    Alexander quandle lower bounds for link genera

    We denote by Q_F the family of the Alexander quandle structures supported by finite fields. For every k-component oriented link L, every partition P of L into h:=|P| sublinks, and every labelling z of such a partition by the natural numbers z_1,...,z_n, the number of X-colorings of any diagram of (L,z) is a well-defined invariant of (L,P), of the form q^(a_X(L,P,z)+1) for some natural number a_X(L,P,z). Letting X and z vary in Q_F and among the labellings of P, we define a derived invariant A_Q(L,P)=sup a_X(L,P,z). If P_M is such that |P_M|=k, we show that A_Q(L,P_M) is a lower bound for t(L), where t(L) is the tunnel number of L. If P is a "boundary partition" of L and g(L,P) denotes the infimum among the sums of the genera of a system of disjoint Seifert surfaces for the L_j's, then we show that A_Q(L,P) is at most 2g(L,P)+2k-|P|-1. We set A_Q(L):=A_Q(L,P_m), where |P_m|=1. By elaborating on a suitable version of a result by Inoue, we show that when L=K is a knot then A_Q(K) is bounded above by A(K), where A(K) is the breadth of the Alexander polynomial of K. However, for every g we exhibit examples of genus-g knots having the same Alexander polynomial but different quandle invariants A_Q. Moreover, in such examples A_Q provides sharp lower bounds for the genera of the knots. On the other hand, A_Q(L) can give better lower bounds on the genus than A(L), when L has at least two components. We show that in order to compute A_Q(L) it is enough to consider only colorings with respect to the constant labelling z=1. In the case when L=K is a knot, if either A_Q(K)=A(K) or A_Q(K) provides a sharp lower bound for the knot genus, or if A_Q(K)=1, then A_Q(K) can be realized by means of the proper subfamily of quandles X=(F_p,*), where p varies among the odd prime numbers.Comment: 36 pages; 16 figure

    The Impact of Exploring Computer Science in Wisconsin: Where Disadvantage is an Advantage

    Assessing the impact of regional or statewide interventions in primary and secondary school (K-12) computer science (CS) education is difficult for a variety of reasons. Qualitative survey data provide only a limited view of impacts, but quantitative data can be notoriously difficult to acquire at scale from large numbers of classrooms, schools, or local educational authorities. In this paper, we use several publicly available data sources to glean insights into public high school CS enrollments across an entire U.S. state. Course enrollments with NCES course codes and local descriptors, school-level demographic data, and school geographic attendance boundaries can be combined to highlight where CS offerings persist and thrive, how CS enrollments change over time, and the ultimate quantitative impact of a statewide intervention. We propose a more appropriate level of data aggregation for these types of quantitative studies than has been undertaken in previous work while demonstrating the importance of a contextual aggregation process. The results of our disparate impact analysis for the first time quantify the impact of a statewide Exploring Computer Science (ECS) program rollout on economic groups across the region. Our blueprint for this analysis can serve as a template to guide and assess large-scale K-12 CS interventions wherever detailed project evaluation methods cannot scale to encompass the entire study area, especially in cases where attribute heterogeneity is a significant issue

    Background Vibrio anguillarum is an extracellular bacterial pathogen that is a causative agent of vibriosis in finfish and crustaceans with mortality rates ranging from 30% to 100%. Mutations in central metabolism (glycolysis and the TCA cycle) of intracellular pathogens often result in attenuated virulence due to depletion of required metabolic intermediates; however, it was not known whether mutations in central metabolism would affect virulence in an extracellular pathogen such as V. anguillarum. Results Seven central metabolism mutants were created and characterized with regard to growth in minimal and complex media, expression of virulence genes, and virulence in juvenile rainbow trout (Oncorhynchus mykiss). Only the isocitrate dehydrogenase (icd) mutant was attenuated in virulence against rainbow trout challenged by either intraperitoneal injection or immersion. Further, the icd mutant was shown to be immunoprotective against wild type V. anguillarum infection. There was no significant decrease in the expression of the three hemolysin genes detected by qRT-PCR. Additionally, only the icd mutant exhibited a significantly decreased growth yield in complex media. Growth yield was directly related to the abundance of glutamate. A strain with a restored wild type icd gene was created and shown to restore growth to a wild type cell density in complex media and pathogenicity in rainbow trout. Conclusions The data strongly suggest that a decreased growth yield, resulting from the inability to synthesize α-ketoglutarate, caused the attenuation despite normal levels of expression of virulence genes. Therefore, the ability of an extracellular pathogen to cause disease is dependent upon the availability of host-supplied nutrients for growth.     The NMDA receptor GluN2C subunit controls cortical excitatoryinhibitory balance, neuronal oscillations and cognitive function

