    Maleic anhydride-modified chicken ovalbumin as an effective and inexpensive anti-HIV microbicide candidate for prevention of HIV sexual transmission

    <p>Abstract</p> <p>Background</p> <p>Previous studies have shown that 3-hydroxyphthalic anhydride (HP)-modified bovine milk protein, β-lactoglobulin (β-LG), is a promising microbicide candidate. However, concerns regarding the potential risk of prion contamination in bovine products and carcinogenic potential of phthalate derivatives were raised. Here we sought to replace bovine protein with an animal protein of non-bovine origin and substitute HP with another anhydride for the development of anti-HIV microbicide for preventing HIV sexual transmission.</p> <p>Results</p> <p>Maleic anhydride (ML), succinic anhydride (SU) and HP at different conditions and variable pH values were used for modification of proteins. All the anhydrate-modified globulin-like proteins showed potent anti-HIV activity, which is correlated with the percentage of modified lysine and arginine residues in the modified protein. We selected maleic anhydride-modified ovalbumin (ML-OVA) for further study because OVA is easier to obtain than β-LG, and ML is safer than HP. Furthermore, ML-OVA exhibited broad antiviral activities against HIV-1, HIV-2, SHIV and SIV. This modified protein has no or low <it>in vitro </it>cytotoxicity to human T cells and vaginal epithelial cells. It is resistant to trypsin hydrolysis, possibly because the lysine and arginine residues in OVA are modified by ML. Mechanism studies suggest that ML-OVA inhibits HIV-1 entry by targeting gp120 on HIV-1 virions and also the CD4 receptor on the host cells.</p> <p>Conclusion</p> <p>ML-OVA is a potent HIV fusion/entry inhibitor with the potential to be developed as an effective, safe and inexpensive anti-HIV microbicide.</p

    Prevalence, associated factors and outcomes of pressure injuries in adult intensive care unit patients: the DecubICUs study

Epidemiologic data is however unavailable. We aimed to provide an international picture of the extent of pressure injuries and factors associated with ICU-acquired pressure injuries in adult ICU patients. Methods: International 1-day point-prevalence study; follow-up for outcome assessment until hospital discharge (maximum 12 weeks). Factors associated with ICU-acquired pressure injury and hospital mortality were assessed by generalised linear mixed-effects regression analysis. Results: Data from 13,254 patients in 1117 ICUs (90 countries) revealed 6747 pressure injuries; 3997 (59.2%) were ICU-acquired. Overall prevalence was 26.6% (95% confidence interval [CI] 25.9–27.3). ICU-acquired prevalence was 16.2% (95% CI 15.6–16.8). Sacrum (37%) and heels (19.5%) were most affected. Factors independently associated with ICU-acquired pressure injuries were older age, male sex, being underweight, emergency surgery, higher Simplified Acute Physiology Score II, Braden score 3 days, comorbidities (chronic obstructive pulmonary disease, immunodeficiency), organ support (renal replacement, mechanical ventilation on ICU admission), and being in a low or lower-middle income-economy. Gradually increasing associations with mortality were identified for increasing severity of pressure injury: stage I (odds ratio [OR] 1.5; 95% CI 1.2–1.8), stage II (OR 1.6; 95% CI 1.4–1.9), and stage III or worse (OR 2.8; 95% CI 2.3–3.3). Conclusion: Pressure injuries are common in adult ICU patients. ICU-acquired pressure injuries are associated with mainly intrinsic factors and mortality. Optimal care standards, increased awareness, appropriate resource allocation, and further research into optimal prevention are pivotal to tackle this important patient safety threat

    The increasing public attention on unceasing food safety incidents prompts the requirements of analytical techniques with high sensitivity, reliability, and reproducibility to timely prevent food safety incidents occurring. Food analysis is critically important for the health of both animals and human beings. Due to their unique physical and chemical properties, nanomaterials provide more opportunities for food quality and safety control. To date, nanomaterials have been widely used in the construction of sensors and biosensors to achieve more accurate, fast, and selective food safety detection. Here, various nanomaterial-based sensors for food analysis are outlined, including optical and electrochemical sensors. The discussion mainly involves the basic sensing principles, current strategies, and novel designs. Additionally, given the trend towards portable devices, various smartphone sensor-based point-of-care (POC) devices for home care testing are discussed

    Using Low Molecular Weight Organic Acids to Enhance Microbial Degradation of Polycyclic Aromatic Hydrocarbons: Current Understanding and Future Perspectives

    Polycyclic aromatic hydrocarbons (PAHs), an organic pollutant with persistence and carcinogenicity, are universally present in the environment and food processing. Biological approaches toward remediating PAHs-contaminated sites are a viable, economical, and environmentally friendly alternative compared to conventional physical and/or chemical remediation methods. Recently, various strategies relating to low molecular weight organic acids (LMWOAs) have been developed to enhance the microbial degradation of PAHs. However, the remaining challenge is to reveal the role of LMWOAs in the PAHs biodegradation process, and the latter limits researchers from expanding the application scope of biodegradation. In this mini-review, we summarized the current understanding of the impact of LMWOAs on (1) the physicochemical behavior of PAHs in the extracellular environment; (2) the interactions between PAHs and the microbial cell surface; and (3) the intracellular metabolization of PAHs. Future perspectives for this field are discussed in this review as well

    Spatial and Temporal Distribution Characteristics of Water Requirements for Maize in Inner Mongolia from 1959 to 2018

    Crop water requirements are crucial for agricultural water management and redistribution. Based on meteorological and agricultural observation data, the effective precipitation (Pe), water requirements (ETc), and irrigation water requirements (Ir) in the maize growing areas of Inner Mongolia were calculated. Furthermore, climatic trends of these variables were analysed to reveal their temporal and spatial distributions. The research results are as follows: the average Pe of maize in Inner Mongolia during the entire growth period was 125.9 mm, with an increasing trend from west to east. The Pe in the middle growth period of maize was the highest and was small in the early and late growth stages. The Pe climate exhibited a negative slope with a decreasing trend. The average ETc of maize during the entire growth period was 480.6 mm. The high-value areas are mainly distributed in the Wulatzhongqi and Linhe areas. The average Ir of maize during the entire growth period was 402.9 mm, and the spatial distribution is similar to that of ETc. In each growth period, Ir showed an increasing trend. Supplemental irrigation should be added appropriately during each growth period to ensure the normal growth of maize. This study can provide an effective basis for the optimisation of irrigation and regional water conservation in the maize cultivation area of Inner Mongolia

    Seedlings are vulnerable to many kinds of fatal abiotic and biotic agents, and examining the causes of seedling dynamics can help understand mechanisms of species coexistence. To disentangle the relative importance of neighborhood densities, habitat factors and phylogenetic relatedness on focal seedling survival, we monitored the survival of 5306 seedlings of 104 species &gt; 15 months. We address the following questions: (i) How do neighborhood densities, habitat variables and phylogenetic relatedness affect seedling survival? What is the relative importance of conspecific densities, habitat variables and phylogenetic relatedness to seedling survival? (ii) Does the importance of the neighborhood densities, habitat variables and phylogenetic relatedness vary among growth forms, leaf habits or dispersal modes?
