    Genetic diversity of local rice varieties (Oryza sativa L.) in Vietnam’s Mekong Delta based on SSR markers and morphological characteristics

    Based on target traits, use of the genetic diversity of rice is beneficial for crop improvement. In this study, 41 rice varieties local to Vietnam’s Mekong Delta were evaluated on the basis of 11 quantitative morphological traits, along with the assessment of genetic diversity according to 50 SSR markers. The actual yield had a significance level of 0.05, while plant height and panicles per square meter had a high significance level of 0.001. Cluster analysis based on 11 quantitative traits also revealed that two were the optimal number of clusters used in this study. The highest polymorphic information content (PIC) value obtained was for RM286 (0.49), with a range of 0.00 to 0.49 and an average PIC of 0.14. Both structure and phylogenetic tree analyses as inferred from 50 SSR markers by the unweighted pair‐group method with arithmetic mean (UPGMA) also indicated that the 41 local rice varieties could be divided into two major groups. This study provides a useful information for Mot bui do cao CM, and Mot bui five varieties for improvements in the yield and intermediate amylose content of local rice‐breeding programs in future, especially for the Mekong Delta region

    Whole-genome sequencing of three local rice varieties (Oryza sativa L.) in Vietnam

    Recently, a new technology, Next-generation sequencing (NGS) has been launched and providing whole-genome sequences that helps identify molecular markers across the genome. DNA markers such as single nucleotides and insertion – deletion (InDel) polymorphisms were widely used for plant breeding particularly to distinguish important traits in rice. These PCR-based markers can be used for the precision detection of polymorphisms. Moreover, PCR-based approaches are simple and effective methods for dealing with the issue of fraudulent labeling and adulteration in the global rice industry. In this study, three local varieties of Oryza sativa L. in Vietnam were sequenced with up to ten times genome depth and at least four times coverage (~83%) using the Illumina HiSeq2000™ system, with an average of 6.5 GB clean data per sample, generated after filtering low-quality data. The data was approximately mapped up to 95% to the reference genome IRGSP 1.0. The results obtained from this study will contribute to a wide range of valuable information for further investigation into this germplasm

    Evaluation of the influence of the hydraulic fluid temperature on power loss of the mining hydraulic excavator

    In the steady state of operation, the temperature of a mining excavator hydraulic fluid is determined by the ambient temperature, hydraulic system design, and power losses. The amount of the hydraulic system power loss depends on the hydraulic fluid physical and thermodynamic properties and the degree of wear of the mining excavator hydraulic system working elements. The main causes of power losses are pressure losses in pipelines, valves and fittings, and leaks in pumps and hydraulic motors. With an increase in the temperature of hydraulic fluid, its viscosity decreases, which leads, on the one hand, to a decrease in power losses due to pressure losses in pipelines, valves and fittings, and, on the other hand, to an increase in volumetric leaks and associated power losses. To numerically determine the level of power losses occurring in the hydraulic system on an example of the Komatsu PC750-7 mining excavator when using Shell Tellus S2 V 22, 32, 46, 68 hydraulic oils with the corresponding kinematic viscosity of 22, 32, 46, 68 cSt at 40 °C, the developed calculation technique and software algorithm in the MatLab Simulink environment was used. The power loss coefficient, obtained by comparing power losses at the optimum temperature for a given hydraulic system in the conditions under consideration with the actual ones is proposed. The use of the coefficient will make it possible to reasonably select hydraulic fluids and set the values of the main pumps limit state and other hydraulic system elements, and evaluate the actual energy efficiency of the mining hydraulic excavator. Calculations have shown that the implementation of measures that ensure operation in the interval with a deviation of 10 % from the optimal temperature value for these conditions makes it possible to reduce energy losses from 3 to 12 %

