Charakterisierung eines Typ III-Sekretionssystems fĂŒr Virulenzproteine aus Salmonella typhimurium
Zur Strukturanalyse des Typ III-Sekretionssystems, kodiert von der Salmonella-
PathogenitÀtsinsel 2, wurden polyklonale Antikörper gegen rekombinante Proteine des
Sekretionsapparates erzeugt. Zur Entfernung unspezifischer Bindungen wurden die
polyklonalen Antiseren an eine CNBr-aktivierte Sepharose 4B, gekoppelt mit Salmonella-
Lysaten, prĂ€adsorbiert. AnschlieĂend konnten die Antiseren fĂŒr verschiedene
Immunoblotanalysen eingesetzt werden.
Durch eine Membranfraktionierung mit N-Lauroyl-Sarcosin konnte die subzellulÀre
Lokalisation der Apparatsproteine SsaC, SsaN, SsaV und SsaB bestimmt werden. Dabei
wurde SsaC in der Ă€uĂeren, SsaN und SsaV in der inneren Membranfraktion
nachgewiesen. SsaB war nur in der löslichen Fraktion detektierbar. Um mögliche
Proteinkomplexe innerhalb des Sekretionsapparates zu identifizieren, wurde der
membranpermeable Quervernetzer DSS eingesetzt. So konnten fĂŒr das Sekretin-bildende
Protein SsaC und fĂŒr das putative Translokatorprotein SseD oligomere Strukturen
nachgewiesen werden.
Bei der Untersuchung der Effekte von Mutationen in verschiedenen SPI2-Genen auf die
Synthese einzelner Proteine des Sekretionsapparates wurde deutlich, dass das Zwei-
Komponenten-Regulationssystem SsrAB sowie alle untersuchten Apparatsproteine eine
essentielle Rolle in der Synthese bzw. im Aufbau des Sekretionsapparates spielen.
Lediglich eine Mutation in ssaV oder ssaG hatte einen weniger starken Effekt auf die
Synthese der Apparatsproteine, so dass SsaC in Wildtyp-Mengen detektiert werden konnte.
DarĂŒber hinaus wurde die Synthese einzelner Proteine des Sekretionsapparates auch in
AbhÀngigkeit des pH-Wertes analysiert. Nach einer Absenkung des pH-Wertes auf pH 5,8
nahmen die Proteinmengen weitaus stÀrker zu als ohne pH-Wert-VerÀnderung. Dabei
konnte nachgewiesen werden, dass dieser Unterschied in den synthetisierten
Proteinmengen, in AbhÀngigkeit des pH-Wertes, nicht auf eine verstÀrkte Transkription
zurĂŒckzufĂŒhren ist und dass es sich um einen SPI2-spezifischen Effekt handelt.
Die Kinetik des Aufbaus des Sekretionsapparates wurde anhand der Sekretion des
putativen Translokatorproteins SseB bestimmt. 1 h nach der Absenkung des pH-Wertes auf
pH 5,8 wurde eine intrazellulÀre Akkumulation von SseB detektiert. Zu diesem Zeitpunkt
konnte kein SseB auf der ZelloberflÀche lokalisiert werden. Eine Sekretion von SseB
wurde erst 3 h nach der SĂ€ure-Induktion beobachtet. Demzufolge muss der
Sekretionsapparat in diesem Zeitraum vollstÀndig aufgebaut und funktional intakt sein, um
die pH-induzierte Sekretion zu ermöglichen.
Das SPI2-Protein SsaB (SpiC) ist fĂŒr die Sekretion der putativen Translokatorproteine
SseB und SseC erforderlich. So könnte SsaB eine Komponente des Typ IIISekretionsapparates
darstellen oder die Funktion eines Chaperones fĂŒr die
Effektorproteine, bzw. eines GSP-spezifischen Chaperones fĂŒr das Sekretin-bildende
Protein SsaC besitzen. Ebenso wĂ€re es möglich, dass SsaB fĂŒr die korrekte Lokalisation
von SsaC in der Ă€uĂeren Membran verantwortlich ist.
