    Interactions between Mytilus galloprovincialis hemocytes and the bivalve pathogens Vibrio aestuarianus 01/032 and Vibrio splendidus LGP32

    none9Marine bivalves can accumulate large numbers of bacteria, in particular Vibrio species, whose persistence in bivalve tissues largely depends on their sensitivity to the bactericidal activity of circulating hemocytes and hemolymph soluble factors. The interactions between vibrios and hemolymph have been investigated, in particular in bivalve species susceptible to infection by certain Vibrio spp. and strains. In this work, the effects of two bivalve pathogens, Vibrio splendidus LGP32 (V.s.) and Vibrio aestuarianus 01/032 (V.a.), isolated from oyster mortality outbreaks, on the hemocytes of Mytilus galloprovincialis were investigated. In vitro, V.s., but not V.a., induced a dramatic decrease in lysosomal membrane stability-LMS in the hemocytes; both vibrios induced a moderate lysozyme release, with V.s. > V.a.. The V.s.-induced decrease in LMS was mediated by activation of PI-3Kinase, as shown by use of different kinase inhibitors. TEM analysis showed rapid internalization of both vibrios; however, V.s. lead to cellular and lysosomal damage and was able to survive within the hemocytes, whereas significant killing of V.a. was observed. In vivo, in mussels challenged with either vibrio and sampled at 6, 24 and 96 h post-injection, transient decreases in hemocyte LMS and progressive increases in serum lysozyme activity were observed, with V.s. > V.a.. Moreover, whereas V.a. was efficiently cleared from hemolymph, V.s. showed significant growth, that was maximal at 24 h p.i. when lowest LMS values were recorded in the hemocytes. Both vibrios also induced significant decreases in LMS in the digestive gland, again with V.s. > V.a.. The results indicate distinct interactions between mussel hemocytes and the two vibrio strains tested. The effects of V.s. may be due to the capacity of this strain to interfere with the signaling pathways involved in hemocyte function, thus escaping the bactericidal activity of the host cell, as observed for certain mammalian pathogens.     Calculating economic values for milk components in a pasture-based-dairy-system: the case of Argentina

    Economic values (EV) of milk components (P: protein, F: fat and V: volume ) were obtained for a dairy system based on pasture, where a large proportion of milk is sold to industry. The EV’s were calculated using a profit function with a restriction in total feed supply. Economic value of protein (EVP) had the largest absolute and relative standardized values. Economic value of volume (EVV) was always small and negative. Multiple regression analyses of EV’s showed all EV’ s to be affected (P<.01) by the ratio of price of protein to price of fat (P:P:F) and level of production. The effects of P:P:F were linear and quadratic, whereas those of level of production were linear. Prediction equations obtained in the study can be used to calculate EV’s for different pricing systems and levels of milk production under grazing systems in Argentina

    Yield prediction of retail product from beef carcasses in Argentina.

    Currently the Argentine beef industry does not have an objective system of quality control at purchase, with which to guide production by means of differential prices. The objective of this work was to develop equations to predict the yield of retail cuts of the pistol hind quarter (Kg retail cuts/kg carcass, RC). For this purpose 343 pistol hind quarters (PHQ), of six biotypes were evaluated: BB, British (123); BC, Britsh x Continental (39); BI, British x Bos indicus (113); HH, Argentine Holstein (25); CC, Continental (20); and CI, Continental, Bos indicus (23). Multiple linear regressions were used to develop prediction equations for RC by biotype. Independent variables used were warm carcass side weight (CW); percentage of PHQ (kg PHQ/kg carcass, % PHQ); percentage of kidney knob and pelvic fat (% KPH); and dental classification (0, 2, 4 or 6 permanent incisors, DC). Equations to predict, RC based on these variables, for the different biotypes, were obtained. We conclude that a proposal to adopt these prediction equations is justified; with subsequent validations, this would enable the industry to apply objective criteria of yields in its purchases, thus guiding produces, through differential prices, to adjust the offer to market requirements

