    Abstrak: Kajian Skala Ruang Kota d/h Dalam Perspektif Soundscapes (Studi Kasus Kawasan Pusat Kota Manado). Bunyi hanyalah satu komponen dari pengalaman orang-orang di tempat tersebut : ada juga pengalaman visual, suhu, angin, vegetasi, bahan yang berbeda dari permukaan lantai, keamanan fisik dari tempat itu, kegiatan mereka sendiri dan kegiatan orang lain terlihat jelas di tempat ini. Baku kebisingan yang ada di Indonesia ialah baku tentang tingkat kebisingan dalam kawasan dan lingkungan menurut Keputusan Menteri Negara Lingkungan Hidup Nomor : Kep-48/Menlh/11/1996. Tujuan penelitian ini untuk menemukan nilai kebisingan dan dimensi skala d/h agar kenyamanan bunyi dapat dirasakan dalam ruang kota. Penelitian dilaksanakan di pusat kota Manado tepatnya di kawasan Taman Kesatuan Bangsa, dengan menggunakan mix methods melalui rumus LAeq 10 menit dan Leq Siang, untuk mendapatkan baku kebisingan dalam kawasan. Hasil penelitian menemukan jika nilai d/h semakin kecil maka akan terasa semakin bising suatu kawasan dan untuk mengimbangi kesan bising suatu kawasan agar terasa normal dan nyaman dilakukan dengan cara membuat perbandingan d/h agar sama dengan nilai 4. Kata Kunci : Kebisingan, Kenyamanan, Skala d/h. Abstract: Study of Urban Spatial Scale d/h in Soundscapes Perspective (Case Study of Manado City Center Area). Sound is only one component of the experience of people in the place: there is also the visual experience, temperature, wind, vegetation, different materials of the floor surface, the physical safety of the place, their activities, and the activities of others visible in this place. The existing noise standard in Indonesia is the standard for noise levels in the area and the environment according to the Decree of the State Minister of the Environment Number: Kep-48/Menlh/11/1996. The purpose of this study was to find the noise value and the dimensions of the d/h scale so that the comfort of sound can be felt in urban spaces. The research was carried out in the center of Manado city, precisely in the National Unity Park area, using mixed methods using the LAeq 10 minutes formula and Leq Siang, to get the noise standard in the area. The results of the study found that if the d/h value is smaller, the area will feel noisier, and to compensate for the noisy impression of an area so that it feels normal and comfortable, it is done by making a d/h comparison to be equal to a value of 4.  Keywords: Noise, Comfort, d/h Scale &nbsp

    Induced pluripotent stem cells from hair follicles as a cellular model for neurodevelopmental disorders

    AbstractDisease-specific induced pluripotent stem cells (iPSC) allow unprecedented experimental platforms for basic research as well as high-throughput screening. This may be particularly relevant for neuropsychiatric disorders, in which the affected neuronal cells are not accessible. Keratinocytes isolated from hair follicles are an ideal source of patients' cells for reprogramming, due to their non-invasive accessibility and their common neuroectodermal origin with neurons, which can be important for potential epigenetic memory. From a small number of plucked human hair follicles obtained from two healthy donors we reprogrammed keratinocytes to pluripotent iPSC. We further differentiated these hair follicle-derived iPSC to neural progenitors, forebrain neurons and functional dopaminergic neurons.This study shows that human hair follicle-derived iPSC can be differentiated into various neural lineages, suggesting this experimental system as a promising in vitro model to study normal and pathological neural developments, avoiding the invasiveness of commonly used skin biopsies