    Despite strong evidence for NMDA receptor (NMDAR) hypofunction as an underlying factor for cognitive disorders, the precise roles of various NMDAR subtypes remains unknown. The GluN2Ccontaining NMDARs exhibit unique biophysical properties and expression pattern, and lower expression of GluN2C subunit has been reported in postmortem brains from schizophrenia patients. We found that loss of GluN2C subunit leads to a shift in cortical excitatory-inhibitory balance towards greater inhibition. Specifically, pyramidal neurons in the medial prefrontal cortex (mPFC) of GluN2C knockout mice have reduced mEPSC frequency and dendritic spine density and a contrasting higher frequency of mIPSCs. In addition a greater number of perisomatic GAD67 puncta was observed suggesting a potential increase in parvalbumin interneuron inputs. At a network level the GluN2C knockout mice were found to have a more robust increase in power of oscillations in response to NMDAR blocker MK- 801. Furthermore, GluN2C heterozygous and knockout mice exhibited abnormalities in cognition and sensorimotor gating. Our results demonstrate that loss of GluN2C subunit leads to cortical excitatoryinhibitory imbalance and abnormal neuronal oscillations associated with neurodevelopmental disorders

    The impact of post-operative sepsis on mortality after hospital discharge among elective surgical patients: a population-based cohort study

    Our aim in the present study was to assess the mortality impact of hospital-acquired post-operative sepsis up to 1 year after hospital discharge among adult non-short-stay elective surgical patients.We conducted a population-based, retrospective cohort study of all elective surgical patients admitted to 82 public acute hospitals between 1 January 2007 and 31 December 2012 in New South Wales, Australia. All adult elective surgical admission patients who stayed in hospital for ≥4 days and survived to discharge after post-operative sepsis were identified using the Admitted Patient Data Collection records linked with the Registry of Births, Deaths, and Marriages. We assessed post-discharge mortality rates at 30 days, 60 days, 90 days and 1 year and compared them with those of patients without post-operative sepsis.We studied 144,503 survivors to discharge. Of these, 1857 (1.3%) had experienced post-operative sepsis. Their post-discharge mortality rates at 30 days, 60 days, 90 days and 1 year were 4.6%, 6.7%, 8.1% and 13.5% (vs 0.7%, 1.2%, 1.5% and 3.8% in the non-sepsis cohort), respectively (P < 0.0001 for all). After adjustment for patient and hospital characteristics, post-operative sepsis remained independently associated with a higher mortality risk (30-day mortality HR 2.75, 95% CI 2.14-3.53; 60-day mortality HR 2.45, 95% CI 1.94-3.10; 90-day mortality HR 2.31, 95% CI 1.85-2.87; 1-year mortality HR 1.71, 95% CI 1.46-2.00). Being older than 75 years of age (HR 3.50, 95% CI 1.56-7.87) and presence of severe/very severe co-morbidities as defined by Charlson co-morbidity index (severe vs normal HR 2.05, 95% CI 1.45-2.89; very severe vs normal HR 2.17, 95% CI 1.49-3.17) were the only other significant independent predictors of increased 1-year mortality.Among elective surgical patients, post-operative sepsis is independently associated with increased post-discharge mortality up to 1 year after hospital discharge. This risk is particularly high in the first month, in older age patients and in the presence of severe/very severe co-morbidities.     Preconception Counseling in Women With Diabetes: A population-based study in the North of England

