Tetrazine-Triggered Release of Carboxylic-Acid-Containing Molecules for Activation of an Anti-inflammatory Drug.
In addition to its use for the study of biomolecules in living systems, bioorthogonal chemistry has emerged as a promising strategy to enable protein or drug activation in a spatially and temporally controlled manner. This study demonstrates the application of a bioorthogonal inverse electron-demand Diels-Alder (iEDDA) reaction to cleave trans-cyclooctene (TCO) and vinyl protecting groups from carboxylic acid-containing molecules. The tetrazine-mediated decaging reaction proceeded under biocompatible conditions with fast reaction kinetics (<2 min). The anti-inflammatory activity of ketoprofen was successfully reinstated after decaging of the nontoxic TCOprodrug in live macrophages. Overall, this work expands the scope of functional groups and the application of decaging reactions to a new class of drugs
Mindfulness, Acceptance and Catastrophizing in Chronic Pain
OBJECTIVES: Catastrophizing is often the primary target of the cognitive-behavioral treatment of chronic pain. Recent literature on acceptance and commitment therapy (ACT) suggests an important role in the pain experience for the concepts mindfulness and acceptance. The aim of this study is to examine the influence of mindfulness and general psychological acceptance on pain-related catastrophizing in patients with chronic pain. METHODS: A cross-sectional survey was conducted, including 87 chronic pain patients from an academic outpatient pain center. RESULTS: The results show that general psychological acceptance (measured with the AAQ-II) is a strong predictor of pain-related catastrophizing, independent of gender, age and pain intensity. Mindfulness (measured with the MAAS) did not predict levels of pain-related catastrophizing. DISCUSSION: Acceptance of psychological experiences outside of pain itself is related to catastrophizing. Thus, acceptance seems to play a role in the pain experience and should be part of the treatment of chronic pain. The focus of the ACT treatment of chronic pain does not necessarily have to be on acceptance of pain per se, but may be aimed at acceptance of unwanted experiences in general. Mindfulness in the sense of "acting with awareness" is however not related to catastrophizing. Based on our research findings in comparisons with those of other authors, we recommend a broader conceptualization of mindfulness and the use of a multifaceted questionnaire for mindfulness instead of the unidimensional MAAS

Strange Quark Contributions to Parity-Violating Asymmetries in the Backward Angle G0 Electron Scattering Experiment
We have measured parity-violating asymmetries in elastic electron-proton and
quasi-elastic electron-deuteron scattering at Q^2 = 0.22 and 0.63 GeV^2. They
are sensitive to strange quark contributions to currents in the nucleon, and to
the nucleon axial current. The results indicate strange quark contributions of
< 10% of the charge and magnetic nucleon form factors at these four-momentum
transfers. We also present the first measurement of anapole moment effects in
the axial current at these four-momentum transfers.Comment: 5 pages, 2 figures, changed references, typo, and conten
The G0 Experiment: Apparatus for Parity-Violating Electron Scattering Measurements at Forward and Backward Angles
In the G0 experiment, performed at Jefferson Lab, the parity-violating
elastic scattering of electrons from protons and quasi-elastic scattering from
deuterons is measured in order to determine the neutral weak currents of the
nucleon. Asymmetries as small as 1 part per million in the scattering of a
polarized electron beam are determined using a dedicated apparatus. It consists
of specialized beam-monitoring and control systems, a cryogenic hydrogen (or
deuterium) target, and a superconducting, toroidal magnetic spectrometer
equipped with plastic scintillation and aerogel Cerenkov detectors, as well as
fast readout electronics for the measurement of individual events. The overall
design and performance of this experimental system is discussed.Comment: Submitted to Nuclear Instruments and Method
Transverse Beam Spin Asymmetries at Backward Angles in Elastic Electron-Proton and Quasi-elastic Electron-Deuteron Scattering
We have measured the beam-normal single-spin asymmetries in elastic
scattering of transversely polarized electrons from the proton, and performed
the first measurement in quasi-elastic scattering on the deuteron, at backward
angles (lab scattering angle of 108 degrees) for Q2 = 0.22 GeV^2/c^2 and 0.63
GeV^2/c^2 at beam energies of 362 MeV and 687 MeV, respectively. The asymmetry
arises due to the imaginary part of the interference of the two-photon exchange
amplitude with that of single photon exchange. Results for the proton are
consistent with a model calculation which includes inelastic intermediate
hadronic (piN) states. An estimate of the beam-normal single-spin asymmetry for
the scattering from the neutron is made using a quasi-static deuterium