Aus S. typhimurium konnten die als Nadel-Komplexe bezeichneten makromolekularen
Strukturen, die den gesamten Sekretionsapparat darstellen, isoliert aber nicht eindeutig
identifiziert werden. Da in einer SPI1-Mutante, die einen Defekt im Typ IIISekretionsapparat
hat, keine Nadel-Komplexe mehr angereichert werden konnten, liegt die
Vermutung nahe, dass es sich bei den Nadel-Komplexen, isoliert aus S. typhimurium-
Wildtyp, um die SPI1-Strukturen handelt
"Spatial Joint Models through Bayesian Structured Piece-wise Additive Joint Modelling for Longitudinal and Time-to-Event Data"
Joint models for longitudinal and time-to-event data have seen many
developments in recent years. Though spatial joint models are still rare and
the traditional proportional hazards formulation of the time-to-event part of
the model is accompanied by computational challenges. We propose a joint model
with a piece-wise exponential formulation of the hazard using the counting
process representation of a hazard and structured additive predictors able to
estimate (non-)linear, spatial and random effects. Its capabilities are
assessed in a simulation study comparing our approach to an established one and
highlighted by an example on physical functioning after cardiovascular events
from the German Ageing Survey. The Structured Piecewise Additive Joint Model
yielded good estimation performance, also and especially in spatial effects,
while being double as fast as the chosen benchmark approach and performing
stable in imbalanced data setting with few events
COMPLEMENTARY USE OF DESIGN METHODOLOGIES IN EDUCATIONAL ENGINEERING DESIGN PROJECTS: A CASE STUDY
The challenge of a nowadays well-grounded engineering education is preparing industry ready graduates, that is, to provide students with the skills to master the complexity of products in terms of innovation, invention and problem solving combined with soft skills abilities. This addresses particularly the engineering design education, which in general should be mainly based on practical studies represented by engineering design projects. In order to supply industry with work-ready practitioners, the effectiveness and efficacy of design education respectively design projects is a crucial aspect. This article presents some findings of an efficient use of design methodologies and techniques when carrying through design projects in the faculty of Mechanical Engineering and Mechatronics at the University of Applied Sciences, Deggendorf, Germany. Engineering design projects are compulsory for students within their curriculum. The projects aim at fostering and stimulating students creativity by means of a complementary use of design methodologies and techniques. By taking advantage of this approach the systematic generation of ideas in the course of a design project is presented, which eventually led to the development of an innovative product
Sharp lines in the absorption edge of EuTe and PbEuTe in high magnetic fields
The optical absorption spectra in the region of the \fd transition energies
of epitaxial layers of of EuTe and \PbEuTe, grown by molecular beam epitaxy,
were studied using circularly polarized light, in the Faraday configuration.
Under \sigmam polarization a sharp symmetric absorption line (full width at
half-maximum 0.041 eV) emerges at the low energy side of the band-edge
absorption, for magnetic fields intensities greater than 6 T. The absorption
line shows a huge red shift (35 meV/T) with increasing magnetic fields. The
peak position of the absorption line as a function of magnetic field is
dominated by the {\em d-f} exchange interaction of the excited electron and the
\Euion spins in the lattice. The {\em d-f} exchange interaction energy was
estimated to be eV. In \PbEuTe the same absorption line
is detected, but it is broader, due to alloy disorder, indicating that the
excitation is localized within a finite radius. From a comparison of the
absorption spectra in EuTe and \PbEuTe the characteristic radius of the
excitation is estimated to be \AA.Comment: Journal of Physics: Condensed Matter (2004, at press
Magnetic Ordering Of Eutepbte Multilayers Determined By X-ray Resonant Diffraction
In this work we use resonant x-ray diffraction combined with polarization analysis of the diffracted beam to study the magnetic ordering in EuTePbTe multilayers. The presence of satellites at the (1/2 1/2 1/2) magnetic reflection of a 50 repetition EuTePbTe superlattice demonstrated the existence of magnetic correlations among the alternated EuTe layers.
