    Characterizing the fungal and bacterial microflora and concentrations in fitness centres

    Fitness centres are special places where conditions for microbiological proliferation should be considered. Moisture due to human perspiration and water condensation as a result of human physical activities are prevalent in this type of buildings. Exposure to microbial contaminants is clinically associated with respiratory disorders and people who work out in polluted environments would be susceptible to contaminants. This work studied the indoor air contamination in three gymnasiums in Lisbon. The sampling was performed at two periods: at the opening (morning) and closing (night) of the three gymnasiums. The airborne bacterial and fungal populations were sampled by impaction directly onto Tryptic Soy Agar (for bacteria) and Malt Extract Agar (for fungi) plates, using a Merck MAS-100 air sampler. Higher bacterial concentrations were found at night as compared to the morning but the same behaviour was not found for fungal concentrations. Gram-negative catalase positive cocci were the dominant bacteria in indoor air samples of the studied gymnasiums. In this study, 21 genera/species of fungal colonies were identified. Chrysosporium sp., Chrysonilia sp., Neoscytalidium hialinum, Sepedonium sp. and Penicillium sp. were the most prevalent species identified in the morning, while Cladosporium sp., Penicillium sp., Chrysosporium sp., Acremonium sp. and Chrysonilia sp. were more prevalent at night. A well-designed sanitation and maintenance program for gymnasiums is needed to ensure healthier space for indoor physical activity

    A cryptic RNA-binding domain mediates Syncrip recognition and exosomal partitioning of miRNA targets

    Exosomal miRNA transfer is a mechanism for cell-cell communication that is important in the immune response, in the functioning of the nervous system and in cancer. Syncrip/hnRNPQ is a highly conserved RNA-binding protein that mediates the exosomal partition of a set of miRNAs. Here, we report that Syncrip's amino-terminal domain, which was previously thought to mediate protein-protein interactions, is a cryptic, conserved and sequence-specific RNA-binding domain, designated NURR (N-terminal unit for RNA recognition). The NURR domain mediates the specific recognition of a short hEXO sequence defining Syncrip exosomal miRNA targets, and is coupled by a non-canonical structural element to Syncrip's RRM domains to achieve high-affinity miRNA binding. As a consequence, Syncrip-mediated selection of the target miRNAs implies both recognition of the hEXO sequence by the NURR domain and binding of the RRM domains 5′ to this sequence. This structural arrangement enables Syncrip-mediated selection of miRNAs with different seed sequences. © 2018 The Author(s)

    Population genetic signatures of a climate change driven marine range extension

    Shifts in species distribution, or 'range shifts', are one of the most commonly documented responses to ocean warming, with important consequences for the function and structure of ecosystems, and for socio-economic activities. Understanding the genetic signatures of range shifts can help build our knowledge of the capacity of species to establish and persist in colonised areas. Here, seven microsatellite loci were used to examine the population connectivity, genetic structure and diversity of Octopus tetricus, which has extended its distribution several hundred kilometres polewards associated with the southwards extension of the warm East Australian Current along south-eastern Australia. The historical distribution and the range extension zones had significant genetic differences but levels of genetic diversity were comparable. The population in the range extension zone was sub-structured, contained relatively high levels of self-recruitment and was sourced by migrants from along the entire geographic distribution. Genetic bottlenecks and changes in population size were detected throughout the range extension axis. Persistent gene flow from throughout the historical zone and moderate genetic diversity may buffer the genetic bottlenecks and favour the range extension of O. tetricus. These characteristics may aid adaptation, establishment, and long-term persistence of the population in the range extension zone

    Different carbon nanostructured materials obtained in catalytic chemical vapor deposition

