    Inapproximability of the Standard Pebble Game and Hard to Pebble Graphs

    Pebble games are single-player games on DAGs involving placing and moving pebbles on nodes of the graph according to a certain set of rules. The goal is to pebble a set of target nodes using a minimum number of pebbles. In this paper, we present a possibly simpler proof of the result in [CLNV15] and strengthen the result to show that it is PSPACE-hard to determine the minimum number of pebbles to an additive n1/3ϵn^{1/3-\epsilon} term for all ϵ>0\epsilon > 0, which improves upon the currently known additive constant hardness of approximation [CLNV15] in the standard pebble game. We also introduce a family of explicit, constant indegree graphs with nn nodes where there exists a graph in the family such that using constant kk pebbles requires Ω(nk)\Omega(n^k) moves to pebble in both the standard and black-white pebble games. This independently answers an open question summarized in [Nor15] of whether a family of DAGs exists that meets the upper bound of O(nk)O(n^k) moves using constant kk pebbles with a different construction than that presented in [AdRNV17].Comment: Preliminary version in WADS 201

    In the last few years the Pichia pastoris expression system has been gaining more and more interest for the expression of recombinant proteins. Many groups have employed fermentation technology in their investigations because the system is fairly easy to scale up and suitable for the production in the milligram to gram range. A large number of heterologous proteins from different sources has been expressed, but the fermentation process technology has been investigated to a lesser extent. A large number of fermentations are carried out in standard bioreactors that may be insufficiently equipped to meet the demands of high-cell-density fermentations of methylotrophic yeasts. In particular, the lack of on-line methanol analysis leads to fermentation protocols that may impair the optimal expression of the desired products. We have used a commercially available methanol sensor to investigate in detail the effects of supplementary glycerol feeding while maintaining a constant methanol concentration during the induction of a Mut+ strain of Pichia pastoris. Specific glycerol feed rates in the range of 38-4.2 mg × g(exp -1) × h(exp -1) (mg glycerol per gram fresh weight per hour) were investigated. Expression of the recombinant scFv antibody fragment was only observed at specific feed rates below 6 mg × g(exp -1) × h(exp -1). At low specific feed rates, growth was even lower than with methanol as the sole carbon source and the harvest expression level of the scFv was only half of that found in the control fermentation. These results show that glycerol inhibits expression driven by the AOX1 promoter even at extremely limited availability and demonstrate the benefits of on-line methanol control in Pichia fermentation research

    Searching of gapped repeats and subrepetitions in a word

    A gapped repeat is a factor of the form uvuuvu where uu and vv are nonempty words. The period of the gapped repeat is defined as u+v|u|+|v|. The gapped repeat is maximal if it cannot be extended to the left or to the right by at least one letter with preserving its period. The gapped repeat is called α\alpha-gapped if its period is not greater than αv\alpha |v|. A δ\delta-subrepetition is a factor which exponent is less than 2 but is not less than 1+δ1+\delta (the exponent of the factor is the quotient of the length and the minimal period of the factor). The δ\delta-subrepetition is maximal if it cannot be extended to the left or to the right by at least one letter with preserving its minimal period. We reveal a close relation between maximal gapped repeats and maximal subrepetitions. Moreover, we show that in a word of length nn the number of maximal α\alpha-gapped repeats is bounded by O(α2n)O(\alpha^2n) and the number of maximal δ\delta-subrepetitions is bounded by O(n/δ2)O(n/\delta^2). Using the obtained upper bounds, we propose algorithms for finding all maximal α\alpha-gapped repeats and all maximal δ\delta-subrepetitions in a word of length nn. The algorithm for finding all maximal α\alpha-gapped repeats has O(α2n)O(\alpha^2n) time complexity for the case of constant alphabet size and O(nlogn+α2n)O(n\log n + \alpha^2n) time complexity for the general case. For finding all maximal δ\delta-subrepetitions we propose two algorithms. The first algorithm has O(nloglognδ2)O(\frac{n\log\log n}{\delta^2}) time complexity for the case of constant alphabet size and O(nlogn+nloglognδ2)O(n\log n +\frac{n\log\log n}{\delta^2}) time complexity for the general case.     Investigation of a Bubble Detector based on Active Electrolocation of Weakly Electric Fish

    Weakly electric fish employ active electrolocation for navigation and object detection. They emit an electric signal with their electric organ in the tail and sense the electric field with electroreceptors that are distributed over their skin. We adopted this principle to design a bubble detector that can detect gas bubbles in a fluid or, in principle, objects with different electric conductivity than the surrounding fluid. The evaluation of the influence of electrode diameter on detecting a given bubble size showed that the signal increases with electrode diameter. Therefore it appears that this detector will be more appropriate for large sized applications such as bubble columns than small sized applications such as bubble detectors in dialysis

