    Infección experimental por herpesvirus bovino 1 en conejas gestantes

    Natural infection with Bovine herpesvirus 1 (BoHV-1) produces several clinical manifestations including conjunctivitis, respiratory signs, genital diseases and abortion. The rabbit is a good model for studying of latency, pathogenicity of BoHV-5 and other bovine herpesviruses. This study was conducted in order to analyze the response to experimental infection with BoHV-1 in rabbits during different periods of pregnancy. The results obtained could be useful for better understanding of the infection in cattle. A new method of infection was used. The rabbits developed clinical signs. The virus was recovered from nasal swabs and histological lesions were found in the analyzed samples. The humoral response was demonstrated and viral DNA was detected from the placentas. This work showed for the first time the arrival of BoHV-1 to blood after intranasal infection with the virus. It also provides a useful tool that can be used to evaluate the pathogenicity of by BoHV-1 strains, the immune response and to study viral latency and reactivation.La infección natural por Herpesvirus bovino 1 (BoHV-1) se manifiesta por conjuntivitis, signos respiratorios (rinotraqueitis), lesiones genitales (vulvovaginitis pustular infecciosa o balanopostitis) y abortos. El conejo, hasta el momento, ha resultado ser el mejor modelo experimental para estudiar los diferentes aspectos de la infección por BoHV-1, el fenómeno de latencia, la neuropatogenicidad de BoHV-5 y el comportamiento de diferentes cepas virales. Este trabajo se desarrolló con el objetivo de estudiar la respuesta de conejas infectadas con BoHV-1 en diferentes períodos de la gestación para que los datos resultantes puedan ser utilizados para la mejor comprensión de la infección en el bovino. Se utilizó un nuevo método de infección intranasal. Los animales desarrollaron signos clínicos. Se recuperó virus a partir de hisopados nasales, se observaron lesiones histopatológicas en las muestras analizadas, se demostró la respuesta inmune humoral y se detectó ADN viral a partir de placentas de los animales gestantes infectados. En este trabajo se evidenció por primera vez en el modelo conejo la llegada del virus al torrente sanguíneo luego de la infección intranasal. Además se aporta una herramienta de utilidad que puede ser utilizada para evaluar la virulencia de diferentes cepas de BoHV-1 y la respuesta inmune a distintos inmunógenos como también para realizar estudios de latencia y reactivación

    Complete genome amplification of Equine influenza virus subtype 2

    This work reports a method for rapid amplification of the complete genome of equine influenza virus subtype 2 (H3N8). A ThermoScriptTM reverse transcriptase instead of the avian myeloblastosis virus reverse transcriptase or Moloney murine leukemia virus reverse transcriptase was used. This enzyme has demonstrated higher thermal stability and is described as suitable to make long cDNA with a complex secondary structure. The product obtained by this method can be cloned, used in later sequencing reactions or nested-PCR with the purpose of achieving a rapid diagnosis and characterization of the equine influenza virus type A. This detection assay might be a valuable tool for diagnosis and screening of field samples as well as for conducting molecular studies.En este trabajo comunicamos un método rápido que permite la amplificación del genoma completo del subtipo 2 (H3N8) del virus de la influenza equina. Se utilizó la enzima transcriptasa reversa ThermoScriptTM en lugar de la transcriptasa reversa del virus de la mieloblastosis aviar o la transcriptasa reversa del virus de la leucemia murina de Moloney. Esta enzima ha demostrado tener una alta estabilidad térmica y la capacidad de hacer largas copias de ADN con una estructura secundaria compleja. El producto obtenido por esta técnica puede ser clonado y utilizado posteriormente en reacciones de secuenciación o de PCR anidada con la finalidad de lograr un diagnóstico rápido y la caracterización del virus de la influenza equina tipo A. Este ensayo de detección puede llegar a ser una valiosa herramienta para el diagnóstico y el análisis de muestras de campo, así como para la realización de estudios moleculares

