    Low Dose of BPA Induces Liver Injury through Oxidative Stress, Inflammation and Apoptosis in Long–Evans Lactating Rats and Its Perinatal Effect on Female PND6 Offspring

    Bisphenol A (BPA) is a phenolic compound used in plastics elaboration for food protection or packaging. BPA-monomers can be released into the food chain, resulting in continuous and ubiquitous low-dose human exposure. This exposure during prenatal development is especially critical and could lead to alterations in ontogeny of tissues increasing the risk of developing diseases in adulthood. The aim was to evaluate whether BPA administration (0.036 mg/kg b.w./day and 3.42 mg/kg b.w./day) to pregnant rats could induce liver injury by generating oxidative stress, inflammation and apoptosis, and whether these effects may be observed in female postnatal day-6 (PND6) offspring. Antioxidant enzymes (CAT, SOD, GR, GPx and GST), glutathione system (GSH/GSSG) and lipid-DNA damage markers (MDA, LPO, NO, 8-OHdG) were measured using colorimetric methods. Inducers of oxidative stress (HO-1d, iNOS, eNOS), inflammation (IL-1β) and apoptosis (AIF, BAX, Bcl-2 and BCL-XL) were measured by qRT-PCR and Western blotting in liver of lactating dams and offspring. Hepatic serum markers and histology were performed. Low dose of BPA caused liver injury in lactating dams and had a perinatal effect in female PND6 offspring by increasing oxidative stress levels, triggering an inflammatory response and apoptosis pathways in the organ responsible for detoxification of this endocrine disruptor.Depto. de FisiologíaSección Deptal. de Bioquímica y Biología Molecular (Medicina)Fac. de MedicinaTRUEUnión Europea.     The role of outdoor and indoor air quality in the spread of SARS-CoV-2: Overview and recommendations by the research group on COVID-19 and particulate matter (RESCOP commission)

    There are important questions surrounding the potential contribution of outdoor and indoor air quality in the transmission of SARS-CoV-2 and perpetuation of COVID-19 epidemic waves. Environmental health may be a critical component of COVID-19 prevention. The public health community and health agencies should consider the evolving evidence in their recommendations and statements, and work to issue occupational guidelines. Evidence coming from the current epidemiological and experimental research is expected to add knowledge about virus diffusion, COVID-19 severity in most polluted areas, inter-personal distance requirements and need for wearing face masks in indoor or outdoor environments. The COVID-19 pandemic has highlighted the need for maintaining particulate matter concentrations at low levels for multiple health-related reasons, which may also include the spread of SARS-CoV-2. Indoor environments represent even a more crucial challenge to cope with, as it is easier for the SARS-COV2 to spread, remain vital and infect other subjects in closed spaces in the presence of already infected asymptomatic or mildly symptomatic people. The potential merits of preventive measures, such as CO2 monitoring associated with natural or controlled mechanical ventilation and air purification, for schools, indoor public places (restaurants, offices, hotels, museums, theatres/cinemas etc.) and transportations need to be carefully considered. Hospital settings and nursing/retirement homes as well as emergency rooms, infectious diseases divisions and ambulances represent higher risk indoor environments and may require additional monitoring and specific decontamination strategies based on mechanical ventilation or air purificatio

    B cell response after SARS-CoV-2 mRNA vaccination in people living with HIV

    Background: Limited longitudinal data are available on immune response to mRNA SARS-CoV-2 vaccination in people living with HIV (PLWHIV); therefore, new evidence on induction and persistence of spike-specific antibodies and B cells is needed. Methods: In this pilot study we investigated the spike-specific humoral and B cell responses up to six months after vaccination with two doses of mRNA vaccines in 84 PLWHIV under antiretroviral therapy compared to 79 healthy controls (HCs). Results: Spike-specific IgG persisted six months in PLWHIV with no significant differences compared to HCs, even though a significantly lower IgG response was observed in patients with CD4+ T cells < 350/mmc. The frequency of subjects with antibodies capable of inhibiting ACE2/RBD binding was comparable between PLWHIV and HCs a month after the second vaccine dose, then a higher drop was observed in PLWHIV. A comparable percentage of spike-specific memory B cells was observed at month six in PLWHIV and HCs. However, PLWHIV showed a higher frequency of spike-specific IgD- CD27- double-negative memory B cells and a significantly lower rate of IgD- CD27+ Ig-switched memory B cells compared to HCs, suggesting a reduced functionality of the antigen-specific memory B population. Conclusions: The mRNA vaccination against SARS-CoV-2 elicits humoral and B cell responses quantitatively similar between PLWHIV and HCs, but there are important differences in terms of antibody functionality and phenotypes of memory B cells, reinforcing the notion that tailored vaccination policies should be considered for these patients