    A Comparison of E-book and Print Book Discovery, Preferences and Usage by Science and Engineering Faculty and Graduate Students at the University of Kansas

    See below for the accompanying dataset and the survey instrument used to collect the data.The availability of science and technology e-books through the University of Kansas Libraries is growing rapidly through approval plans, e-book packages, and electronic demand-driven acquisitions. Based on informal conversations with faculty, questions still lingered as to the acceptance of books in the electronic format by faculty and graduate students in the STEM disciplines. To learn more about book format preferences, a survey was distributed via e-mail to 1,898 faculty and graduate students in science and technology at the University of Kansas. The survey included questions focused on print book use, e-book use, format preferences, and demographics. A majority of the 357 respondents indicated a preference for print books indicating many of the oft-repeated comments about the disadvantages of reading books on a computer. Patrons using tablets were more inclined to access e-books. The survey indicated a continuing need to purchase books in both print and electronic formats, and to market the availability of e-books to University of Kansas patrons

    Observing CMB polarisation through ice

    Ice crystal clouds in the upper troposphere can generate polarisation signals at the uK level. This signal can seriously affect very sensitive ground based searches for E- and B-mode of Cosmic Microwave Background polarisation. In this paper we estimate this effect within the ClOVER experiment observing bands (97, 150 and 220 GHz) for the selected observing site (Llano de Chajnantor, Atacama desert, Chile). The results show that the polarisation signal from the clouds can be of the order of or even bigger than the CMB expected polarisation. Climatological data suggest that this signal is fairly constant over the whole year in Antarctica. On the other hand the stronger seasonal variability in Atacama allows for a 50% of clean observations during the dry season.Comment: 7 Pages, 4 figure

    Optimal Color Range Reporting in One Dimension

    Color (or categorical) range reporting is a variant of the orthogonal range reporting problem in which every point in the input is assigned a \emph{color}. While the answer to an orthogonal point reporting query contains all points in the query range QQ, the answer to a color reporting query contains only distinct colors of points in QQ. In this paper we describe an O(N)-space data structure that answers one-dimensional color reporting queries in optimal O(k+1)O(k+1) time, where kk is the number of colors in the answer and NN is the number of points in the data structure. Our result can be also dynamized and extended to the external memory model

    A model sensitivity study of the impact of clouds on satellite detection and retrieval of volcanic ash

    Volcanic ash is commonly observed by infrared detectors on board Earth-orbiting satellites. In the presence of ice and/or liquid-water clouds, the detected volcanic ash signature may be altered. In this paper the sensitivity of detection and retrieval of volcanic ash to the presence of ice and liquid-water clouds was quantified by simulating synthetic equivalents to satellite infrared images with a 3-D radiative transfer model. The sensitivity study was made for the two recent eruptions of Eyjafjallajokull (2010) and Grimsvotn (2011) using realistic water and ice clouds and volcanic ash clouds. The water and ice clouds were taken from European Centre for Medium-RangeWeather Forecast (ECMWF) analysis data and the volcanic ash cloud fields from simulations by the Lagrangian particle dispersion model FLEXPART. The radiative transfer simulations were made both with and without ice and liquid-water clouds for the geometry and channels of the Spinning Enhanced Visible and Infrared Imager (SEVIRI). The synthetic SEVIRI images were used as input to standard reverse absorption ash detection and retrieval methods. Ice and liquid-water clouds were on average found to reduce the number of detected ash-affected pixels by 6-12 %. However, the effect was highly variable and for individual scenes up to 40% of pixels with mass loading > 0 : 2 gm 2 could not be detected due to the presence of water and ice clouds. For coincident pixels, i. e. pixels where ash was both present in the FLEXPART (hereafter referred to as "Flexpart") simulation and detected by the algorithm, the presence of clouds overall increased the retrieved mean mass loading for the Eyjafjallajokull (2010) eruption by about 13 %,while for the Grimsvotn (2011) eruption ash-mass loadings the effect was a 4% decrease of the retrieved ash-mass loading. However, larger differences were seen between scenes (standard deviations of +/- 30 and +/- 20% for Eyjafjallajokull and Grimsvotn, respectively) and even larger ones within scenes. The impact of ice and liquid-water clouds on the detection and retrieval of volcanic ash, implies that to fully appreciate the location and amount of ash, hyperspectral and spectral band measurements by satellite instruments should be combined with ash dispersion modelling