    Different carbon nanostructured materials, such as nanotubes, nanofibers, nanosprings and nanooctopus, were grown by changing the metal catalyst and experimental parameters of the thermal chemical vapor deposition process. These experiments were performed using a tubular furnace and methane or acetylene as carbon feedstock gases. Thin films of Ni or Cu were deposited onto a SiO2/Si substrate and employed as catalysts. The effect of the growth temperature, metal catalyst and carbon gas precursor (methane or acetylene) on the final carbon nanoestructured material was studied by scanning electron microscopy, Raman spectroscopy and grazing incidence X-ray diffraction. Growth of multiwall carbon nanotubes (MWCNTs) was observed using both metal films and carbon precursor gases, whereas partially oxidized Ni films promoted formation of nanosprings. Experiments with reduced supply of methane resulted in octopus-like carbon nanostructures when a Cu film was used as a catalyst.Diferentes materiais nanoestruturados à base de carbono, tais como nanotubos, nanofibras, nanomolas e nanooctopus, foram obtidos através do processo de deposição química de vapor. Tais experimentos foram realizados em um forno tubular e variações nos parâmetros experimentais permitiram a obtenção das diferentes nanoestruturas de carbono. Filmes finos de Ni e Cu foram depositados sobre substratos de SiO2/Si e empregados como catalisadores. O efeito de diferentes gases precursores de carbono, da temperatura de crescimento e do metal catalítico sobre as características do material final foi investigado por microscopia eletrônica de varredura, espectroscopia Raman e difratometria de raios X com ângulo rasante. O uso dos gases metano e acetileno levaram à formação de nanotubos de carbono para ambos os filmes metálicos, enquanto Ni parcialmente oxidado promoveu o crescimento de nanomolas.     Agreement between two photoplethysmography-based wearable devices for monitoring heart rate during different physical activity situations : a new analysis methodology

    Wearables are being increasingly used to monitor heart rate (HR). However, their usefulness for analyzing continuous HR in research or at clinical level is questionable. The aim of this study is to analyze the level of agreement between different wearables in the measurement of HR based on photoplethysmography, according to different body positions and physical activity levels, and compared to a gold-standard ECG. The proposed method measures agreement among several time scales since different wearables obtain HR at different sampling rates. Eighteen university students (10 men, 8 women; 22 ± 2.45 years old) participated in a laboratory study. Participants simultaneously wore an Apple Watch and a Polar Vantage watch. ECG was measured using a BIOPAC system. HR was recorded continuously and simultaneously by the three devices, for consecutive 5-min periods in 4 different situations: lying supine, sitting, standing and walking at 4 km/h on a treadmill. HR estimations were obtained with the maximum precision offered by the software of each device and compared by averaging in several time scales, since the wearables obtained HR at different sampling rates, although results are more detailed for 5 s and 30 s epochs. Bland-Altman (B-A) plots show that there is no noticeable difference between data from the ECG and any of the smartwatches while participants were lying down. In this position, the bias is low when averaging in both 5 s and 30 s. Differently, B-A plots show that there are differences when the situation involves some level of physical activity, especially for shorter epochs. That is, the discrepancy between devices and the ECG was greater when walking on the treadmill and during short time scales. The device showing the biggest discrepancy was the Polar Watch, and the one with the best results was the Apple Watch. We conclude that photoplethysmography-based wearable devices are suitable for monitoring HR averages at regular intervals, especially at rest, but their feasibility is debatable for a continuous analysis of HR for research or clinical purposes, especially when involving some level of physical activity.     Translating COGNISTAT and the Use of the Cognitive Interview Approach: Observations and Challenges

    Cognitive functioning is a salient issue among people in the late adulthood stage where mental health declines with age. A common cognitive disability among elders is Dementia including Alzheimer’s disease. Cognitive screening tools such as the Mini- Mental Status Examination and the Montreal Cognitive Assessment are most commonly used to measure the cognitive ability areas leading to a diagnostic evaluation. COGNISTAT as a neuropsychological instrument is a recent screener being introduced to a few outpatient clinics. In this study; COGNISTAT is translated to Filipino for the first time using a rigorous procedure and pilot tested on elderly volunteers in local settings. The translation was done by an interdisciplinary team of a Geriatric Physician; a Speech Pathologist; and a Clinical Psychologist. The Filipino-translated COGNISTAT was administered to 22 elders in two batches using the cognitive interview method. Challenges were identified in the initial testing phase which has implications for future adaptations of foreign instruments

    Unravelling the biological potential of Pinus pinaster bark extracts

    Natural compounds from agro-food by-products have fostered interest in food industries. The aim of this study was to unravel potential uses for Pinus pinaster bark extracts (PBE). As functional features of this type of extracts are usually attributed to phenolic compounds, the extraction process was studied. Different PBEs were achieved, with high content in phenolic compounds, using different water/ethanol combinations as a solvent. These PBEs were chemically characterized, and their bioactivity and in vitro cell viability were evaluated. Extracts obtained with hydroethanolic solvents had higher content in phenolic and flavonoid compounds. All the PBEs presented high antioxidant, antibacterial and antihyperglycemic activities. Moreover, PBEs have low cytotoxicity and a selective activity against cancer cells as these were negatively affected.