Immunization of adult female mice with V. cholerae outer-membrane vesicles (OMVs) passively protects suckling mice from challenge. Although V. cholerae is unable to colonize protected pups, the bacteria survive passage and have the potential to be transmitted to susceptible individuals.     Evaluation of Plant and Fungal Extracts for Their Potential Antigingivitis and Anticaries Activity

    The link between diet and health has lead to the promotion of functional foods which can enhance health. In this study, the oral health benefits of a number of food homogenates and high molecular mass and low molecular mass fractions were investigated. A comprehensive range of assays were performed to assess the action of these foods on the development of gingivitis and caries using bacterial species associated with these diseases. Both antigingivitis and anticaries effects were investigated by assays examining the prevention of biofilm formation and coaggregation, disruption of preexisting biofilms, and the foods' antibacterial effects. Assays investigating interactions with gingival epithelial cells and cytokine production were carried out to assess the foods' anti- gingivitis properties. Anti-caries properties such as interactions with hydroxyapatite, disruption of signal transduction, and the inhibition of acid production were investigated. The mushroom and chicory homogenates and low molecular mass fractions show promise as anti-caries and anti-gingivitis agents, and further testing and clinical trials will need to be performed to evaluate their true effectiveness in humans

    1-17Basic information about watershed relief, drawn from analytical-descriptive methodologies, allows for the physical parameters necessary to the study of natural resources, such as watershed integrated management, environmental impact, soil degradation, deforestation, water resource conservation, among others. All these processes linked to a strong spatial component allow the use of geographic information systems. Digital elevation models (DEMs) and their derivatives are an important component of these data sources. In this work we evaluated DEMs generated from indirect source data (digitized points from existing contour maps) to determine whether elevation accuracy varies with DEM spatial resolution (grid size). We also assessed the possible existence of some interaction between grid size and the software used that could influence the altimetric quality of the DEM. It was concluded that spatial resolution affects DEM quality. We also found that it is possible to find an ideal grid size in terms of the altimetric accuracy and computer weight size of the generated DEM. Regarding the existence of an interaction between grid size and the software used, which could affect the quality of models obtained based on the methodology of source data acquisition, we found that this possibility does exist when the database is generated from digitized points

    Fish as Reservoirs and Vectors of Vibrio cholerae

    Vibrio cholerae, the etiologic agent of cholera, is autochthonous to various aquatic environments, but despite intensive efforts its ecology remains an enigma. Recently, it was suggested that copepods and chironomids, both considered as natural reservoirs of V. cholerae, are dispersed by migratory waterbirds, thus possibly distributing the bacteria between water bodies within and between continents. Although fish have been implicated in the scientific literature with cholera cases, as far as we know, no study actually surveyed the presence of the bacteria in the fish. Here we show for the first time that fish of various species and habitats contain V. cholerae in their digestive tract. Fish (n = 110) were randomly sampled from freshwater and marine habitats in Israel. Ten different fish species sampled from freshwater habitats (lake, rivers and fish ponds), and one marine species, were found to carry V. cholerae. The fish intestine of Sarotherodon galilaeus harboured ca. 5×103 V. cholerae cfu per 1 gr intestine content—high rates compared with known V. cholerae cfu numbers in the bacteria's natural reservoirs. Our results, combined with evidence from the literature, suggest that fish are reservoirs of V. cholerae. As fish carrying the bacteria swim from one location to another (some fish species move from rivers to lakes or sea and vice versa), they serve as vectors on a small scale. Nevertheless, fish are consumed by waterbirds, which disseminate the bacteria on a global scale. Moreover, V. cholerae isolates had the ability to degrade chitin, indicating a commensal relationship between V. cholerae and fish. Better understanding of V. cholerae ecology can help reduce the times that human beings come into contact with this pathogen and thus minimize the health risk this poses
