Die Vegetationsentwicklung des Interglazials von Wallensen in der Hilsmulde
1. Von dem zuerst 1907 von Menzel beschriebenen, vor kurzem (1951) auch von P. W. Thomson pollenanalytisch untersuchten Interglazial von Wallensen wurden noch 3 weitere Diagramme aufgestellt, ein Profil und über 50 Einzelproben auf Großreste untersucht. Die nachgewiesene Folge der Waldperioden: 5. Kiefern-Eichenmischwaldzeit — 6. Hasel-Eichenmischwaldzeit — 7. Hainbuchenzeit — 8. Tannenzeit — 9. Fichtenzeit — 10. Kiefernzeit, stimmt — mutatis mutandis — völlig mit der Periodenfolge überein, die in Dänemark und im nordwestdeutschen Flachland als für das letzte Interglazial bezeichnend erkannt worden ist. (Die ältesten frühinterglazialen Abschnitte sind nicht erfaßt worden). Danach kann (mit Thomson) Wallensen dem letzten Interglazial zugeordnet und die erwähnte Waldfolge auch für die nordwestlichen Mittelgebirge als gültig angesehen werden. Von Fagus wurde kein einziges Pollenkorn nachgewiesen. 2. Die pollenanalytischen Befunde konnten durch die Untersuchung von Großresten, u. a. durch zahlreiche Funde von Knospenschuppen ergänzt und erweitert werden. Insgesamt wurden 80 Sippen, meist Arten, z. T. Gattungen oder Familien, bestimmt, wonach Wallensen als artenreiche Interglazialflora bezeichnet werden kann. Die Wasserflora war während der Hasel-Eichenmischwaldzeit am reichhaltigsten; für diese Zeit konnten auch die in Europa heute ausgestorbenen interglazialen Leitpflanzen Brasenia purpurea und Dulichium spathaceum nachgewiesen werden. Bemerkenswert ist auch der Nachweis der heute südosteuropäischen Tilia tomentosa. Ein merkwürdiger Fruchtrest (Steinkern?), der unter Nr. 79 beschrieben worden ist und wahrscheinlich von keiner mitteleuropäischen Art stammt, gehört sowohl der Hasel-Eichenmischwald- wie der Hainbuchenzeit an. 3. Reiche Funde von Hex, Hedera, Taxus und Osmunda bezeugen eine gewisse thermische Ozeanität des Gebiets während aller Zeiten. Während der Hasel-Eichenmischwaldzeit und der Hainbuchenzeit dürfte das Gebiet aber sommerwärmer gewesen sein als heute.researc
Limited utility of qPCR-based detection of tumor-specific circulating mRNAs in whole blood from clear cell renal cell carcinoma patients
BACKGROUND:
RNA sequencing data is providing abundant information about the levels of dysregulation of genes in various tumors. These data, as well as data based on older microarray technologies have enabled the identification of many genes which are upregulated in clear cell renal cell carcinoma (ccRCC) compared to matched normal tissue. Here we use RNA sequencing data in order to construct a panel of highly overexpressed genes in ccRCC so as to evaluate their RNA levels in whole blood and determine any diagnostic potential of these levels for renal cell carcinoma patients.
METHODS:
A bioinformatics analysis with Python was performed using TCGA, GEO and other databases to identify genes which are upregulated in ccRCC while being absent in the blood of healthy individuals. Quantitative Real Time PCR (RT-qPCR) was subsequently used to measure the levels of candidate genes in whole blood (PAX gene) of 16 ccRCC patients versus 11 healthy individuals. PCR results were processed in qBase and GraphPadPrism and statistics was done with Mann-Whitney U test.
RESULTS:
While most analyzed genes were either undetectable or did not show any dysregulated expression, two genes, CDK18 and CCND1, were paradoxically downregulated in the blood of ccRCC patients compared to healthy controls. Furthermore, LOX showed a tendency towards upregulation in metastatic ccRCC samples compared to non-metastatic.
CONCLUSIONS:
This analysis illustrates the difficulty of detecting tumor regulated genes in blood and the possible influence of interference from expression in blood cells even for genes conditionally absent in normal blood. Testing in plasma samples indicated that tumor specific mRNAs were not detectable. While CDK18, CCND1 and LOX mRNAs might carry biomarker potential, this would require validation in an independent, larger patient cohort
GRAVITY: The AO-Assisted, Two-Object Beam-Combiner Instrument
We present the proposal for the infrared adaptive optics (AO) assisted,
two-object, high-throughput, multiple-beam-combiner GRAVITY for the VLTI. This
instrument will be optimized for phase-referenced interferometric imaging and
narrow-angle astrometry of faint, red objects. Following the scientific
drivers, we analyze the VLTI infrastructure, and subsequently derive the
requirements and concept for the optimum instrument. The analysis can be
summarized with the need for highest sensitivity, phase referenced imaging and
astrometry of two objects in the VLTI beam, and infrared wavefront-sensing.
