    Transition on the entropic elasticity of DNA induced by intercalating molecules

    We use optical tweezers to perform stretching experiments on DNA molecules when interacting with the drugs daunomycin and ethidium bromide, which intercalate the DNA molecule. These experiments are performed in the low-force regime from zero up to 2 pN. Our results show that the persistence length of the DNA-drug complexes increases strongly as the drug concentration increases up to some critical value. Above this critical value, the persistence length decreases abruptly and remains practically constant for larger drug concentrations. The contour length of the molecules increases monotonically and saturates as drugs concentration increases. Measured in- tercalants critical concentrations for the persistence length transition coincide with reported values for the helix-coil transition of DNA-drug complexes, obtained from sedimentation experiments.Comment: This experimental article shows and discuss a transition observed in the persistence length of DNA molecules when studied as a function of some intercalating drug concentrations, like daunomycin and ethidium bromide.     Towards absolute calibration of optical tweezers

    Aiming at absolute force calibration of optical tweezers, following a critical review of proposed theoretical models, we present and test the results of MDSA (Mie-Debye-Spherical Aberration) theory, an extension of a previous (MD) model, taking account of spherical aberration at the glass/water interface. This first-principles theory is formulated entirely in terms of experimentally accessible parameters (none adjustable). Careful experimental tests of the MDSA theory, undertaken at two laboratories, with very different setups, are described. A detailed description is given of the procedures employed to measure laser beam waist, local beam power at the transparent microspheres trapped by the tweezers, microsphere radius and the trap transverse stiffness, as a function of radius and height in the (inverted microscope) sample chamber. We find generally very good agreement with MDSA theory predictions, for a wide size range, from the Rayleigh domain to large radii, including the values most often employed in practice, and at different chamber heights, both with objective overfilling and underfilling. The results asymptotically approach geometrical optics in the mean over size intervals, as they should, and this already happens for size parameters not much larger than unity. MDSA predictions for the trapping threshold, position of stiffness peak, stiffness variation with height, multiple equilibrium points and `hopping' effects among them are verified. Remaining discrepancies are ascribed to focus degradation, possibly arising from objective aberrations in the infrared, not yet included in MDSA theory.Comment: 15 pages, 20 figure

    The silicon stable isotope distribution along the GEOVIDE section (GEOTRACES GA-01) of the North Atlantic Ocean

    The stable isotope composition of dissolved silicon in seawater (δ30SiDSi) was examined at 10 stations along the GEOVIDE section (GEOTRACES GA-01), spanning the North Atlantic Ocean (40–60∘ N) and Labrador Sea. Variations in δ30SiDSi below 500 m were closely tied to the distribution of water masses. Higher δ30SiDSi values are associated with intermediate and deep water masses of northern Atlantic or Arctic Ocean origin, whilst lower δ30SiDSi values are associated with DSi-rich waters sourced ultimately from the Southern Ocean. Correspondingly, the lowest δ30SiDSi values were observed in the deep and abyssal eastern North Atlantic, where dense southern-sourced waters dominate. The extent to which the spreading of water masses influences the δ30SiDSi distribution is marked clearly by Labrador Sea Water (LSW), whose high δ30SiDSi signature is visible not only within its region of formation within the Labrador and Irminger seas, but also throughout the mid-depth western and eastern North Atlantic Ocean. Both δ30SiDSi and hydrographic parameters document the circulation of LSW into the eastern North Atlantic, where it overlies southern-sourced Lower Deep Water. The GEOVIDE δ30SiDSi distribution thus provides a clear view of the direct interaction between subpolar/polar water masses of northern and southern origin, and allow examination of the extent to which these far-field signals influence the local δ30SiDSi distribution

    Form factors of heavy-light systems in point-form relativistic quantum mechanics: the Isgur-Wise function

    We investigate electromagnetic and weak form factors of heavy-light mesons in the context of point-form relativistic quantum mechanics. To this aim we treat the physical processes from which such electroweak form factors are extracted by means of a coupled channel approach which accounts for the dynamics of the intermediate gauge bosons. It is shown that heavy-quark symmetry is respected by this formulation. A simple analytical expression is obtained for the Isgur-Wise function in the heavy-quark limit.     Imaging 3D seismic velocity along the seismogenic zone of Algarve region (southern Portugal)

    The present seismic tomographic study is focused around Algarve region, in South of Portugal. To locate the seismic events and find the local velocity structure of epicentral area, the P and S arrival times at 38 stations are used. The data used in this study were obtained during the Algarve campaign which worked from January/2006 to July/2007. The preliminary estimate of origin times and hypocentral coordinates are determined by the Hy- poinverse program. Linearized inversion procedure was applied to comprise the following two steps: 1) finding the minimum 1D velocity model using Velest and 2) simultaneous relocation of hypocenters and determination of local velocity structure. The velocity model we have reached is a 10 layer model which gave the lowest RMS, after several runnings of eight different velocity models that we used “a priori”. The model parameterization assumes a continuous velocity field between 4.5 km/s and 7.0 km/s until 30 km depth. The earth structure is represented in 3D by velocity at discrete points, and velocity at any intervening point is determined by linear interpolation among the surrounding eight grid points. A preliminary analysis of the resolution capabilities of the dataset, based on the Derivative Weight Sum (DWS) distribution, shows that the velocity structure is better resolved in the West part of the region between the surface to15 km. The resulting tomographic image has a prominent low-velocity anomaly that shows a maximum decrease in P-wave velocity in the first 12 kms in the studied region. We also identified the occurrence of local seismic events of reduced magnitude not catalogued, in the neighbourhood of Almodôvar (low Alentejo). The spatial distribution of epicentres defines a NE-SW direction that coincides with the strike of the mapped geological faults of the region and issued from photo-interpretation. Is still expectable to refine the seismicity of the region of Almodôvar and establish more rigorously its role in the seismotectonic picture of the region. This work is expected to produce a more detailed knowledge of the structure of the crust over the region of Algarve, being able to identify seismogenic zones, potentially generators of significant seismic events and also the identification of zones of active faults

    Quality improvement of traditional dry fermented sausages based on innovative technological strategies

    Dry fermented sausages are highly appreciated food specialties, mainly in Portugal and other southern European countries. Therefore, all research efforts aiming at improving the food quality and safety of traditional dry sausages are of interest, since they are likely to result in products with higher added value and quality standards most suited to the requirements and concerns of the modern consumers. Among those efforts, it may be highlighted the studies involving innovative processing parameters and technologies to overcome practical problems gathered in the meat industry, which are mostly associated with food quality and safety. Additionally, characterization of traditional dry sausages and rationalization of their processing are essential for further achievement of any official certification. Thus, this article attempts to point out some research lines of highest interest in meat science (and particularly to the broad variety of regional dry fermented sausages), towards to the valorisation of technological, nutritional and commercial features. In addition, it is here emphasized the importance for the continuous improvement of the quality and safety of meat products as a way to respond to the current concerns regarding its consumption and the general advices in reducing its daily intake

    DNA-psoralen: single-molecule experiments and first principles calculations

    The authors measure the persistence and contour lengths of DNA-psoralen complexes, as a function of psoralen concentration, for intercalated and crosslinked complexes. In both cases, the persistence length monotonically increases until a certain critical concentration is reached, above which it abruptly decreases and remains approximately constant. The contour length of the complexes exhibits no such discontinuous behavior. By fitting the relative increase of the contour length to the neighbor exclusion model, we obtain the exclusion number and the intrinsic intercalating constant of the psoralen-DNA interaction.
