    Construction of an expression vector for Lactococcus lactis based on an indigenous cryptic plasmid

    To construct an expression vector for Lactococcus lactis, the EmPMT fragment which contained the erythromycin resistance gene, P32 promoter, multiple cloning site (MCS) and terminator (T) was subcloned into the small cryptic plasmid pAR141. The resulting vector, designated as pAR1411, wasfound to be stably maintained in L. lactis MG1363 after transformation for at least 100 generations under non-selective conditions. The vector was also demonstrated to be able to express the gene coding for chloramphenicol acetyltransferase (cat) in L. lactis

    Screening for the optimal induction parameters for periplasmic producing interferon-&#945 2b in Escherichia coli

    Screening for optimum induction parameters to improve the production of periplasmic interferon-α2b (PrIFN-α2b) by recombinant Escherichia coli was conducted using shake flask culture. Recombinant E. coli Rosetta-gami 2(DE3) harboring the plasmid pET26b containing IFN-α2b gene under the control of the T7lac promoter was used, where the induction was accomplished by isopropyl β-D-1- thiogalactopyranoside (IPTG). The induction parameters (inducer concentration, point of induction, induction temperature and the length of induction) were analyzed to find the suitable range to be used for further optimization process. From the analysis, narrow range of induction temperature from 16 to 30°C and IPTG lower than 2 mM were found suitable for induction of PrIFN-α2b. On the other hand, early log phase was the preferred time to initiate the induction and the length of induction was dependent on the combination of other induction parameters used.Key words: Interferon-2b (IFN-2b), induction parameter, Escherichia coli, periplasm, shake flask culture

    Comparative study of methods for extraction and purification of environmental DNA from high-strength wastewater sludge

    DNA extraction from wastewater sludge (COD 50000 and BOD 25000 mg/l) was conducted using nine different methods normally used for environmental samples including a procedure used in this study and the results obtained were compared. The quality of the differently extracted DNAs was subsequently assessed by measuring humic acid concentration, cell lysis efficiency, polymerase chain reaction (PCR) amplification of methanogenic and eubacterial 16S rDNA. The protocol developed in this study was further evaluated by extracting DNA from various high-strength wastewater sludge samples, denaturing gradient gel electrophoresis (DGGE) and fluorescent in situ hybridization (FISH) analyses. The results revealed that great differences existed among the nine procedures and only a few produced satisfactory results when applied to high-strength wastewater sludge. Thermal shock alone was shown inefficient to disrupt the methanogenic cell wall to release the DNA.     Effect of promoter strength and signal sequence on the periplasmic expression of human interferon- &#9452b in Escherichia coli

    Two plasmids, pFLAG-ATS and pET 26b(+), were studied for the periplasmic expression of recombinant human interferon-2b (IFN-2b) in Escherichia coli. The pFLAG-ATS contains ompA signal sequence and tac promoter while pET 26b(+) contains pelB signal sequence and T7lac promoter. It was observedthat periplasmic expression of IFN-2b from pET 26b(+) was around 3000 times higher than pFLAGATS. Difference in the expression level was attributed to the difference in the promoters and the signal sequences. In silico analysis of mRNA secondary structures were analyzed using Vienna RNA packageand MFOLD. The results suggested that the increase of expression would mainly due to the difference in the translation initiation associated with secondary structure of mRNA transcribed by both plasmids

    Predicting Household Water Consumption Events: Towards a Personalised Recommender System to Encourage Water-conscious Behaviour

Recommender systems assist customers to make decisions; however, the modest adoption of digital technology in the water industry means no such system exists for household water users. Such a system for the water industry would suggest to consumers the most effective ways to conserve water based on their historical data from smart water meters. The advantage for water utilities in metropolitan areas is in managing demand, such as low pressure during peak hours or water shortages during drought. For customers, effective recommendations could save them money. This paper presents a novel vision of a recommender system prototype and discusses the benefits both for the consumers and the water utility companies. The success of this type of system would depend on the ability to anticipate the time of the next major water use so as to make useful, timely recommendations. Hence, the prototype is based on a long short-term memory (LSTM) neural network that predicts significant water consumption events (i.e., showers, baths, irrigation, etc.) for 83 households. The preliminary results show that LSTM is a useful method of prediction with an average root mean square error (RMSE) of 0.403. The analysis also provides indications of the scope of further research required for developing a commercially successful recommender system

    Corrosion Resistance of AA6063-Type Al-Mg-Si Alloy by Silicon Carbide in Sodium Chloride Solution for Marine Application

    The present work focused on corrosion inhibition of AA6063 type Al-Mg-Si alloy in sodium chloride (NaCl) solution with a silicon carbide inhibitor, using the potentiodynamic electrochemical method. The aluminium alloy surface morphology was examined, in the as-received and as-corroded in the un-inhibited state, with scanning electron microscopy equipped with energy dispersive spectroscopy (SEM-EDS). The results obtained via linear polarization indicated a high corrosion potential for the unprotected as-received alloy. Equally, inhibition efficiency as high as 98.82% at 10.0 g/v silicon carbide addition was obtained with increased polarization resistance (Rp), while the current density reduced significantly for inhibited samples compared to the un-inhibited aluminium alloy. The adsorption mechanism of the inhibitor aluminium alloy follows the Langmuir adsorption isotherm.     The Suitability of Photocatalyst Precursor Materials in Geopolymer Coating Applications: A Review