Consequently our proposed instrument allows the observations of faint, red
objects with its internal infrared wavefront sensor, pushes the optical
throughput by restricting observations to K-band at low and medium spectral
resolution, and is fully enclosed in a cryostat for optimum background
suppression and stability. Our instrument will thus increase the sensitivity of
the VLTI significantly beyond the present capabilities. With its two fibers per
telescope beam, GRAVITY will not only allow the simultaneous observations of
two objects, but will also push the astrometric accuracy for UTs to 10

Wirtsgalaxien von Quasaren und der Laserleitstern fĂĽr das Very Large Telescope
Astronomische Beobachtungen mit groĂźen bodengebundenen
Teleskopen sind, bedingt durch die statistischen Prozesse der
Lichttransmission durch die Erdatmosphäre, in der räumlichen
Auflösung begrenzt. Mittels adaptiver Optik, einer schnellen
Korrektur der Lichtwellenfront, kann diese Einschränkung behoben
werden. Damit diese Technologie auch fĂĽr lichtschwache Objekte
eingesetzt werden kann, ist eine kĂĽnstliche Referenzquelle in der
Hochatmosphäre nötig. Die vorliegende Dissertation gliedert sich
in zwei Teile: Im astronomischen ersten Teil werden Beobachtungen
von Quasaren mittlerer bis hoher Rotverschiebung beschrieben. Der
zweite Teil beschäftigt sich mit der Entwicklung eines
Lasersystems fĂĽr das Very Large Telescope in Chile, mit dem ein
kĂĽnstlicher Leitstern erzeugt
werden kann.\\
Mittels optischer und nahinfraroter Aufnahmen radioschwacher
Quasare in mehreren Rotverschiebungsbereichen wird im Rahmen
dieser Arbeit eine Messung der Leuchtkraft des Quasarkerns sowie
der beherbergenden Galaxie vorgenommen. Durch den Vergleich mit
der räumlichen Intensitätsverteilung von Punktquellen im selben
Feld mit den Beobachtungsobjekten konnte die geringfĂĽgig
größere räumliche Ausdehnung der Galaxie detektiert werden. Das
Helligkeitsverhältnis von Kern und Galaxie wurde mit Hilfe einer
Modellierung der Oberflächenhelligkeit und der Anpassung an die
Messwerte ermittelt. Dieses Verhältnis wird im Vergleich mit
Messungen anderer Gruppen sowie Modellrechnungen der Galaxie- und
Quasarbildung im jungen Universum diskutiert. Ăśber den hier
betrachteten Rotverschiebungsbereich von z=0.8 bis z=2.7 liegt
dieses Verhältnis innerhalb der zu erwartenden Streuung der
Modellrechnungen, wobei ein eindeutiger Trend mit dem
kosmologischen Alter innerhalb der Fehlergrenzen nicht
festgestellt werden konnte. Vergleicht man die Leuchtkraft der
hier vermessenen radioschwachen Wirtsgalaxien der Quasare mit
radiolauten Objekten sowie anderen Galaxientypen ähnlicher
Rotverschiebung zeigt sich, dass diese eher mit Lyman-break
Galaxien ĂĽbereinstimmen und in der mittleren Helligkeit
vergleichbar mit 'normalen' L* Galaxien sind.
