Studies on rat ovarian receptors for lutropin (luteinizing hormone). Interaction with β-subunit of sheep lutropin

By using radioimmunoassay, the interaction of sheep lutropin (luteinizing hormone, LH) β-subunit with rat ovarian receptors was investigated. The binding of β-subunit was specific, although of much lower order than that of lutropin. Sheep lutropin β-subunit effectively inhibited the binding of human choriogonadotropin (chorionic gonadotropin, gCG) to the ovary, showing that both occupy the same sites. The binding of sheep lutropin β-subunit to ovary was not followed by any detectable increase in cyclic AMP. The ovarian response to lutropin in terms of cyclic AMP production was inhibited in the presence of free β-subunit. The α-subunit of lutropin, when used at concentrations where contamination with whole lutropin was negligible, enhanced the degree of binding of β-subunit; this did not lead to increased cyclic AMP in the tissue. Surprisingly, the binding of β-subunit in vitro was drastically decreased by the prior removal of all endogenous rat lutropin bound to receptors. The implications of these data are discussed in the light of the reported biological activity of the β-subunit

Ability of human chorionic gonadotropin β-subunit to inhibit the steroidogenic response to lutropin

Ability of the β-subunit of human chorionic gonadotropin to inhibit the response to lutropin (luteinizing hormone, LH) was tested in the immature rat ovarian system and pregnant-mare-serum-gonadotropin-primed rat ovarian system with progesterone production being used as the response. Human chorionic gonadotropin β-subunit was found to inhibit human and ovine lutropin-stimulated progesterone production. At a constant dose of lutropin, inhibition was dependent on the concentration of β-subunit. When concentration of the β-subunit was kept constant at 5.0 μg/ml and the concentration of lutropin was varied, the inhibition was maximum at the saturating concentration of the native hormone. The α-subunit of the human chorionic gonadotropin did not inhibit the response to lutropin. The lutropin/β-subunit ratio required to produce an inhibition of response was much lower than that required to bring about an observable inhibition of binding

Performance Comparison of Straight and Curved Diffusers

    Get PDF
Experimental studies have been carried out to compare the performance of two dimensional straight and curved diffusers of same area ratio and effective divergence angle in the Reynolds number range of 7.8 X 10(5) to 1.29 X 10(6). Free stream turbulence effects have also been studied at the increased turbulence level to 3.4 per cent. The results indicate that straight diffuser pressure recovery is slightly higher as compared to the curved diffusers. However, stream turbulence, which improves the pressure recovery in both cases, has been observed to have greater effect in case of curved diffuser. Boundary layer velocity profiles on the diffuser surfaces have also been presented at various streamwise stations. It is observedthat the growth of inner surface boundary layer has a major effects on losses in case of a curved diffuser

CPK: The new tool in the diagnosis of ectopic pregnancy

    Get PDF
Background: Ectopic pregnancy is still a diagnostic dilemma presenting with various complaints. The classic triad of amenorrhea, abdominal pain, vaginal bleeding and /or syncope is not always seen. Misdiagnosis can lead to delay in treatment, blood loss is found to be the major cause of death. Early and accurate diagnosis is critical in bringing down the maternal mortality and morbidity. Prompt and effective treatment of an ectopic pregnancy can help preserve the chances of future healthy pregnancies. Aim of present study was to investigate whether creatinine phosphokinase (cpk) can be used as an effective diagnostic tool in the early diagnosis of ectopic pregnancy which can help in decreasing the maternal mortality and morbidityMethods: This observational comparative three group clinical study was conducted at Chinmaya Mission Hospital, Bangalore, between May 2016 to January 2017.120 women in their early trimester were studied of which 40 were diagnosed cases of ectopic pregnancies, 40 women presented with intrauterine abortive pregnancies and 40 women had normal healthy pregnancies. Serum CPK, serum B-HCG, vaginal scans were done in all, along with routine investigations. Results: The mean CPK values in normal, abortive and ectopic pregnancies were 36.92±6.44, 43.95±11.96 and 91.55±30.43 respectively. It was found to be significantly higher in ectopic Pregnancies. Also, the mean CPK in ruptured and unruptured ectopic pregnancy were 97.26±25.97 and 63.82±34.92 respectively. Conclusions: Present study shows that maternal CPK levels are significantly higher in women with ectopic pregnancies. CPK can serve as the reliable biochemical marker to diagnose ectopic pregnancy particularly ruptured. CPK can be used to increase the diagnostic efficacy in ectopic pregnancy, which followed by rapid and appropriate treatment can reduce the mortality, morbidity and preserve future fertility

YBCO-buffered NdBCO film with higher thermal stability in seeding REBCO Growth

In this work, we report a strengthened superheating effect caused by a buffering YBa2Cu3Oy (Y123 or YBCO) layer in the Nd1+xBa2-xCu3O7-y (Nd123 or NdBCO) thin film with MgO substrate (i.e., NdBCO/YBCO/MgO thin film). In the cold-seeding melt-textured (MT) growth, the NdBCO/YBCO/MgO film presented an even higher superheating level, about 20 {\deg}C higher than that of non-buffered NdBCO film (i.e., NdBCO/MgO film). Using this NdBCO/YBCO/MgO film as seeds and undergoing a maximum processing temperature (Tmax) up to 1120 {\deg}C, we succeeded in growing various RE1+xBa2-xCu3O7-y (REBCO, RE=rare elements) bulk superconductors, including Gd1+xBa2-xCu3O7-y (GdBCO), Sm1+xBa2-xCu3O7-y (SmBCO) and NdBCO that have high peritectic temperatures (Tp). The pole figure (X-Ray \phi-scan) measurement reveals that the NdBCO/YBCO/MgO film has better in-plane alignment than the NdBCO/MgO film, indicating that the induced intermediate layer improves the crystallinity of the NdBCO film, which could be the main origin of the enhanced thermal stability. In short, possessing higher thermal stability and enduring a higher Tmax in the MT process, the NdBCO/YBCO/MgO film is beneficial to the growth of bulk superconductors in two aspects: (1) broad application for high-Tp REBCO materials; (2) effective suppression against heterogeneous nucleation, which is of great assistance in growing large and high-performance REBCO crystals.Comment: 9 pages, 4 figure
