    Dynamic Multipath Model of Low Angle Passive Radar Tracking with Experimental Evaluation

    The problem of multipath propagation encountered in radar tracking of low elevation targets in the presence of reflections from the sea is addressed. A detailed model of the multipath  propagation considering both the specular and the diffuse reflection components in target tracking using passive radars is established. Based on the geometry of the specular and the diffuse reflections, expressions for the reflection coefficient and the scattering field are derived. Experiments in the outfield indicate that the model proposed agrees with the test data well, which can provide an accurate prediction of the angle measurement errors in the presence of multipath effects.Defence Science Journal, 2012, 62(5), pp.331-337, DOI:http://dx.doi.org/10.14429/dsj.62.100

    Effect of land use patterns on stability and distributions of organic carbon in the hilly region of Western Sichuan, China

    Soil aggregation is important for the resistance of land surfaces to erosion, and it influences the ability of soils to remain productive. At the same time, it is also an important process of carbon sequestration. The objectives of this study were to elucidate the effects of different land use patterns on soil aggregate stability and the distribution of organic carbon in different aggregate fractions in order to prove that the different land use patterns enhance soil aggregate stability. Five kinds of soil samples were collected from the hilly area of western Sichuan under different land use patterns, such as abandoned farmlands, eucalyptus plantations, Chinese fir plantations, tea plantations and loquat orchards. The results demonstrated that the five land use patterns had high proportions of aggregates at the size of >2 mm after dry sieving, and had high proportions of aggregates at the size of < 0.5 mm after wet sieving. The aggregation abilities of the soils were significantly different depending on land use patterns. Waterstable aggregate stability was highest in the Chinese fir plantations, followed by eucalyptus plantations and tea plantations. Water-stable aggregate stability was the lowest in loquat  orchards and abandoned farmlands. Except for coarse particle, soil particle contents of the same size were affected according to the different land use patterns. As the size of aggregates decreased, the organic carbon content of the soil aggregates in tea plantations increased after a decrease, and then reduced again. However, the organic carbon contents of soil aggregates in other land uses increased continuously with the decreasing size of aggregates. Organic carbon content of the soil aggregates was strongly increased in land areas that had been converted from abandoned farmland to Chinese fir plantations, tea plantations and loquat orchards, while it was decreased when abandoned farmland was converted to eucalyptus plantations.     Evolutionary Enhancement of Zika Virus Infectivity in Aedes aegypti Mosquitoes

    Zika virus (ZIKV) remained obscure until the recent explosive outbreaks in French Polynesia (2013-2014) and South America (2015-2016). Phylogenetic studies have shown that ZIKV has evolved into African and Asian lineages. The Asian lineage of ZIKV was responsible for the recent epidemics in the Americas. However, the underlying mechanisms through which ZIKV rapidly and explosively spread from Asia to the Americas are unclear. Non-structural protein 1 (NS1) facilitates flavivirus acquisition by mosquitoes from an infected mammalian host and subsequently enhances viral prevalence in mosquitoes. Here we show that NS1 antigenaemia determines ZIKV infectivity in its mosquito vector Aedes aegypti, which acquires ZIKV via a blood meal. Clinical isolates from the most recent outbreak in the Americas were much more infectious in mosquitoes than the FSS13025 strain, which was isolated in Cambodia in 2010. Further analyses showed that these epidemic strains have higher NS1 antigenaemia than the FSS13025 strain because of an alanine-to-valine amino acid substitution at residue 188 in NS1. ZIKV infectivity was enhanced by this amino acid substitution in the ZIKV FSS13025 strain in mosquitoes that acquired ZIKV from a viraemic C57BL/6 mouse deficient in type I and II interferon (IFN) receptors (AG6 mouse). Our results reveal that ZIKV evolved to acquire a spontaneous mutation in its NS1 protein, resulting in increased NS1 antigenaemia. Enhancement of NS1 antigenaemia in infected hosts promotes ZIKV infectivity and prevalence in mosquitoes, which could have facilitated transmission during recent ZIKV epidemics

