    Effects of high-protein intake on bone turnover in long-term bed rest in women

    Bed rest (BR) causes bone loss, even in otherwise healthy subjects. Several studies suggest that ambulatory subjects may benefit from high-protein intake to stimulate protein synthesis and to maintain muscle mass. However, increasing protein intake above the recommended daily intake without adequate calcium and potassium intake may increase bone resorption. We hypothesized that a regimen of high-protein intake (HiPROT), applied in an isocaloric manner during BR, with calcium and potassium intake meeting recommended values, would prevent any effect of BR on bone turnover. After a 20-day ambulatory adaptation to a controlled environment, 16 women participated in a 60-day, 6\ub0 head-down-tilt (HDT) BR and were assigned randomly to 1 of 2 groups. Control (CON) subjects (n = 8) received 1 g/(kg body mass\ub7day)-1 dietary protein. HiPROT subjects (n = 8) received 1.45 g protein/(kg body mass\ub7day)-1 plus an additional 0.72 g branched-chain amino acids per day during BR. All subjects received an individually tailored diet (before HDTBR: 1888 \ub1 98 kcal/day; during HDTBR: 1604 \ub1 125 kcal/day; after HDTBR: 1900 \ub1 262 kcal/day), with the CON group's diet being higher in fat and carbohydrate intake. High-protein intake exacerbated the BR-induced increase in bone resorption marker C-telopeptide (>30%) (p < 0.001) by the end of BR. Bone formation markers were unaffected by BR and high-protein intake. We conclude that high-protein intake in BR might increase bone loss. Further long-duration studies are mandatory to show how the positive effect of protein on muscle mass can be maintained without the risk of reducing bone mineral density

    Thermal decomposition of allantoin as probed by matrix isolation FTIR spectroscopy

    The optimized geometries, energies of the possible conformers of allantoin (2,5-dioxo-4-imidazolidinyl urea, the diureide of glyoxylic acid) as well as the barriers for conformational interconversion have been calculated using the density functional theory [DFT(B3LYP)/6-311++G(d,p)] method. The calculations predicted the existence of four conformers (gC, tT, gâ€ČC, and gâ€ČT; where the first and second symbols in the name of the conformers designate the conformation around the exocyclic NHC–NHCO and CNH–CO axes, respectively), with the gC form contributing to more than 98% of the population in gas phase at room temperature. This conformer is different from that corresponding to the monomeric unit found in crystalline RS-allantoin (gâ€ČC; Mootz, D. Acta Crystallogr.1965, 19, 726), stressing the importance of intermolecular H-bonding in determining the structure of the crystal. Upon sublimation under vacuum (10−6 mbar), the compound was found to undergo extensive decomposition to urea, isocyanic acid, NH3, and carbon. The identification of the decomposition products was made by using matrix isolation infrared spectroscopy. In consonance with the theoretical predictions, the allantoin molecules surviving thermal decomposition were found to undergo conformational isomerization and be present in the cryogenic argon matrix in both the gC and gâ€ČC conformations.     Entgegen der These der Auflösungserscheinungen des BĂŒrgertums stellt der Autor die Annahme auf den PrĂŒfstand, dass wir es nach wie vor mit gesellschaftlichen Fraktionierungen bĂŒrgerlicher Lebensweisen zu tun haben. Am Beispiel autobiographischer Schriften von deutschen Topmanagern stellt der Text ein modernes VerstĂ€ndnis des WirtschaftsbĂŒrgertums vor, das organisational (durch die Karrieremechanismen der Organisation) und institutionell (im Feld der Wirtschaft) verankert ist. Die moderne Sozialformation des WirtschaftsbĂŒrgertums ist nur noch auf der Grundlage von Organisationen denkbar. Sie lĂ€sst sich, jenseits von Klasse und Stand, als Positionselite beschreiben. Anhand der Autobiographien lĂ€sst sich die Reproduktion dieser Elite auf Basis einer engen VerknĂŒpfung zwischen familialer Herkunft, an organisationale Karrieren gebundene Leistungsbereitschaft und hoher formaler Bildung nachzeichnen. Die Abgrenzung in der Statusreproduktion zwischen Bildungs- und WirtschaftsbĂŒrgertum weist der Autor am jeweiligen VerhĂ€ltnis zur Bildung nach; zwar können beide einen hohen Bildungsgrad in Form von Bildungspatenten nachweisen, doch im Falle des WirtschaftsbĂŒrgertums herrscht ein instrumentelles VerhĂ€ltnis zur Bildung vor. Der hohe Bildungsgrad folgt hier dem BedĂŒrfnis, den Status mittels formaler Bildung abzusichern und damit die Gefahr der eigenen Austauschbarkeit - als Personal der Organisation - zu kompensieren. Der Text macht außerdem generationale Effekte sichtbar; insbesondere indem er darlegt, inwieweit der "moderne Manager" einerseits in der Betonung seines Status seinen VorgĂ€ngern gleicht und sich doch gleichzeitig in der Art der UnternehmensfĂŒhrung abgrenzt - indem er bspw. die Managementkonzepte seiner Zeit aufgreift

    Informal and formal reconciliation strategies of older peoples’ working carers: the European carers@work project

    Faced with a historically unprecedented process of demographic ageing, many European societies implemented pension reforms in recent years to extend working lives. Although aimed at rebalancing public pension systems, this approach has the unintended side effect that it also extends the number of years in which working carers have to juggle the conflicting demands of employment and caregiving. This not only impinges on working carers’ well-being and ability to continue providing care but also affects European enterprises’ capacity to generate growth which increasingly relies on ageing workforces. The focus of this paper will thus be a cross-national comparison of individual reconciliation strategies and workplace-related company policies aimed at enabling working carers to reconcile both conflicting roles in four different European welfare states: Germany, Italy, Poland, and the United Kingdom

    In-Vivo Expression Profiling of Pseudomonas aeruginosa Infections Reveals Niche-Specific and Strain-Independent Transcriptional Programs

    Pseudomonas aeruginosa is a threatening, opportunistic pathogen causing disease in immunocompromised individuals. The hallmark of P. aeruginosa virulence is its multi-factorial and combinatorial nature. It renders such bacteria infectious for many organisms and it is often resistant to antibiotics. To gain insights into the physiology of P. aeruginosa during infection, we assessed the transcriptional programs of three different P. aeruginosa strains directly after isolation from burn wounds of humans. We compared the programs to those of the same strains using two infection models: a plant model, which consisted of the infection of the midrib of lettuce leaves, and a murine tumor model, which was obtained by infection of mice with an induced tumor in the abdomen. All control conditions of P. aeruginosa cells growing in suspension and as a biofilm were added to the analysis. We found that these different P. aeruginosa strains express a pool of distinct genetic traits that are activated under particular infection conditions regardless of their genetic variability. The knowledge herein generated will advance our understanding of P. aeruginosa virulence and provide valuable cues for the definition of prospective targets to develop novel intervention strategies