    GNAO1 encephalopathy: broadening the phenotype and evaluating treatment and outcome

    OBJECTIVE: To describe better the motor phenotype, molecular genetic features, and clinical course of GNAO1-related disease. METHODS: We reviewed clinical information, video recordings, and neuroimaging of a newly identified cohort of 7 patients with de novo missense and splice site GNAO1 mutations, detected by next-generation sequencing techniques. RESULTS: Patients first presented in early childhood (median age of presentation 10 months, range 0-48 months), with a wide range of clinical symptoms ranging from severe motor and cognitive impairment with marked choreoathetosis, self-injurious behavior, and epileptic encephalopathy to a milder phenotype, featuring moderate developmental delay associated with complex stereotypies, mainly facial dyskinesia and mild epilepsy. Hyperkinetic movements were often exacerbated by specific triggers, such as voluntary movement, intercurrent illnesses, emotion, and high ambient temperature, leading to hospital admissions. Most patients were resistant to drug intervention, although tetrabenazine was effective in partially controlling dyskinesia for 2/7 patients. Emergency deep brain stimulation (DBS) was life saving in 1 patient, resulting in immediate clinical benefit with complete cessation of violent hyperkinetic movements. Five patients had well-controlled epilepsy and 1 had drug-resistant seizures. Structural brain abnormalities, including mild cerebral atrophy and corpus callosum dysgenesis, were evident in 5 patients. One patient had a diffuse astrocytoma (WHO grade II), surgically removed at age 16. CONCLUSIONS: Our findings support the causative role of GNAO1 mutations in an expanded spectrum of early-onset epilepsy and movement disorders, frequently exacerbated by specific triggers and at times associated with self-injurious behavior. Tetrabenazine and DBS were the most useful treatments for dyskinesia

    Analysis of the antibiotic resistance profiles in methicillin-sensitive s. Aureus pathotypes isolated on a commercial rabbit farm in Italy

    The breeding of meat rabbits is an important sector in the livestock industry in Italy. The focus of this study was to describe the antibiotic resistance profile distribution among the Methicillin-sensitive Staphylococcus aureus isolated in a rabbit farm. From 400 animals of different ages and three farm workers, 96 randomly selected strains isolated from various anatomical sites and lesions were analysed. According to spa typing and the resistance profiles towards veterinary and human antibiotics, 26 pathotypes were identified. The highest resistance was observed against Tetracyclines (92.3%) and Macrolides (80.8%), while almost all were susceptible to Penicillins, according to the limited use of β-lactams on the farm. In total, 92.3% of pathotypes were multidrug resistant (MDRs). Two MDR pathotypes belonging to the t2802 spa type were isolated from both farmers and rabbits. Age categories harboured significantly different pathotypes (p = 0.019), while no association was found between pathotypes and lesions (p = 0.128) or sampling sites (p = 0.491). The antibiotic resistance was observed to increase with the time spent in the farm environment (age category). The selective pressure exerted by antibiotic use acted by giving advantage to more resistant strains rather than by lowering susceptibility to various drug categories within strains