    Igf2 Gene Expression Levels in Wild-Type and Mutant Mice

    Genomic imprinting occurs where only one allele of a gene is expressed depending on its parental origin. The imprinted Igf2 gene (Insulin-like growth factor 2) is encoding a growth factor, which play an important role in embryonic development and formation of the placenta. The regulation and expression of Igf2 is carried out by different promoters. Promoter expression is extremely complex in wild-type mice during development and is altered in several mutant mice bearing deletions at the Igf2/H19 locus. In this work, we analyzed the Igf2 RNA expression of the placenta-specific P0 promoter in placental tissue (embryonic day 17) of both wild-type and mutant mice. For all the other promoters, we used RNA extracted from liver tissues (postnatal day 7.5). All these RNAs were reverse transcribed to cDNA before quantifying expression levels of the promoters by quantitative PCR (qPCR).Our results show that transcriptions of Igf2 P2 and P3 promoters are the highest in all mice analyzed, except in ΔU2/Dom mutant mice where P0 and P1 promoters were highly expressed, while they display low expression in all the other mice strains analyzed.     Amynthas rodericensis is a common earthworm species in Vietnam. The feeding material and substrate affect the growth and reproduction of earthworms. The individual size, total number, weight, and gain weight of earthworms under laboratory conditions were investigated. The experiment was designed with four treatments (100% pig manure; 75% pig manure and 25% substrate; 50% pig manure and 50% substrate; 25% pig manure and 75% substrate) with six replicates in a completely randomized design; the experiments lasted ten weeks. The maximum growth and reproduction of A. rodericensis are observed with 75% pig manure and 25% substrate.Amynthas rodericensis là loài giun đất phổ biến ở Việt Nam. Nguồn thức ăn và cơ chất là những yếu tố ảnh hưởng đến sự sinh trưởng của giun đất nói chung và A. rodericensis nói riêng. Các yếu tố ảnh hưởng đến sự sinh trưởng bao gồm kích thước, khối lượng cơ thể, số lượng cá thể và tăng trọng cơ thể. Thí nghiệm có bốn nghiệm thức tương ứng với các tỷ lệ phối trộn phân lợn và chất nền khác nhau (NT1: 100% phân lợn; NT2: 75% phân lợn và 25% chất nền; NT3: 50% phân lợn và 50% chất nền; NT4: 25% phân lợn và 75% chất nền); mỗi nghiệm thức lặp lại sáu lần, bố trí theo phương pháp ngẫu nhiên hoàn toàn và được theo dõi đến 10 tuần. Kết quả cho thấy tỷ lệ phối trộn phân lợn với chất nền có ảnh hưởng khác nhau đến khả năng sinh trưởng của giun đất A. rodericensis.     Genetic variation of Nang Thom Cho Dao rice variety based on whole genome sequencing

    High-performance sequences are generating increasingly comprehensive catalogs of crop genetic variation. To make optimal use of this vast collection of data for research purposes, a robust and reproducible analytical pipeline discipline is required that is capable of accurately detecting and favoring variants. The entire genome sequencing data from the rice variety Nang Thom Cho Dao was analyzed using the appropriate bioinformatic pipeline. A total of 21 million reads with 6,6 GB of data were analyzed. SNPs and indels from the Nang Thom Cho Dao genome were found to be variable when compared to the Nipponbare reference rice genome. The result showed that the novel Indel of BADH2 gene in Nang Thom Cho Dao genome. The study will contribute valuable information to the development of genetic markers for rice breeding strategies using Nang Thom Cho Dao rice varieties

    Preliminary data on the aquatic invertebrate fauna of the Ma River, Thanh Hoa province: Research article

    A field survey for the invertebrate fauna conducted in the Ma River, Thanh Hoa province in 2013. The research applied multivariable analysis performed by the Primer v.6 software, such as CLUSTER, one-way ANOSIM, BEST and DIVERSE. The results showed a list of 138 aquatic invertebrate species. Of these, most were freshwater wide-distributing species coupled with others characterized for brackish and marine waters. The biodiversity status was quite high compared to several other rivers in the North of Vietnam. The list contained many economic-valued species and 2 of these were listed in the Red Data Book of Vietnam. The aquatic invertebrates showed a significant relation to the two different combinations of physiochemical factors for zooplanktons and zoobenthos, respectively. The values of the species number, abundance and Shannon-Weiner index for both of zooplanktons and zoobenthos showed a curved trend from the upper river segments to lower river segments. These figures for zooplanktons peaked in the middle river segments, whereas the numbers for zoobenthos achieved the highest numbers in the estuaries. The species composition of the estuaries differentiated significantly from that of other freshwater habitats.Năm 2013 đã tiến hành một đợt điều tra khu hệ động vật không xương sống sông Mã, tỉnh Thanh Hóa. Nghiên cứu sử các phân tích đa biến thông qua phần mền Primer v.6, bao gồm: CLUSTER, one-way ANOSIM, BEST và DIVERSE. Kết quả phân tích thu được 138 loài với thành phần loài chủ yếu là những loài nước ngọt thường gặp và phân bố rộng, ngoài ra còn có các loài đặc trưng cho nước lợ và mặn. Trong số các loài thu được, nhiều loài có giá trị kinh tế và 2 loài có tên trong Sách Đỏ Việt Nam. Khu hệ động vật không xương sống sông Mã có quan hệ chặt với hai nhóm chỉ số thủy lý hóa học khác nhau, tương ứng cho động vật nổi và động vật đáy. Giá trị các chỉ số sinh học gồm số lượng loài, mật độ và Shannon-Weiner hồi quy theo đường cong phi tuyến từ thượng lưu tới hạ lưu; đạt giá trị cao nhất tại cửa sông đối với động vật đáy và vùng trung lưu với động vật nổi.     An Outbreak of Severe Infections with Community-Acquired MRSA Carrying the Panton-Valentine Leukocidin Following Vaccination

