment followed by ventilation at optimal titrated PEEP may reduce ventilator-induced lung injury and improve oxygenation in patients with ARDS, but the effects on mortality and other clinical outcomes remain unknown. This article reports the rationale, study design, and analysis plan of the Alveolar Recruitment for ARDS Trial (ART). Methods/Design: ART is a pragmatic, multicenter, randomized (concealed), controlled trial, which aims to determine if maximum stepwise alveolar recruitment associated with PEEP titration is able to increase 28-day survival in patients with ARDS compared to conventional treatment (ARDSNet strategy). We will enroll adult patients with ARDS of less than 72 h duration. The intervention group will receive an alveolar recruitment maneuver, with stepwise increases of PEEP achieving 45 cmH(2)O and peak pressure of 60 cmH2O, followed by ventilation with optimal PEEP titrated according to the static compliance of the respiratory system. In the control group, mechanical ventilation will follow a conventional protocol (ARDSNet). In both groups, we will use controlled volume mode with low tidal volumes (4 to 6 mL/kg of predicted body weight) and targeting plateau pressure <= 30 cmH2O. The primary outcome is 28-day survival, and the secondary outcomes are: length of ICU stay; length of hospital stay; pneumothorax requiring chest tube during first 7 days; barotrauma during first 7 days; mechanical ventilation-free days from days 1 to 28; ICU, in-hospital, and 6-month survival. ART is an event-guided trial planned to last until 520 events (deaths within 28 days) are observed. These events allow detection of a hazard ratio of 0.75, with 90% power and two-tailed type I error of 5%. All analysis will follow the intention-to-treat principle. Discussion: If the ART strategy with maximum recruitment and PEEP titration improves 28-day survival, this will represent a notable advance to the care of ARDS patients.     Abstract The great atherogenic potential of oxidized lowdensity lipoprotein has been widely described in the literature. The objective of this study was to investigate whether the state of oxidized low-density lipoprotein in human plasma measured by the Z-scan technique has an association with different cardiometabolic biomarkers. Total cholesterol, highdensity lipoprotein cholesterol, triacylglycerols, apolipoprotein A-I and apolipoprotein B, paraoxonase-1, and glucose were analyzed using standard commercial kits, and lowdensity lipoprotein cholesterol was estimated using the Friedewald equation. A sandwich enzyme-linked immunosorbent assay was used to detect electronegative low-density lipoprotein. Low-density lipoprotein and high-density lipoprotein sizes were determined by Lipoprint ® system. The Z-scan technique was used to measure the non-linear optical response of low-density lipoprotein solution. Principal component analysis and correlations were used respectively to resize the data from the sample and test association between the θ parameter, measured with the Z-scan technique, and the principal component. A total of 63 individuals, from both sexes, with mean age 52 years (±11), being overweight and having high levels of total cholesterol and low levels of high-density lipoprotein cholesterol, were enrolled in this study. A positive correlation between the θ parameter and more anti-atherogenic pattern for cardiometabolic biomarkers together with a negative correlation for an atherogenic pattern was found. Regarding the parameters related with an atherogenic low-density lipoprotein profile, the θ parameter was negatively correlated with a more atherogenic pattern. By using Z-scan measurements, we were able to find an association between oxidized low-density lipoprotein state and multiple cardiometabolic biomarkers in samples from individuals with different cardiovascular risk factors