    To investigate the association of preconception counseling with markers of care and maternal characteristics in women with pregestational diabetes. The study includes data from a regional multi-center survey on 588 women with pregestational diabetes who delivered a singleton pregnancy between 2001 and 2004. Logistic regression was used to obtain crude and adjusted estimates of association. Preconception counseling was associated with better glycemic control 3 months preconception (odds ratio 1.91, 95% CI 1.10-3.04) and in the first trimester (2.05, 1.39-3.03), higher preconception folic acid intake (4.88, 3.26-7.30), and reduced risk of adverse pregnancy outcome (P = 0.027). Uptake of preconception counseling was positively associated with type 1 diabetes (1.87, 1.14-3.07) and White British ethnicity (2.56, 1.17-5.6) and negatively with deprivation score (0.78, 0.70-0.87). Efforts are needed to improve preconception counseling rates. Uptake is associated with maternal sociodemographic characteristics

Whilst there is a growing body of research considering how adults experience automobility, that is the increasingly central role of cars within societies, there has been little equivalent research exploring children's perspectives. Drawing upon a variety of methods including personal diaries, photographs, in‐depth interviews and surveys amongst schools within Buckinghamshire and North London, the paper contributes to filling this gap in existing research through exploring how cars are not only journey spaces for children, but are also sites for play, relaxation, homework, companionship, technology and the consumption of commodities. Using a Foucauldian analysis of power, insights into wider familial processes relating to mobility are provided by exploring how cars are sites of conflicting power relations between parents and children. The paper also explores how children's everyday experiences of cars were framed by wider sets of power relations, including car corporations which design and manufacture these spaces, and the role of capital which commodifies everyday activities in cars. In doing so, the paper challenges existing research on automobility for only focusing upon adults' experiences of cars and begins to theorise a more inclusive account of automobility which incorporates children and young people

    Progression of renal cell carcinoma is inhibited by genistein and radiation in an orthotopic model

    BACKGROUND: We have previously reported the potentiation of radiotherapy by the soy isoflavone genistein for prostate cancer using prostate tumor cells in vitro and orthotopic prostate tumor models in vivo. However, when genistein was used as single therapy in animal models, it promoted metastasis to regional para-aortic lymph nodes. To clarify whether these intriguing adverse effects of genistein are intrinsic to the orthotopic prostate tumor model, or these results could also be recapitulated in another model, we used the orthotopic metastatic KCI-18 renal cell carcinoma (RCC) model established in our laboratory. METHODS: The KCI-18 RCC cell line was generated from a patient with papillary renal cell carcinoma. Following orthotopic renal implantation of KCI-18 RCC cells and serial in vivo kidney passages in nude mice, we have established a reliable and predictable metastatic RCC tumor model. Mice bearing established kidney tumors were treated with genistein combined with kidney tumor irradiation. The effect of the therapy was assessed on the primary tumor and metastases to various organs. RESULTS: In this experimental model, the karyotype and histological characteristics of the human primary tumor are preserved. Tumor cells metastasize from the primary renal tumor to the lungs, liver and mesentery mimicking the progression of RCC in humans. Treatment of established kidney tumors with genistein demonstrated a tendency to stimulate the growth of the primary kidney tumor and increase the incidence of metastasis to the mesentery lining the bowel. In contrast, when given in conjunction with kidney tumor irradiation, genistein significantly inhibited the growth and progression of established kidney tumors. These findings confirm the potentiation of radiotherapy by genistein in the orthotopic RCC model as previously shown in orthotopic models of prostate cancer. CONCLUSION: Our studies in both RCC and prostate tumor models demonstrate that the combination of genistein with primary tumor irradiation is a more effective and safer therapeutic approach as the tumor growth and progression are inhibited both in the primary and metastatic sites