approximation, and is also in agreement with theory
Falls in young, middle-aged and older community dwelling adults: perceived cause, environmental factors and injury
BACKGROUND: Falls in older people have been characterized extensively in the literature, however little has been reported regarding falls in middle-aged and younger adults. The objective of this paper is to describe the perceived cause, environmental influences and resultant injuries of falls in 1497 young (20–45 years), middle-aged (46–65 years) and older (> 65 years) men and women from the Baltimore Longitudinal Study on Aging. METHODS: A descriptive study where participants completed a fall history questionnaire describing the circumstances surrounding falls in the previous two years. RESULTS: The reporting of falls increased with age from 18% in young, to 21% in middle-aged and 35% in older adults, with higher rates in women than men. Ambulation was cited as the cause of the fall most frequently in all gender and age groups. Our population reported a higher percentage of injuries (70.5%) than previous studies. The young group reported injuries most frequently to wrist/hand, knees and ankles; the middle-aged to their knees and the older group to their head and knees. Women reported a higher percentage of injuries in all age groups. CONCLUSION: This is the first study to compare falls in young, middle and older aged men and women. Significant differences were found between the three age groups with respect to number of falls, activities engaged in prior to falling, perceived causes of the fall and where they fell
Indirect measurements of neutron-induced reaction cross sections at storage rings
Neutron-induced reaction cross sections of unstable nuclei are essential for understanding the synthesis of heavy elements in stars. However, their measurement is very difficult due to the radioactivity of the targets involved. We propose to circumvent this problem by using for the first time the surrogate reaction method in inverse kinematics at heavy-ion storage rings. In this contribution, we describe the developments we have done to perform surrogate-reaction studies at the storage rings of GSI/FAIR. In particular, we present the first results of the proof of principle experiment, which we conducted recently at the Experimental Storage Ring (ESR)
Site-selective protein-modification chemistry for basic biology and drug development.
Nature has produced intricate machinery to covalently diversify the structure of proteins after their synthesis in the ribosome. In an attempt to mimic nature, chemists have developed a large set of reactions that enable post-expression modification of proteins at pre-determined sites. These reactions are now used to selectively install particular modifications on proteins for many biological and therapeutic applications. For example, they provide an opportunity to install post-translational modifications on proteins to determine their exact biological roles. Labelling of proteins in live cells with fluorescent dyes allows protein uptake and intracellular trafficking to be tracked and also enables physiological parameters to be measured optically. Through the conjugation of potent cytotoxicants to antibodies, novel anti-cancer drugs with improved efficacy and reduced side effects may be obtained. In this Perspective, we highlight the most exciting current and future applications of chemical site-selective protein modification and consider which hurdles still need to be overcome for more widespread use.We thank FCT Portugal (FCT Investigator to G.J.L.B.), the EU (Marie-Curie CIG to G.J.L.B. and Marie-Curie IEF to O.B.) and the EPSRC for funding. Measurement of the Parity-Violating Asymmetry in Inclusive Electroproduction of π- near the Δ0 Resonance
The parity-violating (PV) asymmetry of inclusive π-production in electron scattering from a liquid deuterium target was measured at backward angles. The measurement was conducted as a part of the G0 experiment, at a beam energy of 360 MeV. The physics process dominating pion production for these kinematics is quasifree photoproduction off the neutron via the Δ0 resonance. In the context of heavy-baryon chiral perturbation theory, this asymmetry is related to a low-energy constant dΔ- that characterizes the parity-violating γNΔ coupling. Zhu et al. calculated dΔ- in a model benchmarked by the large asymmetries seen in hyperon weak radiative decays, and predicted potentially large asymmetries for this process, ranging from Aγ-=-5.2 to +5.2  ppm. The measurement performed in this work leads to Aγ-=-0.36±1.06±0.37±0.03  ppm (where sources of statistical, systematic and theoretical uncertainties are included), which would disfavor enchancements considered by Zhu et al. proportional to Vud/Vus. The measurement is part of a program of inelastic scattering measurements that were conducted by the G0 experiment, seeking to determine the N-Δ axial transition form factors using PV electron scattering