    Origin of the unusual ground-state spin S = 9 in a Cr10 single-molecule magnet

    The molecular wheel [Cr10(OMe)20(O2CCMe3)10], abbreviated {Cr10}, with an unusual intermediate total spin S = 9 and non-negligible cluster anisotropy, D/kB = −0.045(2) K, is a rare case among wheels based on an even number of 3d-metals, which usually present an antiferromagnetic (AF) ground state (S = 0). Herein, we unveil the origin of such a behavior. Angular magnetometry measurements performed on a single crystal confirmed the axial anisotropic behavior of {Cr10}. For powder samples, the temperature dependence of the susceptibility plotted as χT(T) showed an overall ferromagnetic (FM) behavior down to 1.8 K, whereas the magnetization curve M(H) did not saturate at the expected 30 μB/fu for 10 FM coupled 3/2 spin Cr3+ ions, but to a much lower value, corresponding to S = 9. In addition, the X-ray magnetic circular dichroism (XMCD) measured at high magnetic field (170 kOe) and 7.5 K showed the polarization of the cluster moment up to 23 μB/fu. The magnetic results can be rationalized within a model, including the cluster anisotropy, in which the {Cr10} wheel is formed by two semiwheels, each with four Cr3+ spins FM coupled (JFM/kB = 2.0 K), separated by two Cr3+ ions AF coupled asymmetrically (J23/kB = J78/kB = −2.0 K; J34/kB = J89/kB = −0.25 K). Inelastic neutron scattering and heat capacity allowed us to confirm this model leading to the S = 9 ground state and first excited S = 8. Single-molecule magnet behavior with an activation energy of U/kB = 4.0(5) K in the absence of applied field was observed through ac susceptibility measurements down to 0.1 K. The intriguing magnetic behavior of {Cr10} arises from the detailed asymmetry in the molecule interactions produced by small-angle distortions in the angles of the Cr–O–Cr alkoxy bridges coupling the Cr3+ ions, as demonstrated by ab initio and density functional theory calculations, while the cluster anisotropy can be correlated to the single-ion anisotropies calculated for each Cr3+ ion in the wheel.The authors acknowledge financial support from the Spanish Agencia Estatal de Investigación, through Projects MAT2017-83468-R (AEI/FEDER, UE) and PID2020-115159GB-I00/AEI/10.13039/501100011033, Aragonese Project RASMIA E12_20R (co-funded by Fondo Social Europeo) and of the European Union FEDER (ES).     A genome-wide association study for survival from a multi-centre European study identified variants associated with COVID-19 risk of death

    : The clinical manifestations of SARS-CoV-2 infection vary widely among patients, from asymptomatic to life-threatening. Host genetics is one of the factors that contributes to this variability as previously reported by the COVID-19 Host Genetics Initiative (HGI), which identified sixteen loci associated with COVID-19 severity. Herein, we investigated the genetic determinants of COVID-19 mortality, by performing a case-only genome-wide survival analysis, 60 days after infection, of 3904 COVID-19 patients from the GEN-COVID and other European series (EGAS00001005304 study of the COVID-19 HGI). Using imputed genotype data, we carried out a survival analysis using the Cox model adjusted for age, age2, sex, series, time of infection, and the first ten principal components. We observed a genome-wide significant (P-value < 5.0 × 10-8) association of the rs117011822 variant, on chromosome 11, of rs7208524 on chromosome 17, approaching the genome-wide threshold (P-value = 5.19 × 10-8). A total of 113 variants were associated with survival at P-value < 1.0 × 10-5 and most of them regulated the expression of genes involved in immune response (e.g., CD300 and KLR genes), or in lung repair and function (e.g., FGF19 and CDH13). Overall, our results suggest that germline variants may modulate COVID-19 risk of death, possibly through the regulation of gene expression in immune response and lung function pathways