    TRANSFORMASI SPASIAL PENGGUNAAN LAHAN KECAMATAN MODAYAG - MODAYAG BARAT KABUPATEN BOLAANG MONGONDOW TIMUR 2013-2019

    Abstrak: Transformasi Spasial penggunaan lahan di Kecamatan Modayag-Modayag Barat Kabupaten Bolaang Mongondow Timur, mengalami perubahan yang cukup signifikan pada selang 2013 – 2019. Salah satu hal yang memicu terjadinya hal ini adalah posisi Kecamatan Modayag-Modayag Barat yang berbatasan dengan beberapa Kabupaten dan Kota di Provinsi Sulawesi Utara, diantaranya Kabupaten Bolaang Mongondow, Kotamobagu dan Kabupaten Minahasa Selatan, sehingga memiliki daya tarik wilayah tersendiri. Tujuan dari Penelitian ini untuk menganalisis dan mengidentifikasi lahan yang mengalami transformasi dan yang mengalami penambahan luasan terbesar, termasuk mengidentifikasi lahan apa yang terkonversi menjadi permukiman. Data yang digunakan adalah shapefile Tutupan lahan tahun 2013 dan shapefile tutupan lahan tahun 2019. Metode penelitian yang digunakan kuantitatif lewat pendekatan analisis spasial GIS menggunakan ArcGis versi 8.0, dan tools pendukung analisis perubahan lahan. Penelitian ini dapat disimpulkan : 1). Konversi lahan terbesar dan mengalami peningkatan terjadi pada kebun campuran ke hutan sebesar 452 Ha atau 46%, kebun campuran ke perkebunan sebesar 188,45 Ha atau sebesar 19%, dan Kebun Campuran ke Tegalan/ladang sebesar 161,68 Ha atau sebesar 19%, 2). Konversi lahan permukiman mengalami peningkatan dan mengurangi luasan danau 0,01 Ha atau atau 0,002%, Hutan Lahan Kering sebesar 1,13 Ha atau 0,009%, Perkebunan sebesar 45,4 Ha atau 1,40%, dan juga sudah mengkonversi lahan sawah 5,04 Ha atau 1,87%. Kata kunci: Transformasi Spasial; Konversi Lahan; GIS Abstract: Spatial transformation of land use in the Modayag-Modayag Barat District, Bolaang Mongondow Timur Regency, underwent a significant change between 2013 - 2019. One of the things that triggered this was the position of the Modayag-Modayag Barat District which borders several regencies and cities in North Sulawesi Province, including Bolaang Mongondow Regency, Kotamobagu City and South Minahasa Regency, so that they have their own regional appeal. The purpose of this study is to analyze and identify land that is undergoing transformation and which has experienced the largest increase in area, including identifying what lands have been converted into settlements. The data used are the 2013 land cover shapefile and the 2019 land cover shapefile. The research method used is quantitative through a GIS spatial analysis approach using ArcGIS version 8.0, and supporting tools for land change analysis. This research can be concluded: 1). The largest land conversion and an increase occurred in mixed gardens to forest by 452 ha or 46%, mixed gardens to plantations by 188.45 ha or 19%, and mixed gardens to dry fields/fields by 161.68 ha or 19%. 2). Residential land conversion has increased and reduced the lake area by 0.01 Ha or or 0.002%, Dry Land Forest by 1.13 Ha or 0.009%, Plantation by 45.4 Ha or 1.40%, and has also converted rice fields 5, 04 Ha or 1.87%.     Paraoxonase Activity and Expression Is Modulated by Therapeutics in Experimental Rat Nonalcoholic Fatty Liver Disease

    Objective. The objective of the present study is to investigate the effect of rosiglitazone, metformin, ezetimibe, and valsartan (alone or in combinations) on paraoxonase (PON) activity and PON-mRNA expression in nonalcoholic fatty liver disease (NAFLD). Methods. 54 Male Sprague–Dawley rats were divided to 9 groups: chow diet group (15 weeks); methionine-choline-deficient diet (MCDD) group (15 weeks); MCDD-treated groups for the last 6 weeks with either metformin (M), rosiglitazone (R), metformin plus rosiglitazone (M+R), ezetimibe (E), valsartan (V), or a combination of R+M+V or of R+M+V+E for a total period of 15 weeks. Results. PON activities in serum and liver were decreased in MCDD rats. PON activity in serum increased significantly in all treatment groups. PON activity in liver was also increased significantly, except only in groups R, E, V, R+M+V, and R+M+V+E. Liver PON3 mRNA expression increased significantly in groups R+M, E, V, R+M+V, and R+M+V+E whereas liver PON2 mRNA expression increased significantly in MCDD, R+M, E, V, R+M+V, and R+M+V+E. Conclusions. PON activities in serum and liver were decreased in NAFLD. Treatment with insulin sensitizers, ezetimibe, and valsartan increased PON activity and reduced oxidative stress both in serum and liver