    Background: Infections with community-acquired methicillin-resistant Staphylococcus aureus (CA-MRSA) are emerging worldwide. We investigated an outbreak of severe CA-MRSA infections in children following out-patient vaccination. Methods and Findings: We carried out a field investigation after adverse events following immunization (AEFI) were reported. We reviewed the clinical data from all cases. S. aureus recovered from skin infections and from nasal and throat swabs were analyzed by pulse-field gel electrophoresis, multi locus sequence typing, PCR and microarray. In May 2006, nine children presented with AEFI, ranging from fatal toxic shock syndrome, necrotizing soft tissue infection, purulent abscesses, to fever with rash. All had received a vaccination injection in different health centres in one District of Ho Chi Minh City. Eight children had been vaccinated by the same health care worker (HCW). Deficiencies in vaccine quality, storage practices, or preparation and delivery were not found. Infection control practices were insufficient. CA-MRSA was cultured in four children and from nasal and throat swabs from the HCW. Strains from children and HCW were indistinguishable. All carried the Panton-Valentine leukocidine (PVL), the staphylococcal enterotoxin B gene, the gene complex for staphylococcal-cassette-chromosome mec type V, and were sequence type 59. Strain HCM3A is epidemiologically unrelated to a strain of ST59 prevalent in the USA, althoughthey belong to the same lineage. Conclusions. We describe an outbreak of infections with CA-MRSA in children, transmitted by an asymptomatic colonized HCW during immunization injection. Consistent adherence to injection practice guidelines is needed to prevent CA-MRSA transmission in both in- and outpatient settings

    Awareness and preparedness of healthcare workers against the first wave of the COVID-19 pandemic: A cross-sectional survey across 57 countries.

    BACKGROUND: Since the COVID-19 pandemic began, there have been concerns related to the preparedness of healthcare workers (HCWs). This study aimed to describe the level of awareness and preparedness of hospital HCWs at the time of the first wave. METHODS: This multinational, multicenter, cross-sectional survey was conducted among hospital HCWs from February to May 2020. We used a hierarchical logistic regression multivariate analysis to adjust the influence of variables based on awareness and preparedness. We then used association rule mining to identify relationships between HCW confidence in handling suspected COVID-19 patients and prior COVID-19 case-management training. RESULTS: We surveyed 24,653 HCWs from 371 hospitals across 57 countries and received 17,302 responses from 70.2% HCWs overall. The median COVID-19 preparedness score was 11.0 (interquartile range [IQR] = 6.0-14.0) and the median awareness score was 29.6 (IQR = 26.6-32.6). HCWs at COVID-19 designated facilities with previous outbreak experience, or HCWs who were trained for dealing with the SARS-CoV-2 outbreak, had significantly higher levels of preparedness and awareness (p<0.001). Association rule mining suggests that nurses and doctors who had a 'great-extent-of-confidence' in handling suspected COVID-19 patients had participated in COVID-19 training courses. Male participants (mean difference = 0.34; 95% CI = 0.22, 0.46; p<0.001) and nurses (mean difference = 0.67; 95% CI = 0.53, 0.81; p<0.001) had higher preparedness scores compared to women participants and doctors. INTERPRETATION: There was an unsurprising high level of awareness and preparedness among HCWs who participated in COVID-19 training courses. However, disparity existed along the lines of gender and type of HCW. It is unknown whether the difference in COVID-19 preparedness that we detected early in the pandemic may have translated into disproportionate SARS-CoV-2 burden of disease by gender or HCW type