Die hier vorgestellten Messungen wurden an einem 3.5m Teleskop
ohne Korrektur der atmosphärischen Störungen vorgenommen und
sind dadurch prinzipiell in der erreichbaren räumlichen
Auflösung begrenzt. Um zukünftige Messungen von Quasaren und
ähnliche Projekte, bei denen eine hohe Auflösung gefordert ist,
mit größerer Präzision durchführen zu können, ist ein
Laserleitstern an einem großen Teleskop nötig. In Kapitel 2
werden die grundlegenden Eigenschaften der Lichttransmission durch
die Erdatmosphäre und deren Implikation auf die Abbildung mit
astronomischen Teleskopen beschrieben. Im selben Kapitel wird die
Physik der Resonanzstreuung an Natriumatomen in der Mesosphäre
beschrieben, welche zur Erzeugung des kĂĽnstlichen Leitsterns
genutzt wird und die Grundlage fĂĽr die Auslegung des
Laserleitsterns bildet. In Kapitel 3 wird die Entwicklung und der
Test des Lasersystems beschrieben, welches im Rahmen dieser Arbeit
fĂĽr das Very Large Telescope in Chile gebaut wurde. Ziel war es,
einen Laser zu entwickeln, der bei 589 nm mehr als 10 W
Ausgangsleistung in einer einzelnen Mode mit hoher Strahlqualität erreicht. Mit der Aufteilung des Lasersystems in eine Master-Laser- und eine Verstärkerstufe konnten die Problematiken, welche sich durch thermische Störungen bei hoher Leistung ergeben, gelöst werden. Die cw Verstärkerstufe wurde als injektionsstabilisierter Resonator mit nicht-planarer Geometrie und zwei Farbstoffstrahlen verwirklicht, welche von vier leistungsstarken 532nm Lasern optisch gepumpt werden. Mit Hilfe eines detaillierten Modells des Laserprozesses konnte die Auslegung des Verstärkers erfolgen. Für die Stabilisierung des Verstärkers auf die Resonanzspitze wurden mehrere Methoden getestet. Hierbei konnte eine neue polarisationsspektroskopische Messmethode gefunden werden, welche ein eindeutiges Fehlersignal über den gesamten Phasenbereich liefert, womit eine hochstabile Regelung verwirklicht werden konnte. Mit dem Gesamtsystem konnte stabil eine Einmoden-Ausgangsleistung von 24 W mit hervorragender Strahlqualität erreicht werden
ALFA: First Operational Experience of the MPE/MPIA Laser Guide Star System for Adaptive Optics
The sodium laser guide star adaptive optics system ALFA has been constructed
at the Calar Alto 3.5-m telescope. Following the first detection of the laser
beacon on the wavefront sensor in 1997 the system is now being optimized for
best performance. In this contribution we discuss the current status of the
launch beam and the planned improvements and upgrades. We report on the
performance level achieved when it is used with the adaptive optics system, and
relate various aspects of our experience during operation of the system. We

The foraminiferal signature of recent Gulf of Mexico hurricanes
Down-core activities of �Be, ²³�Th, and ²¹�Pb, as well as X-radiography, have indicated the existence of sediments deposited by hurricanes Ivan (2004), Katrina (2005), and Rita (2005) on the continental margin west of the Mississippi Delta between Southwest Pass and the Mississippi Canyon. These radionuclides have fairly short half-lives on the order of days to decades, however, and their utility for the identification of hurricane events decreases with progressively older deposits. Foraminifera, which can be preserved for geologically significant amounts of time, may serve as another proxy for the detection of hurricane event beds. Here I investigate foraminiferal assemblages contained within recent, known hurricane units to determine whether they differ significantly from those of non-hurricane units and a unit deposited by a river flood event, and whether these differences might give insight into where the hurricane-deposited sediment originated. Cores were collected along the same transect during three different cruises in: 2004, 2005, and 2007; the timing of collection reflected periods of hurricane and non-hurricane influenced deposition. The transect runs southwest from Southwest Pass (~30 m water depth) to the head of the Mississippi Canyon (~170 m depth). Surface samples of a unit deposited by the 2011 Mississippi River flood event were also collected to allow comparison with hurricane units. Average relative abundances of foraminifera were analyzed for trends and ANOVA and discriminant analysis were utilized to determine whether the foraminiferal assemblages of hurricane and non-hurricane units were statistically discrete. Hurricane unit foraminifera very close to the mouth of Southwest Pass (9 km; ~30 m water depth) were more abundant per unit volume of sediment than pre-hurricane foraminifera at this location. Hurricane units further out on the shelf (> 80 m water depth) tended to contain less foraminifera per unit volume than did the non-hurricane units. Hurricane units also contained more textulariids and miliolids than the non-hurricane units, and sometimes contained rare marsh taxa. ANOVA results show that the abundances of 13 of the 32 most abundant taxa were statistically different between units when comparing all four unit types (hurricane, pre- and post-hurricane, flood, and bioturbated). Ten of these species were statistically different between hurricane and non-hurricane units; relative abundance of coastal taxa increased in the hurricane units. Discriminant analysis indicates that all four unit types are generally discrete. The species that contributed most to the discrimination of unit types were generally rare, and some were helpful in the determination of the origin of the hurricane-transported sediment. In contrast, the species that were significant in the ANOVA were among the most abundant. Thus, both abundant and rare species are useful for identifying hurricane deposited sediment. A portion of hurricane unit sediments was likely locally resuspended and redeposited (indicated by high relative abundances of those taxa which were abundant in the pre- and post- hurricane units). There is also evidence of seaward transport of sediment; some species that occur in the pre- and post-hurricane units only at the shallow end of the transect also occurred in hurricane units at the deeper end of the transect. In addition, hurricane units contained some rare marsh and upper slope taxa which were absent from the pre- and post-hurricane units, indicating some portion of the sediment in the hurricane units is derived from these environments. In summary, foraminifera can provide information on the provenance of hurricane-deposited sediment soon after deposition, but bioturbation can destroy this signal rapidly.M.S