    Design Impact on Airflow Patterns inΒ Fluidization Units

    The airflow behavior in a fluidization unit was integrally studied by means of experimental work and computational fluid dynamics simulation. The computational domain included the gas inlet pipe, plenum, perforated plate, fluidization chamber, and air outlet pipe. Different scenarios were simulated to allow distinguishing the best way to represent perforated-plate distributors and elucidate the impact of the grid design on the fluidization performance. The simulated pressure drop across the distributor and the plenum flow pattern were in concordance with the experimental data. It was found that the distance between the peripheral holes and walls has a great impact on the airflow downstream the distributor.Fil: Renaudo, Carlos Alberto.     Sp1 Expression Is Disrupted in Schizophrenia; A Possible Mechanism for the Abnormal Expression of Mitochondrial Complex I Genes, NDUFV1 and NDUFV2

    The prevailing hypothesis regards schizophrenia as a polygenic disease, in which multiple genes combine with each other and with environmental stimuli to produce the variance of its clinical symptoms. We investigated whether the ubiquitous transcription factor Sp1 is abnormally expressed in schizophrenia, and consequently can affect the expression of genes implicated in this disorder. promoter by binding to its three GC-boxes. Both activation and binding were inhibited by mithramycin.These findings suggest that abnormality in Sp1, which can be the main activator/repressor or act in combination with additional transcription factors and is subjected to environmental stimuli, can contribute to the polygenic and clinically heterogeneous nature of schizophrenia

    Seasonality of Influenza A(H3N2) Virus: A Hong Kong Perspective (1997–2006)

    BACKGROUND: The underlying basis for the seasonality of influenza A viruses is still uncertain. Phylogenetic studies investigated this phenomenon but have lacked sequences from more subtropical and tropical regions, particularly from Southeast Asia. METHODOLOGY/PRINCIPAL FINDINGS: 281 complete hemagglutinin (HA) and neuraminidase (NA) sequences were obtained from influenza A(H3N2) viruses, collected over 10 years (1997-2006) from Hong Kong. These dated sequences were analyzed with influenza A(H3N2) vaccine strain sequences (Syd/5/97, Mos/10/99, Fuj/411/02, Cal/7/04) and 315 other publicly available dated sequences from elsewhere, worldwide. In addition, the NA sequence alignment was inspected for the presence of any naturally occurring, known, neuraminidase inhibitor (NAI) resistance-associated amino acid mutations (R292K and E119V). Before 2001, the Hong Kong HA and NA sequences clustered more closely with the older vaccine sequences (Syd/5/97, Mos/10/99) than did sequences from elsewhere. After 2001, this trend reversed with significant clusters containing HA and NA sequences from different locations, isolated at different times, suggesting that viral migration may account for much of the influenza A(H3N2) seasonality during this 10-year period. However, at least one example from Hong Kong was found suggesting that in some years, influenza A(H3N2) viruses may persist in the same location, perhaps continuing to circulate, sub-clinically, at low levels between seasons, to re-emerge in the influenza season the following year, relatively unchanged. None of these Hong Kong influenza A(H3N2) NA sequences contained any of the known NAI-resistance associated mutations. CONCLUSIONS/SIGNIFICANCE: The seasonality of influenza A(H3N2) may be largely due to global migration, with similar viruses appearing in different countries at different times. However, occasionally, some viruses may remain within a single location and continue to circulate within that population, to re-emerge during the next influenza season, with relatively little genetic change. Naturally occurring NAI resistance mutations were absent or, at least, very rare in this population

    Neuroanatomical Pattern of Mitochondrial Complex I Pathology Varies between Schizophrenia, Bipolar Disorder and Major Depression

    BACKGROUND:Mitochondrial dysfunction was reported in schizophrenia, bipolar disorderand major depression. The present study investigated whether mitochondrial complex I abnormalities show disease-specific characteristics. METHODOLOGY/PRINCIPAL FINDINGS:mRNA and protein levels of complex I subunits NDUFV1, NDUFV2 and NADUFS1, were assessed in striatal and lateral cerebellar hemisphere postmortem specimens and analyzed together with our previous data from prefrontal and parieto-occipital cortices specimens of patients with schizophrenia, bipolar disorder, major depression and healthy subjects. A disease-specific anatomical pattern in complex I subunits alterations was found. Schizophrenia-specific reductions were observed in the prefrontal cortex and in the striatum. The depressed group showed consistent reductions in all three subunits in the cerebellum. The bipolar group, however, showed increased expression in the parieto-occipital cortex, similar to those observed in schizophrenia, and reductions in the cerebellum, yet less consistent than the depressed group. CONCLUSIONS/SIGNIFICANCE:These results suggest that the neuroanatomical pattern of complex I pathology parallels the diversity and similarities in clinical symptoms of these mental disorders
