Neuropilin-1 Controls Endothelial Homeostasis by Regulating Mitochondrial Function and Iron-Dependent Oxidative Stress.
The transmembrane protein neuropilin-1 (NRP1) promotes vascular endothelial growth factor (VEGF) and extracellular matrix signaling in endothelial cells (ECs). Although it is established that NRP1 is essential for angiogenesis, little is known about its role in EC homeostasis. Here, we report that NRP1 promotes mitochondrial function in ECs by preventing iron accumulation and iron-induced oxidative stress through a VEGF-independent mechanism in non-angiogenic ECs. Furthermore, NRP1-deficient ECs have reduced growth and show the hallmarks of cellular senescence. We show that a subcellular pool of NRP1 localizes in mitochondria and interacts with the mitochondrial transporter ATP-binding cassette B8 (ABCB8). NRP1 loss reduces ABCB8 levels, resulting in iron accumulation, iron-induced mitochondrial superoxide production, and iron-dependent EC senescence. Treatment of NRP1-deficient ECs with the mitochondria-targeted antioxidant compound mitoTEMPO or with the iron chelator deferoxamine restores mitochondrial activity, inhibits superoxide production, and protects from cellular senescence. This finding identifies an unexpected role of NRP1 in EC homeostasis
Genome-wide signatures of population bottlenecks and diversifying selection in European wolves
Genomic resources developed for domesticated species provide powerful tools for studying the evolutionary history of their wild relatives. Here we use 61K single-nucleotide polymorphisms (SNPs) evenly spaced throughout the canine nuclear genome to analyse evolutionary relationships among the three largest European populations of grey wolves in comparison with other populations worldwide, and investigate genome-wide effects of demographic bottlenecks and signatures of selection. European wolves have a discontinuous range, with large and connected populations in Eastern Europe and relatively smaller, isolated populations in Italy and the Iberian Peninsula. Our results suggest a continuous decline in wolf numbers in Europe since the Late Pleistocene, and long-term isolation and bottlenecks in the Italian and Iberian populations following their divergence from the Eastern European population. The Italian and Iberian populations have low genetic variability and high linkage disequilibrium, but relatively few autozygous segments across the genome. This last characteristic clearly distinguishes them from populations that underwent recent drastic demographic declines or founder events, and implies long-term bottlenecks in these two populations. Although genetic drift due to spatial isolation and bottlenecks seems to be a major evolutionary force diversifying the European populations, we detected 35 loci that are putatively under diversifying selection. Two of these loci flank the canine platelet-derived growth factor gene, which affects bone growth and may influence differences in body size between wolf populations. This study demonstrates the power of population genomics for identifying genetic signals of demographic bottlenecks and detecting signatures of directional selection in bottlenecked populations, despite their low background variability.Heredity advance online publication, 18 December 2013; doi:10.1038/hdy.2013.122
Terminal Penumpang Pelabuhan Umum Di Kolonodale (Ekspresi Budaya Mori âTepo Asa Aroa' Dalam Arsitektur)
Dalam suatu perkembangan kota di Indonesia terutama dalam sektor laut, sangatlah penting. Mengingat Indonesia adalah Negara Maritim yang otomatis mempunyai kekayaan alam di bidang kelautan. Dengan melonjaknya arus penumpang masyarakat terutama dalam hal transpotasi laut, sangatlah dibutuhkan akses kapal-kapal laut baik yang berskala besar maupun kecil. Oleh karena itu, dalam prospek perancangan di ambil suatu objek Terminal Penumpang Pelabuhan Umum di Kolonodale. Yang merupakan terminal penumpang dalam hal ini masih belum memenuhi kriteria terminal penumpang pada umunya dengan standar kepelabuhanan di Indonesia. Adapun pendekatan tema perancangan pada objek ini yaitu, Ekspresi Budaya Mori âTepo Asa Aroa' dalam Arsitektur. Tentunya sangat membantu dalam membuat gagasan-gagasan sketsa ide. Didalamnya akan diterapkan berbagai macam kosnep-konsep nilai budaya, norma-norma, dan perilaku dari masyarakat suku Mori

Numerical investigation of oil injection in a Roots blower operated as expander