    Today, the building and construction sector demands environmentally friendly and sustainable protective coatings using inorganic coating materials for safe, non-hazardous, and great performance. Many researchers have been working on sustainable solutions to protect concrete and metal infrastructures against corrosion and surface deterioration with the intention of introducing green alternatives to conventional coatings. This article presents a review of developments of geopolymer pastes doped with different types of photocatalyst precursors including factors affecting geopolymer properties for enhancing coating with photocatalytic performance. Photodegradation using geopolymer photocatalyst has great potential for resolving harmless substances and removing pollutants when energized with ultraviolet (UV) light. Although geopolymer is a potentially new material with great properties, there has been less research focusing on the development of this coating. This study demonstrated that geopolymer binders are ideal precursor support materials for the synthesis of photocatalytic materials, with a significant potential for optimizing their distinctive properties

    Social disparities in the use of colonoscopy by primary care physicians in Ontario

    <p>Abstract</p> <p>Background</p> <p>It is unclear if all persons in Ontario have equal access to colonoscopy. This research was designed to describe long-term trends in the use of colonoscopy by primary care physicians (PCPs) in Ontario, and to determine whether PCP characteristics influence the use of colonoscopy.</p> <p>Methods</p> <p>We conducted a population-based retrospective study of PCPs in Ontario between the years 1996-2005. Using administrative data we identified a screen-eligible group of patients aged 50-74 years in Ontario. These patients were linked to the PCP who provided the most continuous care to them during each year. We determined the use of any colonoscopy among these patients. We calculated the rate of colonoscopy for each PCP as the number of patients undergoing colonoscopies per 100 screen eligible patients. Negative binomial regression was used to identify factors associated with the rate of colonoscopy, using generalized estimating equations to account for clustering of patients within PCPs.</p> <p>Results</p> <p>Between 7,955 and 8,419 PCPs in Ontario per year (median age 43 years) had at least 10 eligible patients in their practices. The use of colonoscopy by PCPs increased sharply in Ontario during the study period, from a median rate of 1.51 [inter quartile range (IQR) 0.57-2.62] per 100 screen eligible patients in 1996 to 4.71 (IQR 2.70-7.53) in 2005. There was substantial variation between PCPs in their use of colonoscopy. PCPs who were Canadian medical graduates and with more years of experience were more likely to use colonoscopy after adjusting for their patient characteristics. PCPs were more likely to use colonoscopy if their patient populations were predominantly women, older, had more illnesses, and if their patients resided in less marginalized neighborhoods (lower unemployment, fewer immigrants, higher income, higher education, and higher English/French fluency).</p> <p>Conclusions</p> <p>There is substantial variation in the use of colonoscopy by PCPs, and this variation has increased as the overall use of colonoscopy increased over time. PCPs whose patients were more marginalized were less likely to use colonoscopy, suggesting that there are inequities in access.</p

    Lifestyle physical activity among urban Palestinians and Israelis: a cross-sectional comparison in the Palestinian-Israeli Jerusalem risk factor study

    <p>Abstract</p> <p>Background</p> <p>Urban Palestinians have a high incidence of coronary heart disease, and alarming prevalences of obesity (particularly among women) and diabetes. An active lifestyle can help prevent these conditions. Little is known about the physical activity (PA) behavior of Palestinians. This study aimed to determine the prevalence of insufficient PA and its socio-demographic correlates among urban Palestinians in comparison with Israelis.</p> <p>Methods</p> <p>An age-sex stratified random sample of Palestinians and Israelis aged 25-74 years living in east and west Jerusalem was drawn from the Israel National Population Registry: 970 Palestinians and 712 Israelis participated. PA in a typical week was assessed by the Multi-Ethnic Study of Atherosclerosis (MESA) questionnaire. Energy expenditure (EE), calculated in metabolic equivalents (METs), was compared between groups for moderate to vigorous-intensity physical activity (MVPA), using the Wilcoxon rank-sum test, and for domain-specific prevalence rates of meeting public health guidelines and all-domain insufficient PA. Correlates of insufficient PA were assessed by multivariable logistic modeling.</p> <p>Results</p> <p>Palestinian men had the highest median of MVPA (4740 METs-min<sub>*</sub>wk<sup>-1</sup>) compared to Israeli men (2,205 METs-min<sub>*</sub>wk<sup>-1 </sup><it>p </it>< 0.0001), or to Palestinian and Israeli women, who had similar medians (2776 METs-min<sub>*</sub>wk<sup>-1</sup>). Two thirds (65%) of the total MVPA reported by Palestinian women were derived from domestic chores compared to 36% in Israeli women and 25% among Palestinian and Israeli men. A high proportion (63%) of Palestinian men met the PA recommendations by occupation/domestic activity, compared to 39% of Palestinian women and 37% of the Israelis. No leisure time PA was reported by 42% and 39% of Palestinian and Israeli men (<it>p </it>= 0.337) and 53% and 28% of Palestinian and Israeli women (<it>p </it>< 0.0001). Palestinian women reported the lowest level of walking. Considering all domains, 26% of Palestinian women were classified as insufficiently active versus 13% of Palestinian men (<it>p </it>< 0.0001) who did not differ from the Israeli sample (14%). Middle-aged and elderly and less educated Palestinian women, and unemployed and pensioned Palestinian men were at particularly high risk of inactivity. Socio-economic indicators only partially explained the ethnic disparity.</p> <p>Conclusions</p> <p>Substantial proportions of Palestinian women, and subgroups of Palestinian men, are insufficiently active. Culturally appropriate intervention strategies are warranted, particularly for this vulnerable population.</p
