    Neutron scattering at high temperature and levitation techniques

    Studies of the liquid state present an obvious fundamental interest and are also important for technological applications since the molten state is an essential stage in various industrial processes (e.g. glass making, single crystal growing, iron and steel making). Most of the physical properties of a high-temperature liquid are related to its atomic structure. Thus it is important to develop devices to probe the local environment of the atoms in the sample. At very high temperature, it is difficult to use conventional furnaces, which present several problems. In particular, physical contact with the container can contaminate the sample and/or modify its structural properties. Such problems encouraged the development of containerless techniques, which are powerful tools to study high-temperature melts. By eliminating completely any contact between sample and container, it is possible to study the sample with a very high degree of control and to access very high temperatures. An additional advantage of levitation methods is that it is possible to supercool hot liquids down to several hundred of degrees below their equilibrium freezing point, since heterogeneous nucleation processes are suppressed

    Electronic spin precession in semiconductor quantum dots with spin-orbit coupling

    The electronic spin precession in semiconductor dots is strongly affected by the spin-orbit coupling. We present a theory of the electronic spin resonance at low magnetic fields that predicts a strong dependence on the dot occupation, the magnetic field and the spin-orbit coupling strength.     B4galnt2-mediated host glycosylation influences the susceptibility to Citrobacter rodentium infection

    Histo-blood group antigens in the intestinal mucosa play important roles in host-microbe interactions and modulate the susceptibility to enteric pathogens. The B4galnt2 gene, expressed in the GI tract of most mammals, including humans, encodes a beta-1,4-N-acetylgalactosaminyltransferase enzyme which catalyzes the last step in the biosynthesis of the Sd(a) and Cad blood group antigens by adding an N-acetylgalactosamine (GalNAc) residue to the precursor molecules. In our study, we found that loss of B4galnt2 expression is associated with increased susceptibility to Citrobacter rodentium infection, a murine model pathogen for human enteropathogenic Escherichia coli. We observed increased histopathological changes upon C. rodentium infection in mice lacking B4galnt2 compared to B4galnt2-expressing wild-type mice. In addition, wild-type mice cleared the C. rodentium infection faster than B4galnt2(-/-) knockout mice. It is known that C. rodentium uses its type 1 fimbriae adhesive subunit to bind specifically to D-mannose residues on mucosal cells. Flow cytometry analysis of intestinal epithelial cells showed the absence of GalNAc-modified glycans but an increase in mannosylated glycans in B4galnt2-deficient mice compared to B4galnt2-sufficient mice. Adhesion assays using intestinal epithelial organoid-derived monolayers revealed higher C. rodentium adherence to cells lacking B4galnt2 expression compared to wild-type cells which in turn was reduced in the absence of type I fimbriae. In summary, we show that B4galnt2 expression modulates the susceptibility to C. rodentium infection, which is partly mediated by fimbriae-mannose interaction

    Cloning and expression of an insect Ca2+-ATPase from Heliothis virescens

    AbstractA complementary DNA for the Tobacco Budworm, Heliothis virescens, sarco(endo)plasmic reticulum-type Ca2+-ATPase (HVSERCA) has been cloned and sequenced. cDNA fragments of adult rabbit fast-twitch muscle Ca2+-ATPase (SERCA1a) were used as heterologous probes to isolate a partial cDNA clone coding for a protein with high homology to the Ca2+-ATPase from Drosophila melanogaster (DRSERCA) and vertebrate ER/SR Ca2+ pumps. The entire cDNA clone contains an ORF encoding a protein of 1000 amino acids which shares the characteristic motifs of a P-type ATPase. HVSERCA shares 89% identity with DRSERCA, 80% identity with the Artemia Ca2+-ATPase and 72% identity with avian and mammalian SERCAs. An insect Ca2+-ATPase-specific polyclonal antiserum has been raised against a fusion protein containing sequence from the cytoplasmic domain of HVSERCA. Heterologous expression of the insect pump in COS-7 cells has been demonstrated by immunocytochemistry and the reticular pattern of staining is consistent with an ER localisation. However, the expressed enzyme from COS-7 cells does not appear to be active

    Female-female competition is influenced by forehead patch expression in pied flycatcher females