The adoption of positive displacement machines as expanders in Organic Rankine Cycles (ORCs) is increasingly common, especially in the low to medium power range. At the same time, these devices often serve as compressor in Vapor-Compression Refrigeration Systems. In both cases, the application of Computation Fluid Dynamics (CFD) to optimize such machines has become an integrated tool in the design process. As a consequence, several challenges associated with the numerical simulation have to be taken into account. For example, the modeling of the gap represents a challenge for the stability of the numerical analysis. The dynamic of the process, combined with deformations of the clearances and of the working chamber has to be considered with extra care. To raise the efficiency of the machine, oil is typically injected. Its numerical modeling imply an extra challenge in the simulation of the actual operation of the machine. The present work is mainly focused on the multi-phase nature of the flow, with a particular analysis of the lubricant oil injected. In this work, a two-lobe Roots blower operated as expander has been simulated with the open-source software OpenFOAM-v1812, using the SCORG-V5.2.2. This analysis highlights the areas that are affected the most by the oil presence in order to highlight the sealing effect it provides
A reversible <i>Renilla </i>luciferase protein complementation assay for rapid identification of protein-protein interactions reveals the existence of an interaction network involved in xyloglucan biosynthesis in the plant Golgi apparatus
A growing body of evidence suggests that proteinâprotein interactions (PPIs) occur amongst glycosyltransferases (GTs) required for plant glycan biosynthesis (e.g. cell wall polysaccharides and N-glycans) in the Golgi apparatus, and may control the functions of these enzymes. However, identification of PPIs in the endomembrane system in a relatively fast and simple fashion is technically challenging, hampering the progress in understanding the functional coordination of the enzymes in Golgi glycan biosynthesis. To solve the challenges, we adapted and streamlined a reversible Renilla luciferase protein complementation assay (Rluc-PCA), originally reported for use in human cells, for transient expression in Nicotiana benthamiana. We tested Rluc-PCA and successfully identified luminescence complementation amongst Golgi-localizing GTs known to form a heterodimer (GAUT1 and GAUT7) and those which homooligomerize (ARAD1). In contrast, no interaction was shown between negative controls (e.g. GAUT7, ARAD1, IRX9). Rluc-PCA was used to investigate PPIs amongst Golgi-localizing GTs involved in biosynthesis of hemicelluloses. Although no PPI was identified among six GTs involved in xylan biosynthesis, Rluc-PCA confirmed three previously proposed interactions and identified seven novel PPIs amongst GTs involved in xyloglucan biosynthesis. Notably, three of the novel PPIs were confirmed by a yeast-based split-ubiquitin assay. Finally, Gateway-enabled expression vectors were generated, allowing rapid construction of fusion proteins to the Rluc reporters and epitope tags. Our results show that Rluc-PCA coupled with transient expression in N. benthamiana is a fast and versatile method suitable for analysis of PPIs between Golgi resident proteins in an easy and mid-throughput fashion in planta
Fluctuating Atlantic inflows modulate Arctic atlantification
Enhanced warm, salty subarctic inflows drive high-latitude atlantification, which weakens oceanic stratification, amplifies heat fluxes, and reduces sea ice. In this work, we show that the atmospheric Arctic Dipole (AD) associated with anticyclonic winds over North America and cyclonic winds over Eurasia modulates inflows from the North Atlantic across the Nordic Seas. The alternating AD phases create a âswitchgear mechanism.â From 2007 to 2021, this switchgear mechanism weakened northward inflows and enhanced sea-ice export across Fram Strait and increased inflows throughout the Barents Sea. By favoring stronger Arctic Ocean circulation, transferring freshwater into the Amerasian Basin, boosting stratification, and lowering oceanic heat fluxes there after 2007, AD+ contributed to slowing sea-ice loss. A transition to an ADâ phase may accelerate the Arctic sea-ice decline, which would further change the Arctic climate system.acceptedVersio