    La apoptosis es un tipo de muerte celular programada que puede ser desencadenada por múltiples factores, tanto internos como externos; dentro de estos últimos se encuentran las infecciones virales. Algunos alphaherpesvirus han desarrollado diversas estrategias para retardar o inhibir la muerte celular obteniendo, de esta manera, su propio beneficio al poder permanecer durante más tiempo en la célula. Hasta el momento no se ha identificado ningún mecanismo relacionado con la modulación de la muerte celular durante la infección con Herpesvirus equino tipo 1 (EHV-1). El objetivo del presente trabajo fue describir el efecto producido por la infección con EHV-1 sobre cultivos celulares inducidos a la muerte por apoptosis. La evaluación de la apoptosis se realizó mediante el reconocimiento de la fragmentación en escalera del ADN, la evaluación de la relación Anexina V/ioduro de propidio (IP) y la determinación del clivaje de la citoqueratina 18, utilizando técnicas de inmunofluorescencia. Los resultados indican una posible interferencia del EHV-1 con la muerte por apoptosis hacia la mitad de su ciclo de replicación, que se incrementa hacia el final del mismo.Apoptosis is a programmed cell death that can be triggered by many factors, both internal and external.     A Nested PCR technique for direct identification of Feline Immunodeficiency Virus (FIV) was developed. Two methods for proviral DNA extraction and two pairs of primers were evaluated. The method for amplification was standardized and its specificity was determined by the restriction endonucleases digestion of PCR products. Antibodies specific to FIV were determined using a commercial kit. Thirty heparinized blood samples obtained from cats with and without clinical signs consistent with FIV infection were analyzed. In 17 of the samples antibodies specific to FIV and proviral DNA were detected. The PCR technique proved sensitive and specific. It could be useful in the presence of doubtful serologic results and to detect provirus in cats with recent infections.Se desarrolló una técnica de doble reacción en cadena de la polimerasa ( Nested PCR ) para la detección directa del virus de la Inmunodeficiencia Felina (VIF). Se evaluaron dos métodos de extracción de DNA proviral y cuatro pares de cebadores. Se estandarizó el método de amplificación y la especificidad fue determinada por digestión de los productos de PCR con enzimas de restricción. La determinación de anticuerpos se realizó con un equipo comercial. Se trabajó con 30 muestras de sangre heparinizada provenientes de gatos con y sin signos clínicos compatibles con la infección con VIF. En 17 de las muestras examinadas se detectaron anticuerpos específicos contra VIF y DNA proviral. La técnica de PCR desarrollada resultó sensible y específica y podría ser utilizada cuando se presentaran resultados serológicos dudosos o para la detección del provirus en gatos con infecciones recientes

    Failure to thrive - An overlooked manifestation of KMT2B-related dystonia: A case presentation

    Background: KMT2B-related dystonia is a recently described form of childhood onset dystonia that may improve with deep brain stimulation. Prior reports have focused on neurologic features including prominent bulbar involvement without detailing general health consequences that may result from orolingual dysfunction. We describe a family with novel KMT2B mutation with several members with failure to thrive to highlight this non-neurologic, but consequential impact of mutation in this gene. Case presentation: We present a case of a 15-year old female who was admitted and evaluated for failure to thrive. On exam, she had severe speech dysfluency, limited ability to protrude the tongue, and generalized dystonia involving the oromandibular region, right upper and left lower extremity with left foot inversion contracture. The proband and her parents underwent whole genome sequencing. A previously undescribed variant, c.4960 T > C (p.Cys1654Arg), was identified in the KMT2B gene in the proband and mother, and this variant was subsequently confirmed in two maternal cousins, one with failure to thrive. Literature review identified frequent reports of prominent bulbar involvement but failure to thrive is rarely mentioned. Conclusion: Failure to thrive is a common pediatric clinical condition that has consequences for growth and development. In the presence of an abnormal neurologic exam, a search for a specific underlying genetic etiology should be pursued. With this case series, we highlight an unusual potentially treatable cause of failure to thrive, reinforce the importance of precise molecular diagnosis for patients with failure to thrive and an abnormal neurologic exam, and underscore the importance of cascade screening of family members

    Características biológicas de cepas de Herpesvirus bovino 1 y 5 utilizando el modelo experimental conejo