    Antibody dependent enhancement infection of Enterovirus 71 in vitro and in vivo

    BACKGROUND: Human enterovirus 71 (EV71) has emerged as a significant cause of acute encephalitis and deaths in young children. The clinical manifestations caused by EV71 varied from mild hand, foot and mouth disease to severe neurological complications and deaths, but its pathogenesis remains elusive. Antibody dependent enhancement (ADE) infection has been reported in various viruses and has been shown to contribute to disease severity. RESULTS: In this study, the presence of sub-neutralizing antibody was demonstrated to enhance EV71 infection in THP-1 cells and increase the mortality of EV71 infection in a suckling mouse model. Further, a secondary infection model was established to characterize the correlation between ADE and disease severity, and primary asymptomatic EV71 infection was shown to increase the mortality of the secondary EV71 infection in suckling mice. CONCLUSIONS: Together, these in vitro and in vivo experiments strongly supported the hypothesis of ADE infection of EV71. The present findings indicate ADE might contribute to the pathogenesis of severe EV71 infection, and raise practical issues of vaccine development and antibody-based therapy

    A duplex real-time RT-PCR assay for detecting H5N1 avian influenza virus and pandemic H1N1 influenza virus

    A duplex real-time reverse transcriptase polymerase chain reaction (RT-PCR) assay was improved for simultaneous detection of highly pathogenic H5N1 avian influenza virus and pandemic H1N1 (2009) influenza virus, which is suitable for early diagnosis of influenza-like patients and for epidemiological surveillance. The sensitivity of this duplex real-time RT-PCR assay was 0.02 TCID50 (50% tissue culture infective dose) for H5N1 and 0.2 TCID50 for the pandemic H1N1, which was the same as that of each single-target RT-PCR for pandemic H1N1 and even more sensitive for H5N1 with the same primers and probes. No cross reactivity of detecting other subtype influenza viruses or respiratory tract viruses was observed. Two hundred and thirty-six clinical specimens were tested by comparing with single real-time RT-PCR and result from the duplex assay was 100% consistent with the results of single real-time RT-PCR and sequence analysis

    In silico and microarray-based genomic approaches to identifying potential vaccine candidates against Leptospira interrogans

    BACKGROUND: Currently available vaccines against leptospirosis are of low efficacy, have an unacceptable side-effect profile, do not induce long-term protection, and provide no cross-protection against the different serovars of pathogenic leptospira. The current major focus in leptospirosis research is to discover conserved protective antigens that may elicit longer-term protection against a broad range of Leptospira. There is a need to screen vaccine candidate genes in the genome of Leptospira interrogans. RESULTS: Bioinformatics, comparative genomic hybridization (CGH) analysis and transcriptional analysis were used to identify vaccine candidates in the genome of L. interrogans serovar Lai strain #56601. Of a total of 4727 open reading frames (ORFs), 616 genes were predicted to encode surface-exposed proteins by P-CLASSIFIER combined with signal peptide prediction, α-helix transmembrane topology prediction, integral β-barrel outer membrane protein and lipoprotein prediction, as well as by retaining the genes shared by the two sequenced L. interrogans genomes and by subtracting genes with human homologues. A DNA microarray of L. interrogans strain #56601 was constructed for CGH analysis and transcriptome analysis in vitro. Three hundred and seven differential genes were identified in ten pathogenic serovars by CGH; 1427 genes had high transcriptional levels (Cy3 signal ≥ 342 and Cy5 signal ≥ 363.5, respectively). There were 565 genes in the intersection between the set encoding surface-exposed proteins and the set of 307 differential genes. The number of genes in the intersection between this set of 565 and the set of 1427 highly transcriptionally active genes was 226. These 226 genes were thus identified as putative vaccine candidates. The proteins encoded by these genes are not only potentially surface-exposed in the bacterium, but also conserved in two sequenced L. interrogans. Moreover, these genes are conserved among ten epidemic serovars in China and have high transcriptional levels in vitro. CONCLUSION: Of the 4727 ORFs in the genome of L. interrogans, 226 genes were identified as vaccine candidates by bioinformatics, CGH and transcriptional analysis on the basis of the theory of reverse vaccinology. The proteins encoded by these genes might be useful as vaccine candidates as well as for diagnosis of leptospirosis