    There is increasing evidence that sexual selection operates in females and not only in males. However, the function of female signals in intrasexual competition has been little studied in species with conventional sex roles. In the Iberian populations of the pied flycatcher (Ficedula hypoleuca), some females express a white forehead patch, a trait that in other European populations, only males exhibit and has become a classical example in studies of sexual selection. Here, we investigated whether the expression of this trait plays a role in female-female competition during early breeding stages. To test this hypothesis, we simulated territorial intrusions by challenging resident females with stuffed female decoys expressing or not a forehead patch. We found that resident females directed more attacks per trial and maintained closer distances to non-patched decoys than to patched ones. Also, patched females were more likely to attack the decoy than non-patched females. Interestingly, females were more aggressive against the decoys when their mate was absent. This may indicate that females relax territory vigilance in the presence of their mate or that males interfere in the interaction between competing females. The behavior of resident males was also observed, although it was not affected by decoy's patch expression. Our findings suggest that the forehead patch plays a role in female intrasexual competition. If the forehead patch signals fighting ability, as it does in males, we may interpret that non-patched females probably avoided repeating costly agonistic encounters with the most dominant rivals.Fil: Morales, J..     Symmetry breaking and the random-phase approximation in small quantum dots

    The random-phase approximation has been used to compute the properties of parabolic two-dimensional quantum dots beyond the mean-field approximation. Special emphasis is put on the ground state correlation energy, the symmetry restoration and the role of the spurious modes within the random-phase approximation. A systematics with the Coulombic interaction strength is presented for the 2-electron dot, while for the 6- and 12-electron dots selected cases are discussed. To access the final edited and published work see DOI: 10.3390/molecules25245986Taxol®, which is also known as paclitaxel, is a chemotherapeutic agent widely used to treat different cancers. Since the discovery of its antitumoral activity, Taxol® has been used to treat over one million patients, making it one of the most widely employed antitumoral drugs. Taxol® was the first microtubule targeting agent described in the literature, with its main mechanism of action consisting of the disruption of microtubule dynamics, thus inducing mitotic arrest and cell death. However, secondary mechanisms for achieving apoptosis have also been demonstrated. Despite its wide use, Taxol® has certain disadvantages. The main challenges facing Taxol® are the need to find an environmentally sustainable production method based on the use of microorganisms, increase its bioavailability without exerting adverse effects on the health of patients and minimize the resistance presented by a high percentage of cells treated with paclitaxel. This review details, in a succinct manner, the main aspects of this important drug, from its discovery to the present day. We highlight the main challenges that must be faced in the coming years, in order to increase the effectiveness of Taxol® as an anticancer agent

    Identification of Candidate Regulators of Embryonic Stem Cell Differentiation by Comparative Phosphoprotein Affinity Profiling

    Embryonic stem cells are a unique cell population capable both of self-renewal and of differentiation into all tissues in the adult organism. Despite the central importance of these cells, little information is available regarding the intracellular signaling pathways that govern self-renewal or early steps in the differentiation program. Embryonic stem cell growth and differentiation correlates with kinase activities, but with the exception of the JAK/STAT3 pathway, the relevant substrates are unknown. To identify candidate phosphoproteins with potential relevance to embryonic stem cell differentiation, a systems biology approach was used. Proteins were purified using phosphoprotein affinity columns, then separated by two-dimensional gel electrophoresis, and detected by silver stain before being identified by tandem mass spectrometry. By comparing preparations from undifferentiated and differentiating mouse embryonic stem cells, a set of proteins was identified that exhibited altered post-translational modifications that correlated with differentiation state. Evidence for altered post-translational modification included altered gel mobility, altered recovery after affinity purification, and direct mass spectra evidence. Affymetrix microarray analysis indicated that gene expression levels of these same proteins had minimal variability over the same differentiation period. Bioinformatic annotations indicated that this set of proteins is enriched with chromatin remodeling, catabolic, and chaperone functions. This set of candidate phosphoprotein regulators of stem cell differentiation includes products of genes previously noted to be enriched in embryonic stem cells at the mRNA expression level as well as proteins not associated previously with stem cell differentiation status

    Quantum dots based on spin properties of semiconductor heterostructures

    The possibility of a novel type of semiconductor quantum dots obtained by spatially modulating the spin-orbit coupling intensity in III-V heterostructures is discussed. Using the effective mass model we predict confined one-electron states having peculiar spin properties. Furthermore, from mean field calculations (local-spin-density and Hartree-Fock) we find that even two electrons could form a bound state in these dots.Comment: 9 pages, 3 figures.     The Effect That Project Management Certification Has on Employability: Agent's Perceptions from Spain

    This study analyses the effects that the project management certification has on employability. This analysis started with a participative process in which various groups of experts who are involved in the certification of people were consulted. A personal interview was carried out amongst 106 professionals —certifying bodies, training institutions, the civil service, and international organisations— and amongst professional who are certified in project management by the International Project Management Association in Spain. The results show that the certification emerges as a powerful tool for improving employability. The effects are demonstrated across two complementary aspects: internal company aspects and external aspects relating to the labour market. Finally, by compiling the different agents’ opinions, a series of measures emerge for improving the accreditation processes as an employability tool and increasing the mutual learning between public and private actors