    Los Herpesvirus bovinos (BoHV) pueden infectar tanto a mustélidos como a conejos y esta última especie ha sido utilizada como modelo de laboratorio para la infección por BoHV-1 y 5. El objetivo de este trabajo fue estudiar la patogenicidad de diferentes cepas argentinas de BoHV-1 y BoHV-5 utilizando el modelo experimental conejo. Se utilizaron conejos de raza neozelandesa que se inocularon por vía intranasal e intravaginal. Los animales inoculados por vía intranasal con cepas de BoHV-5 desarrollaron signos nerviosos en el 83% de los casos, mientras que BoHV-1.1 causó signos nerviosos en el 57% de los animales y BoHV-1.2 no provocó signos clínicos evidentes. El BoHV-5 causó síntomas nerviosos solo en los animales jóvenes mientras que BoHV-1 solo lo hizo ocasionalmente y también en individuos jóvenes. Los conejos inoculados por vía intravaginal no mostraron signos clínicos ni lesiones aparentes en los órganos estudiados; la infección se demostró por seroconversión serológica. El conejo resultó adecuado para estudiar la sintomatología y las lesiones producidas en los distintos órganos, fundamentalmente en el sistema nervioso central. El modelo resultó de utilidad por ser económico, de muy fácil manejo y permitió reconocer diferencias en el comportamiento biológico de las cepas de BoHV-1 y BoHV-5 estudiadas.Bovine Herpesvirus (BoHV) can infect both rabbits and mustelids. Rabbit has been used as a laboratory model for infection with BoHV-1 and 5. The objective of this research was to study the pathogenicity of different Argentinian BoHV-1 and BoHV-5 strains by using the rabbit experimental model. New Zealand rabbits were inoculated by intranasal and intravaginal ways. The animals inoculated intranasally with strains of BoHV-5 developed neurological signs in 83% of the cases. BoHV-1.1 caused neurological signs in 57% of the animals and BoHV-1.2 did not cause clear clinical signs. BoHV-5 caused nervous signs in young animals while BoHV-1 did so occasionally in young rabbits. Animales inoculated intravaginally showed no apparent clinical signs or apparent lesions in the studied organs. The infection was demonstrated by serological seroconversion. The rabbit was appropriate to study the clinical signs and the lesions produced in the different organs, primarily in the central nervous system. The model was useful for being inexpensive and very easy to use, and it enabled to identify differences in the biological behavior of the studied BoHV-1 and BoHV-5 strains

    Adipose micro-grafts enhance tendinopathy healing in ovine model: An in vivo experimental perspective study

    In Europe, approximatively 100 000 to 500 000 tendon repairs are performed every year. These procedures are associated with a considerable rate of postoperative complications (from 6% to 11%). Autologous micro-grafts (AAMG) and stromal vascular fraction (SVF) have been shown to improve tendon healing in 60% to 70% of treated rodents. The purpose of this study was to evaluate the effects of AAMG in a sheep model with tendinopathy. We used sheep models because, as a large animal, they are more comparable to humans. The hypothesis was that SVF injection would improve tendon healing compared with the control group, reducing inflammatory and matrix degrading, while increasing anti-inflammatory expression and collagen synthesis in the early stage of tendon injury. Sixteen Apennine sheep aged 2 to 5 years underwent 500 UI type I collagenase injection into both common calcaneal tendons (CCT) to induce tendinopathy. After 15 days (T0), one CCT in every ovine underwent randomly to 2.5 mL of AAMG obtained by mechanical disruption and the contralateral CCTs received no treatment. Clinical, ecographic, and sonographic evaluations were performed after 4 weeks (T1) and 8 weeks (T2). Histological, immunohistochemical, real-time polymerase chain reaction (RT-PCR), and biomechanical evaluations were performed at T2. At T2, the treated group showed a final tendon diameter (9.1 ± 1.4 mm) and a hardness expression (62%) that were similar to the original healthy tendon (8.1 ± 1.1 mm; 100%), with a significant recovery compared with the control group (9.5 ± 1.7 mm; 39%). Moreover, histological analysis of the treated group revealed an improvement in the fiber orientation score, fiber edema score, infiltrative-inflammatory process, and necrosis score (4.3 ± 3.3) compared with control group (8.8 ± 2.9). Immunohistochemically, the treated group showed high expression of collagen 1, Factor VIII and significantly low expression of collagen 3. These data were confirmed by RT-PCR analysis. The study findings suggested that AAMGs obtained through mechanical disruption present a safe, efficient, and reliable technique, enhancing tendon healing
